Effect of inhibiting Glucose transporters by Glutor or Glupin on gene expression in primary human NK cells

ABSTRACT: To study the effect of Glucose transporter inhibition we isolated human NK cells from PBMCs of six healthy donors and expanded them with feeder cells and cytokines in the presence of 100nM Glutor, 100nM Glupin or 0.1% DMSO for 21 days. We then performed gene expression analysis using RNAseq.

Targeting T Cell Activation and Lupus Autoimmune Phenotypes by Inhibiting Glucose Transporters.

Project description:CD4+ T cells have numerous features of over-activated cellular metabolism in lupus patients and mouse models of the disease. This includes a higher glycolysis than in healthy controls. Glucose transporters play an essential role in glucose metabolism by controlling glucose import into the cell from the extracellular environment. We have previously shown that treatment of lupus-prone mice with 2-deoxy-D-glucose, which inhibits the first step of glycolysis was sufficient to prevent autoimmune activation. However, direct targeting of glucose transporters has never been tested in a mouse model of lupus. Here, we show that CG-5, a novel glucose transporter inhibitor, ameliorated autoimmune phenotypes in a spontaneous lupus-prone mouse model, B6.NZM2410.Sle1.Sle2.Sle3 (Triple-congenic, TC), and in a chronic graft- vs. host-disease (cGVHD) model of induced lupus. In vitro, CG-5 blocked glycolysis in CD4+ T cells, and limited the expansion of CD4+ T cells induced by alloreactive stimulation. CG-5 also modulated CD4+ T cell polarization by inhibiting Th1 and Th17 differentiation and promoting regulatory T (Treg) induction. Moreover, CG-5 treatment reduced lupus phenotypes including the expansion of germinal center B (GC B) cells, as well as the production of autoantibodies in both TC mice and cGVHD models. Finally, CG-5 blocked glycolysis in human T cells. Overall, our data suggest that blocking glucose uptake with a small molecule inhibitor ameliorates autoimmune activation, at least partially due to its inhibition of glycolysis in CD4+ T cells.

Project description:Glucose uptake in Escherichia coli had been studied extensively, but the lack of basic information of glucose uptake systems in C. acetobutylicum has limited the understanding of glucose assimilation in this strain. There is a main glucose transporter which is very efficient for glucose uptake and the maltose, mannose and galactose transport systems are also able to transport glucose into cytoplasm of E.coli. Therefore, it is very interesting to investigate whether a similar glucose transport mechanism exists in C. acetobutylicum.

New xylose transporters support the simultaneous consumption of glucose and xylose in Escherichia coli

Project description:RNA-seq libraries were constructed for six samples, including AE1.0, AE2.0, PS1.0, PS2.0, REV1.0, REV2.0 . For preparation of RNA samples, Co-utilization were assessed using AM1 mineral salt media supplemented with glucose and xylose at each concentration of 50 g·L-1. Seed culture were grown overnight (16 h) in AM1 medium supplemented with 2% sugar. For each experiment, E. coli was grown at 37 ℃ in a pH-controlled fermentation vessel with the initial cell density at OD550nm of 0.1.Total RNA for samples were isolated using TRIzol reagent (Invitrogen, Grand Island, USA), then used for high-throughput RNA sequencing. The 150-nt paired-end RNA-seq libraries were generated commercially at Novogene Biotechnology Co. Ltd (Tianjin, China) by using Illumina’s Hiseq X Ten platform (Illumina, San Diego, USA).
