Comprehensive analysis of aberrantly expressed circRNAs, mRNAs and lncRNAs in patients with nasopharyngeal carcinoma

ABSTRACT: The location of nasophryngeal cancer is hidden,so ti is difficult to diagnose at an early stage.In this study,we aimed to investigate expression profiles of circRNAs,mRNAs and IncRNAs and to provide some basis for further study.The expression profiles of circRNAs, mRNAs and lncRNAs were analyzed by microarray technology. The differentially expressed ncRNA was calculated by bioinformatics. Our study characterized the landscape of circRNAs, mRNAs and lncRNAs in NPC tissue and provided novel insights into the molecular mechanisms of NPC.

Project description:In this study, we investigated the role of microRNAs and circular RNAs in Nasopharyngeal Carcinoma (NPC) by constructing a circRNA-miRNA-mRNA co-expression network and performing differential expression analysis on mRNAs, miRNAs, and circRNAs. By comparing the expression profile of non-cancerous immortalized nasopharyngeal epithelial cell lines and NPC cell lines, we identified differentially expressed coding and non-coding RNAs. By leveraging coexpression and miRNA target prediction tools, we constructed a ceRNA network composed of mRNAs, miRNAs, and circRNAs.

Long non-coding RNAs and mRNAs in nasopharyngeal carcinoma metastasis

Project description:Increasing evidence has demonstrated a significant role for long non-coding RNAs (lncRNAs) in tumorigenesis. However, their functions in nasopharyngeal carcinoma (NPC) metastasis remain largely unknown. In this study, a model compared high and low metastatic NPC cell lines (5-8F vs. 6-10B and S18 vs. S26)was constructed to determine the expression profile of lncRNAs using the microarray analysis, and we found 167 lncRNAs and 209 mRNAs were differentially expressed. Validationof 26 significantly dysregulated lncRNAs by qRT-PCR showed the expression patterns of 22 lncRNAs were in accordance with the microarray data. Furthermore, the expression level of ENST00000470135, which was the most upregulated lncRNA in high metastatic cell lines, was significantly higher in NPC cell lines and tissues with lymph node metastasis（LNM）and knocking down ENST00000470135 suppressed the migration, invasion and proliferation of NPC cells in vitro. In conclusion, our study revealed expression patterns of lncRNAs in NPC metastasis. The dysregulated lncRNAs may act as novel biomarkers and therapeutic targets for NPC.

Long non-coding RNAs and mRNAs expression profilling in human nasopharyngeal carcinoma

Project description:Long non-coding RNAs (lncRNAs) play important roles in the tumorigenesis and progression of cancers. However, the clinical significance of lncRNAs and their regulatory mechanisms in nasopharyngeal carcinogenesis (NPC) are largely unknown. In this study, we used the microarray analysis to study the different expression profiles of lncRNAs and mRNAs in human nasopharyngeal carcinoma tissues. We performed genome-wide lncRNAs expression in 3 pairs of NPC and normal nasopharynx tissues and identified 384 dysregulated lncRNAs (fold change ≥2 and P <0.05).

Circular RNA expression in Nasopharyngeal carcinoma

Project description:Purpose:This study was performed to explore the circRNA profile of nasopharyngeal carcinoma (NPC). Methods: High-throughput sequencing was performed to identify the circRNA expression profile of NPC compared with healthy controls. Results: A total of 93 upregulated circRNAs and 77 downregulated circRNAs were identified. Conclusion: This study provides a leading and fundamental circRNA profile of NPC.

Screening and validation of lncRNAs and circRNAs as miRNA sponges [cell lines, miRNA-2_0]

Project description:Intensive research in past two decades has uncovered the presence and importance of noncoding RNAs (ncRNAs), which includes microRNAs (miRs) and long ncRNAs (lncRNAs). These two classes of ncRNAs interact to a certain extent, as some lncRNAs bind to miRs to sequester them. Such lncRNAs are collectively called 'competing endogenous RNAs' or 'miRNA sponges'. In this study, we screened for lncRNAs that may act as miRNA sponges using the publicly available data sets and databases. To uncover the roles of miRNA sponges, loss-of-function experiments were conducted, which revealed the biological roles as miRNA sponges. LINC00324 is important for the cell survival by binding to miR-615-5p leading to the de-repression of its target BTG2 LOC400043 controls several biological functions via sequestering miR-28-3p and miR-96-5p, thereby changing the expressions of transcriptional regulators. Finally, we also screened for circular RNAs (circRNAs) that may function as miRNA sponges. The results were negative at least for the selected circRNAs in this study. In conclusion, miRNA sponges can be identified by applying a series of bioinformatics techniques and validated with biological experiments. The expressions of miRs were examined in HEK-293, HUVEC and Hs68 cells.

Project description:To investigate the profiles of mRNAs, lncRNAs and circRNAs in browning of human adipose derived stem cells (hADSCs), we obtained hADSCs from omentum adipose tissues of 5 patients on who were performed abdominal operation and derived them into brown adipocytes. Then we employed microarray profiling the expression of lncRNAs, circRNAs and mRNA of hADSCs and their derived brown adipocytes.
