RNA sequencing (RNA-seq) of LAIR-1 overexpression in SKOV-3 ovarian cancer cell lines

ABSTRACT: Leukocyte-associated immunoglobulin-like receptor 1 (LAIR-1), is widely expressed by solid tumor cells and functions as a tumor suppressor. Our previous studies have found that LAIR-1 is expressed in ovarian cancer tumor tissues and in a variety of epithelial ovarian cancer cell lines and that LAIR-1 could inhibit ovarian cancer cell growth by suppressing the PI3K-AKT-mTOR pathway. The aim of the present study was to obtain a comprehensive understanding of the molecular mechanism of LAIR-1 in regulating the biological behavior of ovarian cancer cells. The approach taken was to perform RNA-seq sequencing on SKOV3 cells overexpressing LAIR-1, and to conduct further validation by combinatorial integration of bioinformatics analyses to assess the genetic alterations and using biological experimental findings.

Project description:Type-2 innate lymphoid cells (ILC2s) are central players in type-2 asthma. They initiate eosinophil infiltration and airway hyperreactivity (AHR) through cytokine secretion. Leukocyte-associated immunoglobulin-like receptor 1 (LAIR-1) is an inhibitory receptor considered as immune checkpoint in different inflammatory diseases. Here we assessed the role of LAIR-1 in murine ILC2s. WT and LAIR-1 KO mice were intranasally challenged with IL-33 and pulmonary ILC2s were sorted to perform RNA sequencing. Our results unravel a novel regulatory axis in ILC2s with the capacity to reduce their activation.

Project description:Ovarian cancer has the highest mortality in gynecologic cancers and most patients are diagnosed in advanced stages. EZH2 has been a major tumor markers and effective therapeutic targets for ovarian cancer, which molecular mechanism remain unclear. The present study was investigated the biological effects on EZH2 knock-out SKOV3 cells in vitro and in vivo experiment, and explore the molecular mechanism by integrated analysis of mRNA-seq and CHIP-seq.  CRISPR/Cas9 system was used to establish the knock-out EZH2 SKOV3 cells. Protein expression was detected by Western blot. The effect of EZH2 in ovarian cancer was detected by MTT assay, wound scratch assay and Transwell assay, apoptosis assay in vitro and xenograft model in vivo. mRNA-seq and CHIP-seq was used to explore the molecular mechanism of biological function of EZH2.

Combating pancreatic cancer with ovarian cancer cells

Project description:Pancreatic ductal adenocarcinoma (PDAC) is a highly malignant tumor, and the five-year survival rate is still less than 5%. However, Gemcitabine, a first-line chemotherapy drug of PDAC, has serious side effects and chemotherapy resistance. It is imperative to find new therapies to treat patients with advanced pancreatic cancer. With the computational screening, we found the gene sginature of ovarian serous carcinoma is anti-correlated with that of PDAC. Then we established an animal model of pancreatic cancer xenografts and injected ovarian cancer cell SKOV3 into xenografts to observe whether the SKOV3 could inhibit the growth of pancreatic cancer. We used RNA-sequencing to compare the tumor treated with SKOV3 to that with normal saline.

RNA sequencing approach was used to explore and compare lung macrophage transcriptomic profiles in wild-type and LAIR-1 knock-out mice.
