Gene expression profiling of lung tissues comparing between Ism1-/- and Wildtype mice at postnatal day 7 (P7) stage

ABSTRACT: Isthmin 1 (ISM1) is a secreted protein of 60 kDa which has known functions of anti-apoptosis and anti-angiogenesis. To understand the physiological functions of ISM1, we generated Ism1 knockout (Ism1-/-) mice in C57BL/6J background through CRISPR gene editing technology. Under normal conditions, Ism1-/- mice presented spontaneous and progressive lung emphysema with enhanced immune cell infiltration compared to wildtype mice suggesting the role of ISM1 in pulmonary inflammation. In order to understand the pathways perturbed in the Ism1-/- mice, bulk RNA-sequencing analysis was performed using total RNA isolated from the lungs of WT and Ism1-/- mice at post-natal day 7 (P7) stage, when the process of alveolarization occurs.
