ABSTRACT: To investigate the influence of the feed additive (milk replacer) on biological processes occuring in cow mammary parenchyma, mammary fat pad and liver via miRNA-mediated regulation. To this end, we performed microRNA expression profiling using next-generation sequencing.

Different Diets Change the Bovine Milk Extracellular Vesicle-miRNA Expression

Project description:Given that different diets could alter cow milk yield and composition, the effects of different feed formula on milk extracellular vesicle (EV) miRNAs were detected. Cow milk EVs contained various small RNAs, including miRNAs, snRNAs, tiRNAs, Cis-regulatory elements, and piRNAs. Two hundred and seventy-six known bos taurus miRNAs were identified by sequencing in bovine milk EVs. There were 13 immune-related miRNAs in the top 20 miRNAs in milk EVs. Nine differently expressed known miRNAs were detected in responding to different feed formulations. Cow milk EVs are abundant of small RNAs, especially miRNAs, which might be closely related to the development of maternal mammary gland and neonatal immune maturity.

Dairy cows fed a low energy diet before dry-off show signs of hunger despite ad libitum access.

Project description:Drying-off is one important management step in commercial dairy farms and consists of ceasing milk production artificially at a specific point in time, generally 2 months before the next calving. Drying-off typically comprises dietary changes as well as gradual or abrupt changes in daily milking frequency, which may challenge the welfare of high-yielding cows. This study investigated the isolated and combined effects of different feed energy densities (normal lactation diet versus energy-reduced diet, both offered ad libitum) and daily milking frequencies (twice versus once) on the feeding motivation of dairy cows on two separate days prior to dry-off (i.e. the day of last milking) using a push-gate feeder. During both days, cows on the energy-reduced diet pushed more than five times more weight to earn the final feed reward and were nearly ten times faster to feed on the first reward than cows on the normal lactation diet. Illustrating the importance of developing more animal welfare-friendly dry-off management, these results illustrate that cows show signs of hunger prior to dry-off when provided a diet with reduced energy density, although offered for ad libitum intake.

Project description:Female Ames dwarf mouse (df/df) with ad libitum access to water and standard pelleted food (LabDiet, PMI Feeds, Inc., St Louis, MO) caged with microisolator filter tops. Mice were killed between 24 to 26 months of age, tissues removed, rapidly frozen on dry ice, and stored in liquid nitrogen. Total liver RNA was isolated from frozen tissue as described (T. Tsuchiya, J.M. Dhahbi, X. Cui, P.L. Mote, A. Bartke, S.R. Spindler, Physiological Genomics, Submitted). mRNA levels were measured using the Affymetrix mouse U74Av2 array according to standard protocols. After hybridization, arrays were scanned using a Hewlett-Packard GeneArray Scanner. Image analysis was performed as described (Cao SX, Dhahbi JM, Mote PL, and Spindler SR.
