Single cell RNA sequencing reveals heterogeneity and dynamics of mouse early inguinal white adipose tissue (iWAT) during development

ABSTRACT: In this study, we comprehensively compared the changes in adipose tissue from 1 week (7 d) to 4 weeks (28 d) postnatally. To analyze the different cell groups and how their amount and function vary along with developmental time, we collected iWAT from 7-, 14-, 18-, 21-, and 28-day-old mice and enzymatically disassociated the tissues into single cells and performed single-cell RNA sequencing.

Exercise Training Remodels Inguinal White Adipose Tissue Through Adaptations in Innervation, Vascularization and the Extracellular Matrix

Project description:Inguinal white adipose tissue (iWAT) is essential for the beneficial effects of exercise training on metabolic health. The underlying mechanisms for these effects are not fully understood and here, we test the hypothesis that exercise training results in a more favorable iWAT structural phenotype. Using biochemical, imaging, and multi-omics analyses we find that 11-days of wheel running in male mice causes profound iWAT remodeling including decreased extracellular matrix (ECM) deposition, increased vascularization and innervation. We identify adipose stem cells as one of the main contributors to training-induced ECM remodeling, show that the PRDM16 transcriptional complex is necessary for iWAT remodeling and beiging, and discover neuronal growth regulator 1 (NEGR1) as a link between PRDM16 and neuritogenesis. Moreover, we find that training causes a shift from hypertrophic to insulin-sensitive adipocyte subpopulations. Exercise training leads to remarkable adaptations to iWAT structure and cell-type composition that can confer beneficial changes in tissue metabolism.

Project description:Purpose: The goals of this study are to identify Gs-linked GPCRs that are endogenously expressed by mouse adipose tissue, we subjected RNA prepared from isolated mouse adipocytes (iWAT and eWAT) and BAT tissue to RNA-seq analysis. Methods: Total RNA extracted from mature adipocytes (iWAT and eWAT) and BAT tissue of C57BL/6J mice (16-week old males) maintained on regular chow were used to construct high throughput sequencing libraries. RNAs with RIN >8 (assessed by the Agilent 2100 Bioanalyzer system) were used to prepare transcriptome libraries using the NEBNext Ultra RNA library prep kit (New England Biolabs). High throughput RNA-sequencing was performed using a HiSeq 2500 instrument (Illumina) at the NIDDK Genomic Core Facility (NIH, Bethesda, MD). Raw reads were mapped to the mouse (mm9) genome. GPCRs were extracted from the RNA-seq data using R form. Gs-coupled GPCRs were identified using the IUPHAR/BPS Guide to Pharmacology Database (https://www.guidetopharmacology.org/). Results: Our study demonstrated that mouse adipocytes/BAT tissue express several GPCRs that are selectively coupled to Gs, including the V2 vasopressin receptor, the glucagon receptor, and different melanocortin receptor subtypes.

Differential gene expression analysis in liver, inguinal white adipose tissue (iWAT), skeletal muscle, and hypothalamus of mice with liver-specific Crif1 deficiency

Project description:CR6-interacting factor-1 (CRIF1) interacting with large mitoribosome subunits is essential for the maturation and insertion of oxidative phosphorylation (OxPhos) polypeptides into the mitochondrial inner membrane. Recently, it has reported that the genetic ablation of Crif1 in a tissue-specific manner results in mitochondrial stress response (MSR) including mitochondrial unfolded protein response (UPRmt). In this study, we aim to obtain a comprehensive understanding of systemic energy metabolism in response to hepatic mitochondrial dysfunction. Through the liver-specific Crif1 deficient mice (LKO), we explore the adaptive response not only in the liver but also in inguinal white adipose tissue (iWAT). Moreover, RNA sequencing in liver, iWAT, skeletal muscle, and hypothalamus suggest that hepatic mitochondrial dysfunction enhance metabolic pathways, including glycolysis, fatty acid elongation and degradation, and 1C metabolism in liver and insulin signaling in iWAT. RNA sequencing also suggests that hepato-mitokines, GDF15 and FGF21 are highly increased in liver of LKO mice. To identify the role of hepato-mitokines, we generate double knockout mice (LKO/Gdf15-/- and LKO/Fgf21-/-), suggesting that GDF15 is responsible for the regulation of the body and fat mass and FGF21 has a critical role for the insulin sensitivity and energy expenditure in this mice.

Effect of chronic cold on gene expression in inguinal white adipose tissue.

Project description:Excess weight is associated with an increased risk of insulin resistance and type 2 diabetes. Activating thermogenic mechanisms is a potentially powerful means to reduce excess weight and achieve metabolic benefits. Recent studies show that cold stimuli trigger the conversion of white fat into beige fat, which can mediate non-shivering thermogenesis. The purpose of this study is to determine effect of chronic cold exposure on gene expression in iWAT. To this end, we performed RNA-sequencing of iWAT from mice at 30 ˚C or 7 ˚C for 3 weeks and identified significantly regulated genes under chronic cold in iWAT.

RNA-Seq analysis of inguinal white adipose tissue (iWAT) from control and high fat diet mice treated with THCA and THCA+T007

Project description:Medicinal cannabis has garnered worldwide attention in recent years but has been hampered by the psychotropic activity of Δ9-tetrahydro-cannabinol (Δ9-THC). However, the biological activity of its precursor Δ9-THC acid (Δ9-THCA) remain largely unexplored; yet, it is known that Δ9-THCA is not psychotropic and displays PPARg agonistic activity. We report here that Δ9-THCA is a partial and selective PPARg, albeit with lower adipogenic activity than the full PPARg agonist, rosiglitazone (RGZ). In addition, Δ9-THCA enhanced osteoblastogenesis in human mesenchymal stem cells. Docking and in vitro functional assays indicated that Δ9-THCA binds and activated PPARg by acting at both the canonical and the alternative binding sites in the PPARg ligand-binding pocket. Indeed, transcriptomic signature at inguinal white adipose tissue (iWAT) from mice treated with Δ9-THCA confirmed its mode of action at PPARg. Administration of Δ9-THCA for 3-weeks in a mouse model of high fat diet (HFD)-induced obesity significantly reduced fat mass and body weight gain, and markedly ameliorated glucose intolerance and insulin resistance, while largely preventing liver steatosis, adipogenesis and macrophage infiltration in fat tissues. In addition, immunohistochemistry, transcriptomic and plasmatic biomarkers analyses showed that treatment with Δ9-THCA caused browning of iWAT and displayed potent anti-inflammatory actions in HFD mice. Altogether, our studies collectively document the potent biological activity of Δ9-THCA as a PPARggonist with capacity to substantially improve metabolic syndrome and inflammation associated to obesity. Our findings also imply that non-decarboxylated, Cannabis sativa extracts could be added to the arsenal of cannabis preparations already available in countries where medicinal cannabis is authorized.
