Generation and characterization of Myc knockin mouse model of osteosarcoma

ABSTRACT: To understand the mechanisms that underlie the ability of Myc to alter both the tumor and its surrounding tumor microenvironment (TME) of osteosarcoma, we generated and molecularly characterized an osteoblast-specific Cre-Lox-Stop-Lox;(LSL)-c-MycT58A;p53fl/+ knockin genetically engineered mouse model (GEMM). Phenotypically, the Myc knockin-GEMM had rapid tumor development with a high incidence of metastasis.

Undifferentiated Pleomorphic Sarcoma Model

Project description:Analysis of undifferentiated pleomorphic sarcoma/malignant fibrous histiocytoma-like tumors from LSL-KrasG12D, p53Fl/Fl mouse model of soft tissue sarcoma. Murine soft tissue sarcomas (n = 17) were compared to normal muscle (n = 4). Tumors were isolated surgically from soft tissue sarcomas generated by conditional Kras and p53 alleles. Tumors were induced using an adenovirus expressing Cre recombinase. Normal muscle samples were isolated from mice of the same genotype without tumor induction.

MicroRNA profile of tumorigenic cells during formation of pulmonary adenocarcinoma

Project description:Study designed to obtain microRNA profile of tumorigenic Sca-1+CD34+ cells during the formation of pulmonary adenocarcinoma. After intranasal infection with adenovirus CRE (Ad-Cre), lung adenocarcinoma was identified pathologically in Lox-stop-lox Kras (LSL-Kras) G12D mice. Sca-1+CD34+ cells were sorted by flow cytometry and tested for the tumor-initiating ability to undergo self-renewal and differentiation. MiRNA profile was obtained from the microarray and further confirmed by qRT-PCR.The function of miRNAs was predicted bioinformatically, and miR-294 was verified to explore its relationship with tumor migration and invasion.

A genetically engineered mouse model of NUT carcinoma (CUT&RUN)

Project description:We created a genetically engineered mouse model (GEMM) of NC that forms a Brd4-NUTM1 fusion gene upon tamoxifen-induction of Sox2-driven Cre. Two GEMM-derived cell lines were developed whose transcriptomic and epigenetic landscapes, characterized by RNAseq and CUT&RUN, show striking overlap with those of primary GEMM tumors. GEMM primary tumor and cell lines form very large H3K27ac-enriched super-enhancers that are unique to hNC, termed megadomains, that are invariably associated with key hNC-defining transcriptional oncogenic targets, Myc and Trp63.

Conditional expression of oncogenic KrasG12D and HrasG12D alleles in mouse keratinocytes reveals a dose dependent requirement for tumor formation.

Project description:The discovery that activation of the Hras gene was the driver mutation for tumor development in mouse skin treated with 7,12-dimethylbenz(a)anthracene illuminated the link between environmental and molecular carcinogenesis. Subsequent studies revealed that activating mutations in the closely related Kras gene could also drive skin tumor development and progression under certain conditions. To address a head to head comparison of downstream signaling produced by each activated oncogene primary keratinocytes were isolated from the LSL-HrasG12D and LSL -KrasG12D C57BL/6J mouse models that carry conditionally expressed mutant alleles when a Lox-STOP-Lox segment is excised by Cre recombinase adenovirus treatment. Keratinocytes expressing two mutant alleles of HrasG12D (HH), only one mutant allele of HrasG12D (H), one mutant allele of KrasG12D (K) or one mutant allele of each (HK) were evaluated for differences in downstream signaling, gene expression and the ability to form squamous papillomas orthotopically in vivo.

Choroid Plexus Gene Expression in ApoE3 or ApoE4-Knockin Mouse Choroid Plexuses

Project description:Laser capture microdissected choroid plexuses were obtained and expression arrays were generated to investigate gene expression in ApoE-Knockin choroid plexuses; the choroid plexus forms the cerebrospinal fluid, the cerebrospinal fliod barrier, functions as the major gateway for blood-born leukocytes to enter the brain in degenerative and inflammatory brain diseases, and the principal neuroimmune interface in the brain. We found lipid deposits in the aged choroid plexus of hyperlipidemic ApoE4-Knockin mice but none in normolipidemic ApoE4-Knockin or normolipidemic or hyperlipidemic ApoE3-Knockin control choroid plexuses. Here, we studied the functional impact and gene epressions these choroid plexuses.
