Project description:Control (Villin-CreERT2tg/0; Apcflox/+; Nleflox/+), ApccKO (Villin-CreERT2tg/0; Apcflox/flox; Nleflox/+) and ApccKO; NlecKO (Villin-CreERT2tg/0; Apcflox/flox; Nleflox/null) mice were subjected to three daily intraperitoneal tamoxifen injections and their intestinal crypts were harvested at 24h (d1) or 48h (d2) post last tamoxifen injection. Total RNA was extracted according to the Trizol-chloroform extraction protocol provided by Invitrogen. RNAseq was performed on these extracted RNA to unravel the molecular mechanisms underlying the ApccKO;NlecKO phenotype.

Project description:These RNAseq data are from germinating cyp79B2, cyp79B3, myb28, myb29 (qko), cyp79B2, cyp79B3 (cyp) and myb28 myb29 (myb) mutant seeds. Col-0 genotype was used as the Arabidopsis' WT reference ecotype and all mutant seeds are from the Col-0 genetic background. Samples were sown on agarose 0,8%, supplemented or not by nitrate KNO3 10Mm or potassium thiocyanate KSCN 8µM. RNA isolation was performed using NucleoSpin® RNA Plant and Fungi kit. RNA quantification was obtained with a NanoDrop ND-100 (NanoDrop Technologies, DE, USA) and the RNA Integrity was measured with a 4100 Bioanalyzer (Agilent Technologies, Santa Clara, CA, USA). cDNA library construction and sequencing were performed with DNBseq™ technology at Beijing Genomics Institute BGI. FastQC was used to check read quality, mapping analysis and transcript quantiifcation were performed using a quasi-mapping alignment from Salmon.

Project description:We reported the transcriptomics responses from RNA-seq of AML12 transfected with Chrna4 over-expression plasmid. We used cultured AML12 from 3 to 5 generations and transfected it with pCMV6-AC-GFP or PAV-TBG-P2A-GFP-Chrna4 plasmid. After 24 h, total RNA of cultured AML12 was extracted with RNAiso Plus (TaKaRa). Then we delegated it to Novogene company for RNAseq aiming to identify key factors promoting this transition.

Project description:MC3T3 cells clone 1b were grown in alpha-MEM with 10% fetal calf serum (FCS, Gibco), 1% penicilin/streptomycin (Pen/Strep) and 1% L-Glutamine (L-Glu). For differentiation experiment, cells were seeded on 6 cm dishes (3x105cells/dish in 5 ml medium) and grownt to confluence for 3 days at 37C / 5% CO2. Cells were then stimulated with osteogenic stimulus (10 mM beta-glycerophosphate (GP, Sigma), 50 µM ascorbic acid (AA, Wako) and 1 mg/ml BMP-2 (Novartis)), or with 10mM GP alone for 1 or 3 days.
