Project description:To elucidate the function of AP-1B in small intestinal epithelia, which is well known as one of the components of the AP-1 clathrin adaptor complex, gene expression profiling of AP-1B KO mice was performed. Small intestine of AP-1B +/- and -/- mice were dissected and soaked in Hank's Buffered Salt Solution (HBSS) (GIBCO) containing 30 mM EDTA. After incubation on ice for 20 min, IECs were isolated by manipulation with a fine needle under stereomicroscopic monitoring (n=2).

AP-1B regulates interaction between intraepithelial lymphocyte-epithelial cell in the intestine

Project description:Intraepithelial lymphocytes (IELs) reside in the epithelial layer to protect against foreign pathogens and maintain epithelial barrier functions in the intestine. The interaction between IELs and epithelial cells is required for their functions; however, the underlying molecular machinery still remains to be elucidated. Here, we found that intestinal epithelium-specific deficiency in a clathrin adaptor protein (AP)-1B, which regulates the basolateral protein sorting, led to the massive reduction of IELs. Quantitative proteomics demonstrated that dozens of proteins, including known IEL-interacting proteins—E-cadherin, Btnl2, and Plexin B2—decreased in the basolateral membrane of AP-1B-deficient epithelial cells. Among the downregulated proteins, CD166 is included. CD166 interacted with CD6 on the surface of induced IELs. CD166 knockdown using shRNA in intestinal organoid culture significantly inhibited the recruitment of IELs to an epithelial layer. These data suggest that basolateral sorting by AP-1B is indispensable for maintaining IELs in the epithelial layer and survival of IELs.

The kinesin KIF16B mediates apical transcytosis of transferrin receptor in AP-1B-deficient epithelia.

Project description:Polarized epithelial cells take up nutrients from the blood through receptors that are endocytosed and recycle back to the basolateral plasma membrane (PM) utilizing the epithelial-specific clathrin adaptor AP-1B. Some native epithelia lack AP-1B and therefore recycle cognate basolateral receptors to the apical PM, where they carry out important functions for the host organ. Here, we report a novel transcytotic pathway employed by AP-1B-deficient epithelia to relocate AP-1B cargo, such as transferrin receptor (TfR), to the apical PM. Lack of AP-1B inhibited basolateral recycling of TfR from common recycling endosomes (CRE), the site of function of AP-1B, and promoted its transfer to apical recycling endosomes (ARE) mediated by the plus-end kinesin KIF16B and non-centrosomal microtubules, and its delivery to the apical membrane mediated by the small GTPase rab11a. Hence, our experiments suggest that the apical recycling pathway of epithelial cells is functionally equivalent to the rab11a-dependent TfR recycling pathway of non-polarized cells. They define a transcytotic pathway important for the physiology of native AP-1B-deficient epithelia and report the first microtubule motor involved in transcytosis.

inflammatory response to IL-1b in WT, LDHA KO, NAMPT KO, NFKBIZ KO and FX11 treated chondrocytes

Project description:Our goal is to study the impacts of IL-1b treatment on chondrocyte physiology. Furthermore, we knocked out several genes that we believe are important for modulating the inflammatory response. We find that IL-1b treatment induces metabolic reprogramming towards aerobic glycoylsis. We found that inhibition of LDHA using inbhitiro FX11 inhibited the inflammatory response of chondrocytes. We hypothesized that this effect was mediated by interaction with NADH, leading to regulation of IkB-z protein. Thus, deletion of LDHA, NAMPT or NFKBIZ was performed and found to inhibit inflammatory response genes induced by IL-1b, with similar gene expression changes amongst the three suggesting that they may work in the same pathway.

Project description:This phase I trial is studying the best dose of 3-AP and the side effects of giving 3-AP together with gemcitabine in treating patients with advanced solid tumors or lymphoma. Drugs used in chemotherapy, such as 3-AP and gemcitabine (GEM), work in different ways to stop the growth of cancer cells, either by killing the cells or by stopping them from dividing. 3-AP may help gemcitabine kill more cancer cells by making the cells more sensitive to the drug. 3-AP may also stop the growth of tumor cells by blocking some of the enzymes needed for cell growth.
