Project description:During an incompatible or compatible interaction between rice (Oryza sativa) and the Asian rice gall midge (Orseolia oryzae), a lot of genetic reprogamming occurs in the plant host We used microarray to know the changes occuring in the resistant host (indica rice variety RP2068-18-3-5) when challenged by avirulent biotype of gall midge (GMB 1). During this incompatible interaction the resistance in the host is manifested by a hypersenstive response. Using microarray data, we identified distinct classes of up- and down-regulated genes during this process. Tissues from the stem region (feeding site of insect larvae) of the plants that were exposed to gall midge, were taken for RNA extraction and hybridization on Affymetric microarrays. For control, tissues were taken from the corresponding region of plants that were not exposed to gall midge.

Project description:Yield loss in crop plants due to biotic stresses is a major problem and pyramiding of R genes is often suggested for sustained and durable resistance against target pests. Information is available for single R gene interaction in rice and other crop plants, but not much data is available for multiple R gene interaction against multiple pathogens/pests. In this study we carried out transcriptional analysis of two rice lines introgressed with either R gene against bacterial blight and gall midge or, against bacterial blight and fungal blast. The gene expression changes were investigated through microarray and the expression pattern upon co-infection with multiple pathogens was analyzed to study the synergistic effect of R gene mediated defense responses as well as to explore the possibility of any antagonism between the R genes as defense pathway employed due to R gene mediated resistance variesaccording to the attacking pathogen/pest. Simultaneous RNA-Seq Analysis of a Mixed Transcriptome of Rice and Blast Fungus Interaction

Project description:To understand the plant-pathogen interaction comprehensively, it is valuable to monitor the gene expression profiles of both interacting organisms simultaneously in the same infected plant tissue. Using RNA-Seq, we analyzed the mixed transcriptome of rice and blast fungus in infected leaves at 24 hours post-inoculation. We demonstrated that our method detected the gene expression of both the host plant and pathogen simultaneously in the same infected leaf blades in natural infection conditions without any artificial treatments. Using compatible (Ina86-137) and incompatible (P91-15B) fungal strains as pathogens, we revealed the differential expression profiles of the compatible and incompatible interactions and observed that the responsive gene expression was more drastic in the incompatible interaction. Our mixed transcriptome analysis is useful for the simultaneous elucidation of the tactics of host plant defense and pathogen attack.

Host plant genotypes affect miRNA gene expression in Hessian fly larvae

Project description:MicroRNAs (miRNAs) are small noncoding RNAs that play critical roles in regulating post transcriptional gene expression. Gall midges encompass a large group of insects that are of economic importance and also possess fascinating biological traits. The gall midge Mayetiola destructor, commonly known as the Hessian fly, is a model organism for studying gall midge biology and insect – host plant interactions. In this study, we systematically analyzed miRNAs from the Hessian fly. Deep-sequencing a Hessian fly larval transcriptome led to the identification of 89 miRNA species that are either identical or very similar to known miRNAs from other insects, and 184 novel miRNAs that have not been reported from other species. Microarray analyses revealed the expression of miRNA genes was strictly regulated during Hessian fly larval development and abundance of many miRNA genes were affected by host genotypes. The identification of a large number of miRNAs for the first time from a gall midge provides a foundation for further studies of miRNA functions in gall midge biology and behavior.

Pyrosequencing-based transcriptome analysis of the asian rice gall midge reveals differential response during compatible and incompatible interaction.

Project description:The Asian rice gall midge (Orseolia oryzae) is a major pest responsible for immense loss in rice productivity. Currently, very little knowledge exists with regard to this insect at the molecular level. The present study was initiated with the aim of developing molecular resources as well as identifying alterations at the transcriptome level in the gall midge maggots that are in a compatible (SH) or in an incompatible interaction (RH) with their rice host. Roche 454 pyrosequencing strategy was used to develop both transcriptomics and genomics resources that led to the identification of 79,028 and 85,395 EST sequences from gall midge biotype 4 (GMB4) maggots feeding on a susceptible and resistant rice variety, TN1 (SH) and Suraksha (RH), respectively. Comparative transcriptome analysis of the maggots in SH and RH revealed over-representation of transcripts from proteolysis and protein phosphorylation in maggots from RH. In contrast, over-representation of transcripts for translation, regulation of transcription and transcripts involved in electron transport chain were observed in maggots from SH. This investigation, besides unveiling various mechanisms underlying insect-plant interactions, will also lead to a better understanding of strategies adopted by insects in general, and the Asian rice gall midge in particular, to overcome host defense.
