Effect of A1 on the transcriptome of mesenchymal stem cells

ABSTRACT: To investigate whether pharmacological stimulation of mesenchymal stromal cells (MSCs) could induce cross-presentation abilities, we treated MSCs with A1. We performed gene expression profiling analysis using data obtained from RNA-seq of untreated MSCs, MSCs treated only with the molecule A1, and MSCs treated with the molecule A1 and the model antigen ovalbumin. A1 is an Accum™ variant, a molecule design to enhance intracellular accumulation of associated molecules through endosomal disruption. Endosomal break was targeted for its crucial role in antigen export to the cytosol and cross-presentation of endocytosed antigen.
