Project description:High mobility group A1 (HMGA1) chromatin regulators are upregulated in diverse tumors where they portend adverse outcomes, although how they function in cancer remains unclear. Pancreatic ductal adenocarcinomas (PDACs) are highly lethal tumors characterized by dense desmoplastic stroma composed predominantly of cancer-associated fibroblasts and fibrotic tissue. Here, we uncover an epigenetic program whereby HMGA1 upregulates FGF19 during tumor progression and stroma formation. HMGA1 deficiency disrupts oncogenic properties in vitro while impairing tumor inception and progression in KPC mice and subcutaneous or orthotopic models of PDAC. RNA sequencing revealed HMGA1 transcriptional networks governing proliferation and tumor-stroma interactions, including the FGF19 gene. HMGA1 directly induces FGF19 expression and increases its protein secretion by recruiting active histone marks (H3K4me3, H3K27Ac). Surprisingly, disrupting FGF19 via gene silencing or the FGFR4 inhibitor BLU9931 recapitulates most phenotypes observed with HMGA1 deficiency, decreasing tumor growth and formation of a desmoplastic stroma in mouse models of PDAC. In human PDAC, overexpression of HMGA1 and FGF19 defines a subset of tumors with extremely poor outcomes. Our results reveal what we believe is a new paradigm whereby HMGA1 and FGF19 drive tumor progression and stroma formation, thus illuminating FGF19 as a rational therapeutic target for a molecularly defined PDAC subtype.

Project description:Emerging evidence suggests that tumor cells metastasize by co-opting stem cell transcriptional networks, although the molecular underpinnings of this process are poorly understood. Here, we show for the first time that the high mobility group A1 (HMGA1) gene drives metastatic progression in triple negative breast cancer cells (MDA-MB-231) by reprogramming cancer cells to a stem-like state. We discovered an HMGA1 signature in triple negative breast cancer cells that is highly enriched in embryonic stem cells. Together, these findings indicate that HMGA1 is a master regulator of tumor progression in breast cancer by reprogramming cancer cells through stem cell transcriptional networks. Future studies are needed to determine how to target HMGA1 in therapy. HMGA1 was knocked-down in MDA-MB-231 cells using siRNA as we previously described (Tesfaye A 2007). RNA from three independent knockdown experiements along with 3 control populations were collected by Rneasy miniprep (Qiagen) and analyzed by Affymetrix Human Exon 1.0 ST platform.

Project description:Analysis of MDA-MB-231 breast cancer cells depleted for High Mobility Group A1 (HMGA1) using siRNA. HMGA1 is involved in invasion and metastasis in breast cancer cells. Results identify the specific transcriptional program induced by HMGA1 in highly metastatic breast cancer cells. MDA-MB-231 cells were transfected with HMGA1-specific siRNA or a control siRNA. Transfections were performed by using Lipofectamin RNAiMAX (Invitrogen) according to the manufacturer's procedure. Seventy-two hours after transfection, samples were processed for total RNA extraction and hybridization on Affymetrix microarrays. Four biological replicas (A, B, C, D) were used for each of the two conditions, for a total of 8 samples.

Project description:Metabolic reprogramming is a typical feature of tumors, in which biological macromolecules and energy produced in abnormal metabolism meet the requirements of highly proliferative tumor cells and participate in multiple stages of tumor development. High mobility group A1 (HMGA1) is a structural transcription factor, which plays a carcinogenic role in regulating the transcription of oncogenes. Bioinformatics analysis showed that HMGA1 was overexpressed in human CRC. However, the mechanisms by which HMGA1 promotes the tumorigenesis of CRC remains unknown. Herein, we applied intestinal epithelium conditional knockout (Hmga1△IEC) and knock-in (Hmga1IEC-OE/+) mice of HMGA1 to induce CRC, and identified that HMGA1 promotes colorectal cancer growth by increasing lipid synthesis. Single-cell sequencing (scRNA seq) and immunohistochemical staining showed that HMGA1 was highly expressed in the epithelial cells of CRC. HMGA1 promoted the CRC cell proliferation and accelerated CRC development in HMGA1-knock-in (Hmga1IEC-OE/+) mice. Knockout of HMGA1 (Hmga1△IEC) in intestinal epithelial cells of mice reduced the lipid accumulation and inhibited the occurrence and development of CRC. We further characterized that HMGA1 upregulated the level of fatty acid synthase (FASN) through enhancing the transcriptional binding of sterol regulatory element-binding protein 1 (SREBP1) to the promoter of FASN, leading to an increase in lipid synthesis in intestinal epithelial cells. High fat diet (HFD) aggravated the malignant progression of CRC in Hmga1△IEC mice and reversed the inhibitory effect of HMGA1 depletion on CRC. Administration Orlistat (50 mg/kg), an inhibitor of FASN-mediated lipid synthesis, into Hmga1-knock-in (Hmga1IEC-OE/+) mice markedly reduced lipid accumulation in intestinal epithelial cells and decreased the tumorigenesis of CRC. Taken together, our data suggest that HMGA1 promotes the tumorigenesis of colorectal cancer by up-regulating FASN-mediated de novo fatty acid synthesis.
