Circular RNA circASH1L protects miRNA-129-5p from target-directed microRNA degradation in human skin wound healing [circASH1L knockdown]

ABSTRACT: In an RNAseq analysis, we have identified the circular RNA cASH1L with high levels in acute wounds (day1 to day7) compared to normal skin. The biological function of cASH1L in human epidermal keratinocytes during wound repair has not been studied. To study the genes regulated by cASH1L, we transfected siRNA targeting the diagnostic junctions of cASH1L into human primary epidermal keratinocytes to knockdown cASH1L expression. We performed a global transcriptome analysis of keratinocytes upon circRNA knockdown using Affymetrix arrays.
