Gene expression profile of CDH17 silenced human colorectal cancer cell lines KM12SM and SW620

ABSTRACT: CDH17 is a key protein in the development of hepatic metastasis in colorectal cancer. In this study, stable silencing of CDH17 was achieved in the KM12SM and SW620 cell lines using shRNA technology, with the objective of exploring the cellular functions and pathways in which CDH17 is implicated.

Project description:To investigate the role of TGF-M-NM-21-regulated miRNAs in the progression of colorectal cancer,we performed comprehensive miRMA microarray analysis on RNA derived from typical human colorectal cancer cell lines and TGF-M-NM-21 knock-down human colorectal cancer cell lines. We identified a novel set of TGF-M-NM-21-related miRNAs. Transcription profiling by array of human colorectal cancer SW620 cells upon MSX1 depletion.

Project description:The first step in the development of human colorectal cancer (CRC) is the aberrant hyperactivation of the Wnt signaling pathway, predominantly caused by inactivating mutations in the adenomatous polyposis coli (APC) gene encoding an essential tumor suppressor. In order to identify genes affected by Apc loss, expression profiling of intestinal epithelium isolated from mice harboring the conditional allele of the gene was performed. The gene encoding transcriptional factor msh homeobox 1 (Msx1) displayed robust upregulation upon Apc inactivation. Subsequently, the expression pattern of human MSX1 was examined in a collection of colonic tumors. The MSX1 mRNA level was increased in all types of human intestinal neoplasia tested. In order to identify genes affected by MSX1 loss, expression profiling of human colorectal cancer cells SW620 upon MSX1 gene depletion was performed.

RNA-seq of GAP43 overexpressed and control SW620 cancer cells

Project description:We found GAP43 was significantly downregulated in colorectal cancer. So we reintroduced GAP43 to colorectal cancer cell line, and investigated the influence to cancer cells. In order to get the transcriptome-wide impact of GAP43 overexpression in colorectal cancer, we made stable transfected GAP43 in SW620 cancer cells, and mRNA-seq were performed (Sequenced with PE150, and 20M reads were generated.) This study provides the DEGs of GAP43 overexpressed and control colorectal cancer cells.
