CaP-coated Zn-Mn-Li alloys regulate osseointegration via influencing macrophage polarization in the osteogenic environment

ABSTRACT: To investigate the mechanism by which Zn alloy scaffold regulates macrophage polarization towards M2, we established RAW264.7 cell lines We then performed gene expression profiling using data obtained from RNA-seq in macrophages stimulated by three different sets of materials.

Transcriptome analysis of high-risk cattle supplemented with inorganic or organic Co, Cu, Mn, and Zn

Project description:This objective of this study was to identify gene expression and biological mechanisms related to trace mineral metabolism, central to inflammatory mediation and disease development, through the evaluation of the whole blood transcriptome in post‐weaned cattle receiving different mineral treatments. Here, 56 high‐risk cross-bred beef cattle (~250 kg) were assigned to receive ad libitum access to supplements containing 1 of 3 treatments over a 60-day period: 1) sulfate sources of Cu, Co, Mn, and Zn in feed (INR; custom blend; n= 40); 2) organic complexed source of Cu, Mn, Co, and Zn in feed (AAC; Availa4; n = 40); or 3) organic complexed source of Cu, Mn, Co, and Zn in feed and administration of a drench product [ProFusion] upon arrival (COMBO; n = 40). Whole blood RNA from 26 cattle (~4 calves/pen) were matched and utilized to assess the host transcriptome at days 0, 13, 28, 45, and 60 for the assessment of host transcriptome responses over time.

Combining P and Zn fertilization to enhance yield, grain Zn availability and protein profile in maize grown on Mediterranean soils

Project description:Alkaline soils such as those found in some Mediterranean areas typically have a low phosphorus (P) and zinc (Zn) phytoavailability that detracts from plant growth and yield. We examined the effects of P and Zn fertilization individually and in combination on growth, yield and grain protein content in maize grown in pots filled with three Mediterranean soils. P and Zn translocation was impaired, and yield reduced by 8–85%, in plants treated with Zn or P alone. In contrast, joint fertilization with P and Zn enhanced translocation to grain and nutrient use efficiency, thereby increasing plant growth, yield (31–121%) and grain Zn availability. Fertilization with P or Zn also influenced the abundance of specific proteins affecting grain quality (viz., storage, lys-rich and cell wall proteins), which were more abundant in mature grains from plants fertilized with Zn alone and, to a lesser extent, P + Zn.

Transcriptomic study of rice crowns from genotypes differing in tolerance to Zn deficiency

Project description:The transcriptome of crown tissue from several Zn-efficient and Zn-inefficient rice genotypes, exposed or not to Zn deficiency, was investigated using RNA sequencing. By grouping data from different genotypes within each efficiency group, we were able to 1) identify several well-known and novel Zn transporters involved in Zn retranslocation from the crown to the shoot and roots in response to Zn deficiency; 2) determine that Zn deficiency triggers the conversion of soluble sugars into starch; and 3) detect several candidate genes possibly conferring Zn efficiency.

Project description:Zn-limited cells accumulate Cu in intracellular structures in a reversable manner. We used RNAseq analysis to monitor gene expression in Zn-limited cells, and during a time course (0 - 24 h) after Zn resupply. Comparison of Chlamydomonas reinhardtii gene expression when Zn deficient cells are resupplied with 2.5 M-BM-5M Zn. Time points 0 (before resupply), 1.5, 3, 4.5, 12, and 24 hours after resupply. Four biological replicates per condition.
