Project description:Pterygium is an ocular surface disease that can cause visual impairment if it progressively invades the cornea. Although many pieces of research showed that ultraviolet radiation triggers pterygium pathological progress, the underlying mechanism in pterygium remains indistinct. In this study, we used microarray to evaluate the changes of transcripts between primary pterygium and adjacent normal conjunctiva samples in China, hoping to find underlying pathways involved in pterygium progression.

Project description:Pterygium is an ocular surface disorder with high prevalence that can lead to vision impairment. As a pathological outgrowth of conjunctiva, pterygium involves neovascularization and chronic inflammation, but its pathogenesis remains largely unknown. Over the last decade, various types of disease models have been built to study pterygium. Here, we developed a 3D multicellular in vitro pterygium model using the digital light processing (DLP)-based 3D bioprinting of human conjunctival stem cells (hCjSCs). A novel feeder-free culture system was adopted and efficiently expanded the primary hCjSCs with homogeneity, stemness and differentiation potency. The DLP-based 3D bioprinting was able to fabricate hydrogel scaffolds that support the viability and biological integrity of the encapsulated hCjSCs. The bioprinted 3D pterygium model was fabricated with hCjSCs, immune cells and vascular cells to recapitulate the disease microenvironment. Transcriptomic analysis using RNA sequencing (RNA-seq) identified a distinct profile correlated to inflammation response, angiogenesis, and epithelial mesenchymal transition in the bioprinted 3D pterygium model. In addition, the pterygium signatures and disease relevance of the bioprinted model were validated with the public RNA-seq data from patient-derived pterygium tissues. By integrating the stem cell technology and 3D bioprinting, this is the first reported 3D in vitro disease model for pterygium that can be utilized by future studies towards the personalized medicine and the drug screening.

Project description:Background: Damage due to ultraviolet (UV) light from the sun is implicated in the development of two proliferative lesions of the ocular surface: pterygium and pinguecula. In this study, RNA-sequencing technology was used to investigate underlying molecular mechanisms. Methods: Pterygium and pinguecula specimens were collected at the time of surgical removal, along with adjacent specimens of healthy conjunctiva. RNA was extracted and sequenced. Data from individual samples were grouped as 1) healthy conjunctiva, 2) pterygia-highly exposed to UV (pterygia-E), 3) pterygia-normally exposed (pterygia-NE) and 4) pinguecula. Pairwise comparison was made of differentially expressed genes (DEGs). Randomly selected DEGs were validated in independent tissue specimens by RT-PCR. Gene sets were further analyzed by Ingenuity Pathway Analysis (IPA), and by assembly of cell type, tissue differentiation and pathological process markers. Results: Transcripts from 18,630 genes were identified. Pterygium exhibited 69% similarity, and pinguecula exhibited 73% similarity to healthy conjunctiva. Pinguecula and pterygia exhibited 95% similarity to each other and pterygia-E and pterygia-NE exhibited 98% similarity to each other. Among the most interesting DEGs were four tumor suppressor genes downregulated in pterygia-E and pterygia-NE: C10orf90, RARRES1, DMBT1, and SCGB3A1. C10orf90 and RARRES1 were also downregulated in pinguecula. Comparison of pterygia-E and pterygia-NE uncovered evidence of genomic instability due to failure to repair DNA damage, and associated with inflammation and the immune response; these changes were also observed in pinguecula, but to a lesser extent, and are consistent with atypia and dysplasia documented pathologically. IPA overwhelmingly linked DEGs to cancer and predicted activation of 12 different upstream regulators known to mediate epithelial cell proliferation. Cell type markers were present for all epithelial and stroma cell types quiescent stem cells of both epithelia and stroma were lacking. Cell-type and differentiation marker expression indicated well-differentiated epithelial components of mixed conjunctival, limbal, and corneal cell-types with mucosal differentiation. Upregulation of MUC6 in all three lesions suggested the presence of a glandular cell type. In all three lesions, genes characteristic of palmoplantar epidermis: KRT9 and TRPV3, were upregulated, suggesting transdifferentiation due to stress. Markers of cornification characteristic of inflammation were also upregulated. Matrix metalloproteinase expression was only slightly changed, suggesting the lesions were not invasive, however fibrovascular proliferation was evidenced. Conclusions: A hallmark of cancer, genomic instability begins in the precancerous stage, and can be caused by failure to repair DNA damage due to UV light exposure. This can be exacerbated by inflammation due to products of DNA damage. Our findings suggest that pinguecula and pterygia, while benign lesions, are both on the pathological path towards cancer.

Project description:Pterygium is a relatively common human ocular surface fibroproliferative disease that affects vision. Endogenously produced microRNA (miRNA) regulates gene expression in various ocular surface diseases and possibly pterygium. We aimed to investigate the role of miRNA in pterygium. Paired human pterygium and conjunctival tissues were obtained from patients diagnosed with primary pterygium. miRNA microarray profiling identified statistically significant miRNA changes which were matched to reciprocal significant changes in their target transcripts. We employed quantitative real-time polymerase chain reaction and found that hsa-miR-766 was up-regulated (2.57-fold) whilst hsa-miR-215 was down-regulated (0.49-fold) in pterygium compared to conjunctival control. Localization of miRNA was performed using in-situ hybridization. Transcript levels of predicted hsa-miR-766 targets, nuclear receptor subfamily 4, group A, member 1 and epidermal growth factor-containing fibulin-like extracellular matrix protein 1, were down-regulated in pterygium compared to conjunctiva by 0.53- and 0.64-fold, respectively. Collagens type 3, alpha 1 and type 4, alpha 2, both targets of hsa-miR-215, were up-regulated in pterygium by 3.01- and 3.11-fold, respectively. These changes were confirmed in the protein levels using immunofluorescent staining. Derangement of hsa-miR-766 and hsa-miR-215 may cause dysregulation of matrix rearrangement, cell proliferation and adhesion proteins, resulting in pterygium formation. Targeting miRNA may be a possible therapeutic approach in this disease. 3 pterygium samples and 3 matched conjuctiva samples from patients diagnosed with primary pterygium. A pool of all 6 samples was used as the common reference.
