Effects of Voluntary Exercise on Hippocampal Gene Expression in male C57Bl/6J mice

Project description:This experiment was designed to identify changes in gene expression in the brain (specifically the hipocampus) in relation to variable levels of voluntary exercise. Male C57Bl/6J mice were weaned onto a low fat diet (LF; Research Diets D12329, 11% kcal fat) and given continuous access to exercise wheels (30.3 x 20.6 x 26 cm; Mini Mitter®, Nalgene® Activity Wheel for Rodents) at ten weeks of age, for three days (n=96). Six mice had their wheels locked to act as sedentary controls (sed). Animals were considered to be high runners (HR) if they ran >= 5 km/night and low runners (LR) if they ran <= 2 km/night. Middle runners (MR) were chosen randomly from the population of animals falling between the high and low groups. This series is comprised of array results from three parallel experiments comparing HR versus sed, MR versus sed, and HR versus LR. Six hybridizations were performed for each of the three comparisons (3 forward-labeled, 3-reverse labeled) to account for dye-incorporation differences. One slide from the HR vs LR experiment was broken during processing, so only five are included from that group. cDNA microarray analysis of human skeletal muscle after endurance exercise

Project description:We examined global mRNA expression using cDNA microarrays in skeletal muscle of humans before, and 3h and 48h after a single bout of exhaustive endurance exercise (cycling). Keywords: Time course Healthy, non-trained university-aged subjects performed a single bout of exhaustive cycling. Skeletal muscle biopsies were taken from the vastus lateralis before, 3h and 48h after the exercise bout. Total RNA was extracted, amplified, reverse transcribed, and cDNA was analyzed on a custom made cDNA microarray. Four subjects were analyzed, and samples were not pooled between subjects (i.e. individual microarrays were used for baseline vs. 3H and baseline vs. 48h for EACH subject; repeated measures design).

Temporal pattern of skeletal muscle gene expression following endurance exercise in Alaskan sled dogs

Project description:Background: Muscle responses to exercise are complex, and potentially include acute responses to exercise-induced injury as well as longer-term adaptive training responses. Using Alaskan sled dogs as an experimental model, changes in muscle gene expression were analyzed to better understand the temporal changes that occur after severe exercise. Methods: Dogs were randomly assigned to undertake in a 160 km run (n=9), or to remain at rest (n=4). Biceps femoris muscle was obtained by needle biopsy from the unexercised dogs and two dogs at each of 2, 6 and 12 hours after the exercise and from 3 dogs 24 hours after exercise. RNA was extracted and microarray analysis used to define gene transcriptional changes. Results: Nine hundred sixty three transcripts exhibited statistically significant change over time after exercise as compared to the unexercised dogs. The changes in gene expression after exercise occurred in a clear temporal pattern and included transcripts with increased expression 2 hours after exercise with a return towards resting levels by 6 hours after exercise. Other transcripts demonstrated increased expression which peaked at six hours after exercise, while other transcripts showed sustained induction or repression over the 24 hours after exercise. Increases in a number of known transcriptional regulators, including PPAR-α, CERM and CEBPD, were observed 2-hours after exercise. Pathway analysis demonstrated coordinated changes in expression of genes with known functional relationships, including genes involved in muscle remodeling and growth, intermediary metabolism and immune regulation. Conclusion: Sustained endurance exercise by Alaskan sled dogs induces coordinated changes in gene expression with a clear temporal pattern. RNA expression profiling has the potential to identify novel regulatory mechanisms and responses to exercise stimuli.

The effects of muscular dystrophy, exercise and sActRIIB-Fc on molecular signature of skeletal muscle in mice

Project description:To study the combined effect of myostatin/activin inhibition and exercise on muscle mass and pathophysiology, young mdx mice, a model for Duchenne Muscular Dystrophy, were injected with soluble activin receptor-Fc (sActRIIB-Fc) or placebo (PBS) 1x/week for a 7-week period, in combination with or without voluntary running. C57Bl/10ScSnJ mice injected with PBS acted as wildtype controls. Microarray expression analysis from skeletal muscle was performed using m. gastrocnemies as the sample. We found thatexercise or a combination of exercise and sActRIIB-Fc treatment is more effective in correcting gene expression profiles of dystrophic muscles than the sActRIIB-Fc treatment alone. We also identified several pathways and proteins that were affected by exercise and sActRIIB-Fc together or independently. Total RNA obtained from gastrocnemius muscle of mdx mice divided into four groups: 1) control (injected with PBS, n=5), 2) runners (voluntary wheel running for 7 weeks, injected with PBS, n=5), 3) sActRIIB-Fc -treated (n=5), and 4) runners with sActRIIB-Fc-treatment (n=5). sActRIIB-Fc or PBS was injected intraperitoneally once a week with a 5-mg/kg dose of sActRIIB-Fc. In addition, wildtype mice served as healthy controls (n=4).
