Project description:Cellular senescence, an irreversible proliferative arrest, functions in tissue remodeling during development and is implicated in multiple aging-associated diseases. While senescent cells often manifest an array of senescence-associated phenotypes, such as cell cycle arrest, altered heterochromatin architecture, reprogrammed metabolism and senescence-associated secretory phenotype(SASP), the identification of senescence cells has been hindered by lack of specific and universal biomarkers. To systematically identify universal biomarkers of cellular senescence, we integrated multiple transcriptome data sets of senescent cells obtained through different in vitro manipulation modes as well as age-related gene expression data of human tissues. Our analysis showed that RRAD (Ras-related associated with diabetes) expression is up-regulated in all the manipulation modes and increases with age in human skin and adipose tissues.

Project description:Cellular senescence is described as an irreversible cell cycle arrest induced in response to various stresses. Senescent cells are characterised by heterogeneous signalling alterations, complex secretory phenotype, known as senescence-associated secretory phenotype (SASP), and diverse transcriptomic profile. With the aim to investigate senescence heterogeneity and identify conserved transctiptomic alterations and universal senescence markers, we performed RNA-seq and multiplex proteomic analysis in proteasome inhibition-induced and stress-induced premature senescence models of HFL1 and BJ human fibroblasts. Our data revealed diverse transcriptomic signatures, but also, 231 common differentially expressed genes related to cell division and ECM remodelling, and enriched pathways that remained conserved among the different models with senescence onset. Moreover, we identified a subset of proteins and validated them in replicative senescent models. These proteins are involved in cell cycle arrest and promote a pro-inflammatory environment in premature and replicative senescence models. We suggest that the simultaneous analysis of p21, p-c-JUN, BCL-xL and survivin in cellular lysates, and IL-8, GM-CSF, GDF-15 and GROa in culture supernatants can provide a powerful tool for the identification and monitoring of senescent cells and can support the assessment of the efficacy of potential senotherapeutic approaches.

Project description:Cellular senescence plays critical roles in aging, regeneration, and disease, yet the ability to discern its contributions across various cell types to pathophysiological processes in vivo remains limited. In this study, we generated an in vivotoolbox, consisting of three p16INK4a-related intersectional genetic systems, enabling pulse- chase tracing (Sn-pTracer), Cre-based tracing and ablation (Sn-cTracer), and gene manipulation combined with tracing (Sn-gTracer) of defined senescent cell types. Using liver injury and repair as an example, we found that macrophages and endothelial cells (ECs) represent distinct senescent cell populations with different fates and functions during liver fibrosis and repair.Targeted reprogramming of p16INK4a+ ECs through Kdr overexpression markedly reduces liver fibrosis. This study illuminates the pathophysiological diversity of senescent cells and offers valuable insights for the development of future cell type-specific senolytic therapies. Furthermore, our model provides a versatile platform for investigating cellular senescence in any desired cell type.

Project description:Senescent cells accumulate in tissues over time, favoring the onset and progression of multiple age-related diseases. The senescence-associated secretory phenotype (SASP) is considered the main pathological feature of senescent cells, however, little is known about the mechanism of secretion of SASP factors. Here, we report that human senescent cells present higher levels of the two main types of authophagy, namely macroautophagy (MA) and chaperone-mediated autophagy (CMA), together with an increase in the transcription factor TFEB and in lysosomal mass. Proteomic analyses showed profound and widespread changes in the composition of lysosomal substrates in both CMA- and MA-derived lysosomes. Notably, some proteins previously identified as plasma biomarkers of aging in humans are highly enriched in the lysosomes of senescent cells, as it is the case of cathepsin D (CTSD). Moreover, we found that senescent cells have the ability to perform lysosomal exocytosis in a RAB27A-dependent manner and this is the main route for the secretion of multiple cytokines (such as CCL2, CCL3, CXCL12), the protease inhibitor SERPINE1 and protease CTSD. We conclude that lysosomes are hyperactive in senescent cells, display an altered pattern of constitutive proteins and lysosomal substrates, and undergo exocytosis contributing to the SAS

Project description:The accumulation of senescent cells behaves as a key driver of several age-related cancers and degenerative diseases of the general population. Whether cellular senescence also contributes to psychiatric diseases is still elusive. We report here the characterization of a zebrafish model of psychiatric disorder resulting from the invalidation of the ppp2r2c gene known to be involved in various psychiatric pathologies in human and encoding a neural specific regulatory subunit of the Protein Phosphatase 2A (PP2A). We found that the behavioral abnormalities of adult ppp2r2c-defficient fish were associated to neural cell senescence and that they can be reversed by invalidating the tp53 gene or by treating the fish with a drug that eliminates senescent cells. A molecule commonly used to treat ADHD patients, the methylphenidate, decreases the number of senescent neural cells both in adult PPP2R2C-defficient fish and in wild-type old fish. At the molecular level, methylphenidate attenuates the DNA damage response. We propose that some of the drugs commonly used in neuropsychiatry such as methylphenidate can prevent the appearance of senescent neural cells and that general strategies to clear senescent cells are promising therapies for common psychiatric disorders.

Project description:Cellular senescence is a stress response that activates innate immunity. However, the interplay between senescent cells and the adaptive immune system remains largely unexplored. Here, we show that senescent cells display enhanced MHC class I (MHC-I) antigen processing and presentation. Immunization of mice with senescent syngeneic fibroblasts generates CD8 T cells reactive against both normal and senescent fibroblasts, some of them targeting senescence-associated MHC-I-peptides. In the context of cancer, we demonstrate that senescent cancer cells trigger strong anti-tumor protection mediated by antigen-presenting cells and CD8 T cells. This response is superior to the protection elicited by cells undergoing immunogenic cell death. Finally, induction of senescence in patient-derived cancer cells exacerbates the activation of autologous tumor-reactive CD8 tumor-infiltrating lymphocytes (TILs) with no effect on non-reactive TILs. Our study indicates that immunization with senescent cancer cells strongly activates anti-tumor immunity, and this can be exploited for cancer therapy.
