Project description:Circadian and metabolic physiology are intricately intertwined, as illustrated by Rev-erb , a transcription factor (TF) that functions both as a core repressive component of the cell autonomous clock and as a regulator of metabolic genes. Here we show that Rev-erb modulates the clock and metabolism by different genomic mechanisms. Clock control requires Rev-erb to bind directly to the genome at its cognate sites, where it competes with activating ROR TFs. By contrast, Rev-erb regulates metabolic genes primarily by recruiting the HDAC3 corepressor to sites to which it is tethered by cell type-specific transcription factors. Thus, direct competition between Rev-erb and ROR TFs provides a universal mechanism for self-sustained control of molecular clock across all tissues, whereas Rev-erb utilizes lineage-determining factors to convey a tissue-specific epigenomic rhythm that regulates metabolism tailored to the specific need of that tissue. Project description:We study the genomic and developmental basis of the mammalian gliding membrane, or patagium, an adaptative trait that has repeatedly evolved in different lineages, including in closely related marsupial species. Through comparative genomic analysis of fifteen new marsupial genomes, both from gliding and non-gliding species, we find that the Emx2 locus experienced lineage-specific patterns of accelerated cis-regulatory evolution in gliding species. We confirm our finding via epigenomics, transcriptomics, and in vivo marsupial transgenics.

Whereas Geobacter species completely oxidize organic compounds to CO2, Pelobacter species either ferment or oxidize short chain alcohols to acetate. Pelobacter species also contain far fewer c-type cytochromes, proteins which play a role in electron transfer during Fe(III) respiration, compared to their Geobacter counterparts. Project description:Allogrooming is a behaviour in which a worker uses its mouth parts to remove debris from the body of other colony members. This behaviour, observed in several species of eusocial insects, play a role in defence against parasites and pathogens. In honeybees, allogrooming represents an important resistance mechanism that seems to limit ectoparasites load, especially mites, within colonies and its expression depends on genetic and environmental factors. The degree of specialization of individuals performing this task is still unclear and behavioural and physiological correlates of allogroomers are largely unknown. It would be advantageous for allogroomers to be able to detect nestmates needing to be groomed. Since many stressors, including pathogens and parasites, change the odour of workers in A. mellifera, an enhanced perception of such chemical cues could contribute to a possibly specialized phenotype of allogroomers. Indeed, antennae play a key role in the expression of hygienic behaviours and proteomic investigation also showed that in the honeybee individuals performing different tasks differ for antennal profile of soluble olfactory proteins, which play a crucial role in the first steps of odour recognition. We thus investigated the expression of olfactory proteins in the antennae, predicting that if allogroomers have any degree of chemosensory specialization, this would reflect in a different antennal proteomic profile.

Project description:DNA methylation is an important chromatin modification that is necessary for the structural integrity and proper regulation of the genome for many species. Despite its conservation across the tree of life, little is known about its contribution to complex traits. Reports that differences in DNA methylation between castes in closely related Hymenopteran insects (ants, bees and wasps) contributes to social behaviors has generated hypotheses on the role of DNA methylation in governing social behavior. However, social behavior has evolved multiple times across insecta, and a common role of DNA methylation in social behavior remains outstanding. Using phylogenetic comparative methods we sought to better understand patterns of DNA methylation and social behavior across insects. DNA methylation can be found in social and solitary insects from all orders, except Diptera (flies), which suggests a shared loss of DNA methylation within this order. The lack of DNA methylation is reflected in the absence of the maintenance and de novo DNA methyltransferases (DNMT) 1 and 3, respectively. Interestingly, DNA methylation is found in species without DNMT3. DNA methylation and social behavior (social/solitary) or with division of labor (caste+/caste–) for 123 insect species analyzed from 11 orders are not evolutionary dependent, which is further supported by sequencing of DNA methylomes from 40 species.

Here, the proteomes of three sub-organs of eastern and western honeybee brains were compared. In MBs, protein metabolism and Ca2+ transmembrane transport are the key role players in driving the learning and memory by modulating the synaptic structure and signal transduction to consolidate memory trace. In OLs, ribonucleoside metabolism and energy production play major roles to underpin visual system by maintaining G-protein cycle and membrane electrical charge potential. However, in antennal lobes (ALs), it has evolved distinct proteome settings to prime the olfactory learning and memory in two bee species. In ALs of Apis cerana cerana (Acc), actin cytoskeleton organization is key for plasticity of glomeruli and intracellular transport to sustain the olfactory signaling. Whereas, in ALs of Apis mellifera ligustica (Aml), hydrogen and hydrogen ion transport are vital to support olfactory process by supplying energy and maintaining molecule transport. Noticeably, in ALs of Acc, the exclusively enriched functional groups acting as second messenger and neurontransmitter of signal transduction, and the enhanced protein metabolism to regulate the plasticity of synaptic structure for formation of memory, suggest that Acc may have evolved a better sense of smell than that of Aml.
