ABSTRACT: Female and male pigs were studied. Half of the pigs were exposed to immune activation from virally-infected mothers, and half were not exposed. At 21 days of age half the pigs were weaned and the remainder remained with the sow.

Evaluation of the effect of stresses due to weaning and immune activation on pigs

Project description:Weaning is a stressor that can have significant impacts on health and physiology of the offspring. The effect of this stressor may be modulated by the exposure to other stressors during gestation such as the inflammatory signals of the mother in response to infection. The goal of this study is to determine the combined effects of weaning stress and immune stress during gestation on gene expression in the amygdala, a brain region critical for decision-making and aggression. A RNA-seq study of the amygdala of female and male pigs was undertaken. Within sex group, half of the pigs were weaned at 21 d of age and the rest remain with the gilts and were nursed. Also, half of the pigs in each group were raised from gilts exposed to a viral infection and half were raised by gilts that served as control.

Effects of starter microbiota and early life feeding of medium chain triglycerides on the gastric transcriptome profile of 3 weeks old caesarean derived pigs

Project description:An early settlement of a complex gut microbiota can protect against gastro-intestinal dysbiosis, but the effects of neonatal microbiota colonization and early life feeding of medium chain triglycerides on the maturation of the porcine gastric mucosa are largely unknown. The transcriptome of the oxyntic mucosa of 24 caesarian-derived pigs previously associated with microbiota of different complexity and fed a diet fortified or not with medium chain fatty acids was studied. Pigs received pasteurized sow colostrum at birth (d0), 2 mL of starter microbiota (10^7 CFU of each Lactob. Amylovorus (LAM), Clostr. glycolicum, and Parabacteroides spp.) on d1-d3 of age and either a placebo inoculant (simple association, SA) or an inoculant consisting of diluted feces of an adult sow (complex association, CA) on d3-d4 of age. Then half of pigs was fed a moist diet (CON) or, for the remaining half, CTRL fortified in medium chain triglycerides with 7% coconut oil ( MCT). Gastric samples were obtained at on euthanised pigs at 3 weeks of age.

Transcription profiling by array of pig liver from neonatal pigs fed milk and soy formula

Project description:The majority of babies in the US are formula-fed instead of breast fed. There are major differences in the composition of formulas and breast milk and yet little is known about metabolic differences in babies as the result of feeding these very different diets and how that might affect development or disease risk in later life. One concern is that soy-based formulas might have adverse health effects in babies as a result of the presence of low levels of estrogenic phytochemicals genistein and daidzein which are normally present in soy beans. In the current study, we used a piglet model to look at this question. Piglets were either fed breast milk from the sow or were fed two different infant formulas (cow's milk-based or soy-based) from age 2 days to 21 days when pigs are normally weaned onto solid food. Blood glucose and lipids were measured. Formula-fed pigs were found to have lower cholesterol than breast fed piglets and in addition had larger stores of iron in their liver.Microarray analysis was carried out to see if changes in liver gene expression could explain these effects of formula feeding. It was found that overall gene expression profiles were influenced by formula feeding compared to breast fed neonates. Gender-independent and unique effects of formula influenced cholesterol and iron metabolism. Further, soy formula feeding in comparison to milk-based formula failed to reveal any estrogenic actions on hepatic gene expression in either male or female pigs. Effect of a progressive weaning on behavioural and physiologic indicators in foals (Equus caballus)

Project description:The weaning process has been identified as being associated with stress, weight loss and stereotypies. To limit these problems, some breeders wean the foals gradually. This article presents the effects of a progressive weaning on various physiological parameters (cortisol, C reactive protein, length of telomeres) and on the blood transcriptome, among the foals and the mares. Results indicate that a progressive weaning has beneficial short-term effects in foals, and that in the longer term the foals weaned abruptly have compensated this gap. In mothers, the beneficial effects of the progressive weaning are less important.

Transcript profiles in longissimus dorsi muscle and subcutaneous adipose tissue: A comparison of pigs with different post-weaning growth rates.

Project description:The objective of this experiment was to use transcriptional profiling of skeletal muscle and adipose tissue to develop a better understanding of the metabolic basis for poor weaned-pig transition. A total of 1,054 pigs were reared in commercial conditions and weighed at birth, weaning, and 3 weeks post- weaning. Transition average daily gain (tADG) was calculated as the average daily gain for the 3-week period post-weaning. Nine pigs from each of the lowest 10th percentile (low tADG) and the 60th-70th percentile (high tADG) were harvested at 3 weeks post-weaning. Differential expression analysis was conduced in both tissues using RNA-Seq methodology
