Transcriptional chracterization of ABCC1 and ABCB1 KO and MZ1 treated WT HAP1 cell lines.

ABSTRACT: We used CRISPR/Cas9 to intoroduce frame-shifting mutations into the ABCC1 and ABCB1 genes and treated HAP1 WT cells with varying concentrations of the MZ1 PROTAC. To measure the transcriptional response to both genetic and chemical perturbances, we performed RNA-seq and quantified gene expression levels.

Project description:MZ1 is a newly designed pan-BET-targeting PROTAC that binds to target proteins (BET proteins such as BRD2, BRD3, and BRD4) and recruits them to the ubiquitin/protease pathway for selective destruction. However, the role of MZ1 in NB models has yet to be determined. The effect of MZ1 on NB cells was investigated using RNA-seq analysis in this work. MZ1 is a newly designed pan-BET-targeting PROTAC that binds to target proteins (BET proteins such as BRD2, BRD3, and BRD4) and recruits them to the ubiquitin/protease pathway for selective destruction. Gene expression analysis of human haploid cells (HAP1) depleted of SMARCB1 and SMARCA4

Project description:The aim of this study is to analyze the change in genome wide expression levels in HAP1 cells upon loss of SMARCB1, SMARCA4 or both these genes together. The SMARCB1 and SMARCA4 genes were the hits from a genome wide screen involving genetrap mutagenesis to find new players that are involved in sensitivity to Doxorubicin (Dox). It was found that loss of SMARCB1 and SMARCA4 genes impart resistance in HAP1 cells to Dox. To validate this, the genes were knocked out in HAP1 cells with CRISPR-Cas9 technology. Gene expression levels in SMARCB1 null, SMARCA4 null and SMARCB1-SMARCA4 double null cells were compared to wildtype HAP1 cells using RNAseq. From these experiments it was found that SMARCB1 loss caused several fold increase in ABCB1 gene levels. ABCB1 is an efflux pump in cells responsible for flushing out many small-molecule drugs. Further analysis of this gene confirmed that ABCB1 was the main factor responsible for Dox resistance upon SMARCB1 loss. In total there are four different cell types with two replicates for each cell type. Therefore, 8 samples in total.

Project description:Glioblastoma (GBM) is a relatively more common primary central nervous system tumor with a high degree of malignancy, high mortality, and complex surgical complete resection. MZ1 is a von Hippel-Lindau tumor suppressor (VHL)-based pan-BET-targeting PROTAC, which can bind to the target proteins (BET proteins, including BRD2, BRD3, and BRD4) and recruit them to the ubiquitin/ proteasome system for degradation. However, the function of MZ1 has not been assessed in GBM cells so far. In the present study, ChIP-Seq analysis was performed to explore the effect of MZ1 on GBM cells.

Exploring the cytotoxic effects of MZ1 on multiple molecular subtypes of B-cell acute lymphoblastic leukemia cells

Project description:Acute lymphoblastic leukemia (ALL) is the most common childhood malignancy. B-cell acute myeloid leukemia (B-ALL) predominates in ALL, with current cure rates of ∼80%. However, the long-term survival rate of patients with early relapse or refractory B-ALL is very low. In recent years, Bromodomains and extra-terminal (BET) protein inhibitors have shown considerable prospect in hematological tumors. MZ1 is a novel BET inhibitor that degrades target proteins and inhibits tumor growth through proteolysis-targeting chimeras (PROTAC) technology. This study shows that MZ1 has cytotoxic effects on 697 (TCF3/PBX1) and RS4;11 (MLL-AF4) B-ALL cell lines representing different molecular subtypes of B-ALL. Furthermore, we observed that MZ1 could promote cell apoptosis, induce cells cycle arrest and inhibit B-ALL cell proliferation by depleting BET protein and downregulating the transcription of CCND3. Collectively, our results indicate that MZ1 might be exploited as a novel therapeutic strategy for the treatment of B-ALL.
