HFD versus LFD [RNA-seq]

ABSTRACT: Obesity alters the transcriptional profile of the adipose tissue. Our aim was to characterize the metabolic adaptations induced by diet-induced obesity in the adipose tissue. To this end wild-type mice were fed for 20 weeks a high-fat diet and subcutaneous adipose tissue was analyzed by RNA sequencing.

Human s.c adipose tissue gene expression during diet-induced weight loss

Project description:Obesity has considerable effects on morbidity and mortality and the prevalence of obesity has been increasing rapidly worldwide during the past two decades. Even if obesity affects the entire individual, adipose tissue play a central role in the development of obesity. Expression profiling of adipose tissue may give insights into mechanisms contributing to obesity and obesity-related disorders. 24 obese subjects (6 women and 18 men, BMI 37.6 ± 4.9) received a very low calorie diet (450 kcal/d, Cambridge diet or Modifast) for 16 weeks. On average, patients lost 27.7 kg during the diet. Microarray expression analysis (Affymetrix U133A) in subcutaneous adipose tissue was performed before diet and after 16 weeks of diet.

Project description:The metabolic syndrome (MetS) is characterized by the presence of metabolic abnormalities that include abdominal obesity, dyslipidemia, hypertension, increased blood glucose/insulin resistance, hypertriglyceridemia and increased risk for cardiovascular disease (CVD). The ApoE*3Leiden.human Cholesteryl Ester Transfer Protein (ApoE3L.CETP) mouse model manifests several features of the MetS upon high fat diet (HFD) feeding. Moreover, the physiological changes in the white adipose tissue (WAT) contribute to MetS comorbidities. The aim of this study was to identify transcriptomic signatures in the gonadal WAT of ApoE3L.CETP mice in discrete stages of diet-induced MetS.

Adipose tissue biology and effect of weight loss in women with lipedema

Project description:Lipedema is a lipodystrophic disease characterized by marked increases in lower-body subcutaneous adipose tissue, anecdotally reported to: i) increase inflammation and fibrosis and impair microvascular and lymphatic circulation in the affected adipose tissue, ii) reduce risk of developing obesity-related cardiometabolic abnormalities; and iii) be resistant to diet-induced weight loss. To further our understanding of lipedema, we examined body composition, metabolic health and adipose tissue bology in women with obesity and lipedema (Obese-LIP) at baseline and following ~9% diet-induced weight loss. At baseline, people with Obese-LIP had ~23% greater leg fat mass, ~11% lower android-to-gynoid ratio and ~54% greater insulin sensitivity compared to women matched on age, body mass index and whole-body fat mass. In the Obese-LIP group, total and proinflammatory macrophage content and expression of inflammation and fibrosis-related genes were greater while lymph/angiogenesis-related genes were lower in subcutaneous femoral compared to abdominal adipose tissue. Diet-induced weight loss improved insulin sensitivity and decreased total fat mass due to similar reductions in abdominal and leg fat masses, with minimal effect on markers of adipose tissue inflammation/fibrosis and lymph/angiogenesis. Our study provides important insights into the pathophysiology of lipedema and suggests diet-induced weight loss should be the cornerstone therapy in people with Obese-LIP.

Role of Adipocyte Na,K-ATPase Oxidant Amplification Loop in Cognitive Decline and Neurodegeneration - Expression profiling in Liver and Fat

Project description:Adipocyte-specific expression of Na,K-ATPase signaling antagonist, NaKtide, has been shown to ameliorate the pathophysiological consequences of obesity and improve diseased phenotype. Recent studies have established that the release of adipocytokines and other bioactive mediators in obesity contributes to neurodegeneration. This study utilizes a mechanistic approach and characterizes the hippocampal transcriptome in comparison with transcriptomic changes in liver, visceral and subcutaneous adipose tissue. RNAseq analysis in high fat diet fed transgenic mice showed large scale differential gene expression as well as modulation of several biological pathways in hippocampus, liver, visceral and subcutaneous adipose tissue, which were attenuated by doxycycline induced adipocyte specific NaKtide expression.

Cx3cl1 expression in the adipose tissue is downregulated during development of obesity

Project description:To assess changes in expression level of various chemokines and their receptors on diet-induced obesity, we analysed gene expression in adipose tissue of C56BL/6J mice fed a high-fat (HF) diet or normal chow diet for 8 weeks. HF diet-induced obese (DIO) mice showed adipose tissue inflammation and insulin resistance. Comprehensive gene expression analysis showed that MCP-1–CCR2 and CCL5–CCR5 signalling in epididymal white adipose tissue (eWAT) were enhanced during the development of obesity. Surprisingly, the gene expression of Cx3cl1 was decreased in the eWAT of DIO mice compared with lean mice. While Cx3cr1 expression showed no significant difference between DIO and lean mice. Decreased CX3CL1-CX3CR1 signalling in adipose tissue may also be involved in the development of obesity-induced adipose tissue inflammation and insulin resistance.

MicroRNAs Regulated by Eicosapentaenoic Acid (EPA) in Brown, Subcutaneous, and visceral Adipose Tissue of Diet-Induced Obese Mice

Project description:To understand differences in microRNA (miRNA) signatures between two different diets with and without EPA in brown, subcutaneous, and viscerl tissue from C57BL/6 mice to understand mechanistic insight regarding their contribution to metabolic disorders in obesity. We performed small RNA-sequencing of brown, subcutaneous adipose from high fat diet (45% kcal from fat) and high fat diet supplemented with EPA (45% Kcal from fat, 6.75% EPA). Using the Gunaratne Next Generation pipeline (published in Creighton et al. 2009) miRNA expression profiles were identified. Counts of each unique read were normalized to total usable reads, and had 40 counts added. We mapped about 13.8 million sequence reads per sample to the Mus musculus genome (build mm 10). AS a total 1251 miRNAs were identified in three adipose tissue and out of which in bown adipose tissue 15 showd differential expression between BF-HF and BF-EPA .IN subcutaneous adipose tissue 3 miRNAs showed differntial expression between SUB-HF and SUB-EPA. EPA differentially regulate specific miRNAs expression in brown, subcutaneous, and visceral adipose tissue.

Project description:The arrays were used to explore how parents’ obesity status influence their offspring’s weight. We randomly assigned three-week-old mice to two groups, one group receiving a high-fat diet (HFD), the other group receiving a control diet (chow). Adult females of both groups were mated to males fed with HFD or CD. F1 transcriptome assay data were created from four tissues (liver, epigonal visceral, inguinal subcutaneous, and interscapular brown adipose tissue) of male offspring in relation to their parents’ obesity status.

Time-course microarrays reveal early activation of the immune transcriptome and adipokine dysregulation leads to fibrosis in visceral adipose depots during diet-induced obesity

Project description:Time-course analysis of adipocyte gene expression profiles response to high fat diet. The hypothesis tested in the present study was that in diet-induced obesity, early activation of TLR-mediated inflammatory signaling cascades by CD antigen genes, leads to increased expression of pro-inflammatory cytokines and chemokines, resulting in chronic low-grade inflammation. Early changes in collagen genes may trigger the accumulation of ECM components, promoting fibrosis in the later stages of diet-induced obesity. New therapeutic approaches targeting visceral adipose tissue genes altered early by HFD feeding may help ameliorate the deleterious effects of a diet-induced obesity. Total RNA obtained from isolated epididymal and mesenteric adipose tissue of C57BL/6J mice fed normal diet or high fat diet for 2, 4, 8, 20 and 24weeks
