Analysis of sequences downstream of the polyadenylation signal and their influence on gene expression

ABSTRACT: Using massively parallel reporter assay (MPRA) we study influence of multiple variants of sequences located downstream of the polyadenylation signal (PAS) on the reporter gene transcription in human HEK293T cells. We have found that consensus sequence located downstream of the PAS and providing the strong increase in the reporter gene transcription is extremely T-rich. Since HEK293T cell culture is one of the most prominent host cell lines for recombinant antibody expression, we believe that our results may be useful for improving the efficiency of producing biotherapeutics including therapeutic antibody.
