Development of a new model of Parkinson´s disease

ABSTRACT: To get insight the molecular mechanism underlying neuronal loss in Parkinson´s disease. We performed gene expression profiling analysis using data obtained from RNA-seq of midbrain of mice injected with an AAV control or an AAV expressing human tyrosinase that induces neuromelanin accumulation and subsequent neuronal loss.

Rescuing voluntary motor behavior in a Parkinson´s disease model by aCRISPR mediated reprogramming of striatal astrocytes into induced GABAergic neurons

Project description:Cell replacement strategies of affected cells have been performed in clinical studies of Parkinson´s disease with variable outcome. Conversion of endogenous glial cells into functional neurons might represent a more reliable alternative, thus here we developed a knock-in mouse line carrying a dual dCas9 transactivator system (dCAM), which allowed the conditional activation of the endogenous genes Ascl1, Lmx1a, and Nr4a2 in striatal astrocytes in vivo by delivering sgRNAs specific for their endogenous loci. In order to move this successful approach toward translation, we established an AAV based activator system carrying intein-split-dCas9-VP64, SAM (AAV-dCAS) activators, and the specific sgRNAs targeting transcription factor combinations. Both systems, the dCAM and the AAV-dCAS approach were successful in reprogramming of striatal astrocytes into GABAergic neurons, which functionally integrate into striatal circuits as evidenced by the alleviation of specific motor behaviors in a 6-OHDA Parkinson’s disease model. Thus, the AAV-dCAS approach may enable clinical therapies for Parkinson´s disease by reprogramming striatal astrocytes.

Transcriptome analysis of myeloid cells (CD11b) and astrocytes (ACSA2) from the striatum and midbrain of control and parkinsonian animals

Project description:Inflammation is a common feature in neurodegenerative diseases that participates in the process of neuronal loss. Here, we questioned whether the inflammatory reaction generated in Parkinson´s disease (PD) by dopaminergic neuron degeneration would trigger specific inflammatory reactions in the midbrain and in the striatum that could modify the course of neuronal death. Experimental parkinsonism was induced by overexpressing α-synuclein in the substantia nigra with a viral vector. Bulk RNA sequencing of purified midbrain microglia/myeloid cells showed a phagocytic and anti-inflammatory M2 phenotype, while midbrain astrocytes presented a pro-inflammatory state. In the striatum, microglia but not astrocytes presented a pro-inflammatory state.

Parkinson-associated SNCA enhancer variants revealed by open chromatin in mouse dopamine neurons

Project description:The progressive loss of midbrain (MB) dopaminergic (DA) neurons defines the motor features of Parkinson disease (PD) and modulation of risk by common variation in PD has been well established through GWAS. Anticipating that a fraction of PD-associated genetic variation mediates their effects within this neuronal population, we acquired open chromatin signatures of purified embryonic mouse MB DA neurons. Correlation with >2,300 putative enhancers assayed in mice reveals enrichment for MB cis-regulatory elements (CRE), data reinforced by transgenic analyses of six additional sequences in zebrafish and mice. One CRE, within intron 4 of the familial PD gene SNCA, directs reporter expression in catecholaminergic neurons of transgenic mice and zebrafish. Sequencing of this CRE in 986 PD patients and 992 controls reveals two common variants associated with elevated PD risk. To assess potential mechanisms of action, we identify proteins whose binding is impacted by these enhancer variants. Additional genotyping across the SNCA locus identifies a single PD-associated haplotype, containing the minor alleles of both of the aforementioned PD-risk variants. Our work posits a model for how common variation at SNCA may modulate PD risk and highlights the value of cell context-dependent guided searches for functional non-coding variation.

Project description:High throughput sequencing of poly-A RNA from control- and sporadic Parkinson´s disease patient derived fibroblasts (n=15), induced pluripotent stem cells (n=31) and differentiated midbrain neurons (n=15). Fibroblasts and iPSCs do not show major differences on single gene level. In contrast, midbrain neurons derived from Parkinson´s disease patients show changes known to be associated with neurodegenerative diseases.
