Genome annotation of the blackstripe livebearer Poeciliopsis prolifica, a placental Poeciliid for understanding fish placenta evolution

ABSTRACT: The objective of our study is to obtain a high-quality reference genome of Poeciliopsis prolifica with gene annotation and identify the exonic SNP in inbred lines by RNA sequencing to provide the essential genetic toolkit for evolutionary and comparative genomic studies for P. prolifica and facilitate the reseach of placental evolution.

Transcription profiling of human placenta isolated from first time mothers with placental malaria

Project description:Chronic inflammation during placental malaria (PM) caused by Plasmodium falciparum is most frequent in first-time mothers and is associated with poor maternal and fetal outcomes. In the first genome wide analysis of the local human response to sequestered malaria parasites, we identified genes associated with chronic PM, then localized the corresponding proteins and immune cell subsets in placental cryosections. Experiment Overall Design: Placental samples from twenty first-time mothers were selected based on placental malaria (PM) status and RNA quality. Ten had active PM-episodes, seven of which had inflammatory cells on histology. Of the ten PM-negative women, five had histological evidence of a past PM-episode, including one with inflammatory cells.

Project description:The omega-3 long chain poly-unsaturated fatty acid (n-3 LCPUFA) docosahexaenoic acid (DHA) plays a central role in fetal and neonatal development in humans, in particular the development of the brain and nervous system, and this has led to numerous studies focussed on determining the effect of exposure to an increased supply of DHA on pregnancy and neonatal outcomes. The aim of this study was to determine the impact of treatment with a DHA-enriched fish-oil emulsion on fatty acid composition, proliferation rate and gene expression in human placental HTR8/SVneo cells in vitro.

A mechanism of gene evolution generating mucin function

Project description:How novel gene functions evolve is a fundamental question in biology. Mucin proteins, a functionally but not evolutionarily defined group of proteins, allow to study convergent evolution of gene function. By analyzing the genomic variation of mucins across a wide range of mammalian genomes, we propose that exonic repeats and their copy number variation contribute substantially to the de novo evolution of new gene functions. By integrating bioinformatic, phylogenetic, proteomic, and immunohistochemical approaches, we identified 15 undescribed instances of evolutionary convergence, where novel mucins originated by gaining densely O-glycosylated exonic repeat domains. Our results suggest that secreted proteins rich in proline are natural precursors for acquiring mucin function. Our findings have broad implications for understanding the role of exonic repeats in the parallel evolution of new gene functions, especially those involving protein glycosylation.

Treating the placenta with nanoparticles to prevent altered placental secretion and fetal development in response to hypoxia [Fetal Plasma]

Project description:Hypoxia-related pregnancy complications increase the risk of disease in the child in later life. No prevention is available. Previously we noted that a trophoblast barrier, an in vitro model of the placenta, reacted to oxidative stress by secreting factors that damage neighbouring cells. Application of mitochondrion-targeted antioxidant MitoQ prevented this. Here we tested the effects of MitoQ-bound nanoparticles on trophoblast barriers and in a rat model of gestational hypoxia.A single dose of MitoQ-nanoparticles, administered maternally before a hypoxic episode, reduced oxidative stress in the placental barrier without reaching the fetus and prevented changes to birthweight. MitoQ-nanoparticles further suppressed damaging signalling from the placental barriers. Altered signalling molecules in the fetal plasma and in conditioned media from rat placenta included changes to proteins with relevance to cardiovascular disease. We suggest as a future possibility, treatment of the placenta to prevent disease in the offspring in later life.
