Effect of treatment with Levamisole 1 mM on activated human T-cells

ABSTRACT: To investigate the effects of levamisole treatment on in vitro activated human T-cells, we isolated T-cells from buffy coats from 3 donors. All buffy coats were collected from adult anonymous healthy blood donors (Sanquin Blood Supply, Amsterdam, The Netherlands) who provided written informed consent for usage of the remainder of their blood donation in research. Untouched T cells were isolated from PBMCs by negative Magnetic-activated cell sorting (MACS) using the human pan T cell isolation kit (Miltenyi Biotec) according to the manufacturer’s instructions. Purity of the T cell fraction was assessed by flowcytometry for CD3 (>98% for all donors). To investigate the effects of Levamisole treatment on the T-cell immune response we performed bulk RNA sequencing on 24-hour and 48-hour aCD3/aCD28 dynabeads activated T-cells (Gibco, Thermo Fisher Scientific). Levamisole samples were treated with levamisole 1 mM while control samples were treated with the appropriate solvent controls. 3 separate donors were included in this experiment.

Project description:To increase the dataset of our proteomic naive CD4+ T cell surface atlas, we applied whole genome microarray expression analysis. The NCBI RefSeq accession number of all transcripts which were detected in naive and stimulated CD4+ T cells (aCD3/aCD28 for 3h) of four different donors, was mapped to the corresponding UniProtKB accession number. For these UniProtKB ACs, we extracted the subcellular localization (UniProt_SL) annotation from the UniProtKB/Swiss-Prot database or if not available the subcellular localization was predicted unsing LocTree3 and PolyPhobius. This led to the identification of 908 genes coding for proteins located on the surface of human naive and/or activated CD4+ T cells. Gene expression in human naive T cells and naive T cells which were stimulated with aCD3/aCD28 for 3h was measured. Naive T cells from four different human blood donors were analyzed.

Transcriptome analysis of primary human monocyte differentiation

Project description:In order investigate the control of genes encoding cytoskeletal motor proteins and their interaction partners in the activation program of primary monocytes, we performed whole transcriptome microarray profiling of ex vivo monocyte differentiation to activated macrophages or dendritic cells in monocytes isolated from three anonymous blood donors. Monocytes were isolated from the buffy coats of three anonymous blood donors and differentiated into activated macrophages or activated dendritic cells (DCs) ex vivo. Total RNA from monocytes, activated macrophages or DCs was isolated and submitted for whole transcriptome microarray analysis.

Transcriptional effects of IL-10 neutralization on LPS-activated M-CSF-derived macrophages

Project description:Analysis of the role of IL-10 on the transcriptional signature of LPS-activated human M-CSF-dependent monocyte-derived macrophages. Methods: Human Peripheral Blood Mononuclear Cells (PBMC) were isolated from buffy coats from donors over a Lymphoprep gradient according to standard procedures. Monocytes were purified from PBMC by magnetic cell sorting using anti-CD14 microbeads (>95% CD14+ cells). Monocytes from three independent donors (0.5 x 106 cells/ml, >95% CD14+ cells) were cultured in RPMI 1640 supplemented with 10% fetal bovine serum (FBS) at 37°C in a humidified atmosphere with 5% CO2 and 21% O2 for 7 days in the presence of 1000 U/ml M-CSF, with cytokine addition every two days. Then, monocyte-derived macrophages were treated with an anti-IL-10 blocking antibody (anti-IL10) or an isotype-matched antibody (IgG2b) at 2.5 micrograms/ml for 1 hour, and exposed to LPS (10 ng/ml). After 4 hours of LPS treatment, cells were lysed and RNA isolated for transcriptional analysis.

Project description:Analysis of the role of LXR on the transcriptional signature of monocyte-derived macrophages generated in the presence of tumor ascitic fluids or rheumatoid arthritis synovial fluids. Methods: Human Peripheral Blood Mononuclear Cells (PBMC) were isolated from buffy coats from donors over a Lymphoprep gradient according to standard procedures. Monocytes were purified from PBMC by magnetic cell sorting using anti-CD14 microbeads (>95% CD14+ cells). Monocytes from independent donors (0.5 x 106 cells/ml, >95% CD14+ cells) were treated with DMSO (vehicle), 1 micromolar GW3965 or 1 micromolar GSK2033 for 1 hour, and then cultured in RPMI 1640 10% FBS supplemented with either 50% Tumor-derived Ascitic Fluid (TAF) or 20% Synovial Fluid from patients with active Rheumatoid Arthritis (RASF), at 37°C in a humidified atmosphere with 5% CO2 and 21% O2. After 72 hours, cells were lysed and RNA isolated for transcriptional analysis.
