Gene expression changes induced by overexpression of PJA2 in SW-480 cells

ABSTRACT: To investigate the impact of PJA2 on the progression of colorectal cancer, we employed lentiviral constructs to establish PJA2-overexpressing SW-480 cell lines. Subsequently, we conducted gene expression profiling analysis using RNA-seq data obtained from both vector SW-480 cells and PJA2-overexpressing SW-480 cells.

Targeting AGTR1/NF-κB /CXCR4 axis by miR-155 attenuates oncogenesis in Glioblastoma

Project description:MiR-155 show conserved binding on the 3’UTR of AGTR1, subsequently confirmed by AGTR1-3’UTR-luciferase reporter assay. To determine the pathways regulated by miR-155, we overexpressed miR-155 in SNB19 glioblastoma cells using pLemiR lentiviral constructs. pLemiR vector, with turbo RFP (Open Biosystems) was used as control. Furthermore, stable miR-155 overexpressing GBM cells show a decrease in AGTR1-mediated cell proliferation, invasion, foci formation, anchorage-independent growth, Epithelial to Mesenchymal Transition and stemness.

Identification of alpha-satellite RNA interacting proteins

Project description:We set out to identify proteins that bind alpha-satellite ncRNAs in HCT-116 and SW-480 colorectal cancer human cell lines. We performed RNA pulldown followed by mass spectrometry identification of the associated proteins. Among the identified proteins, we observed a statistical overrepresenttion of some ontology ssociated to mitotic progression in HCT-116 cells, but not in SW-480 cells.

Through comparing microarray results between solvent- and SW-treated cells, differentially expressed genes were obtained (>1.5 fold). The microarray results were validated using real-time RT-PCR. We used the KEGG database, STRING database and GO database for further ananlysis, and therefore got the possible signaling pathways underlying the anticancer effect of SW.
