Project description:Salicylic acid (SA) levels increase in Arabidopsis upon exposure to low temperature for more than a week. This cold-induced SA biosynthesis was found to proceed through the isochorismate synthase (ICS) pathway. The sid2-1 mutant is deficient in ICS1 and does not make SA at low temperature. We used microarray analysis to examine the genes differentially regulated by low temperature that were dependent on SA We used the sid2-1 mutant and a WT control at 22?C and after 3 weeks at 4?C. All samples were in triplicate.

Project description:Effector-Triggered Immunity (ETI) and Pattern-Triggered Immunity (PTI) are well-defined modes of plant immunity triggered by recognition of pathogen effector proteins and microbe-associated molecular patterns, respectively. While ETI and PTI network extensively share signaling components, the shared components are used in different ways, resulting in distinct network properties in the model plant Arabidopsis: immunity is highly robust against network perturbations in ETI but relatively sensitive in PTI. However, the molecular mechanism how the shared network leads to the different properties is not known. Here we show that sustained MAPK activation compensate salicylic acid (SA) signaling. A 12 DNA microarray study using total RNA from Arabidopsis transgenic plants carrying DEX-inducible MKK4DD in Col or sid2-2 background treated with DEX or control.

Polyamine-Induced Hormonal Changes in eds5 and sid2 Mutant Arabidopsis Plants.

Project description:Polyamines are multifaceted compounds which play a role in regulating plant growth and stress tolerance in interactions with plant hormones. The aim of the present study was to reveal how exogenous polyamines influence the synthesis of salicylic acid, with a special emphasis on the effect of salicylic acid deficiency on the polyamine metabolism and polyamine-induced changes in other plant hormone contents. Our hypothesis was that the individual polyamines induced different changes in the polyamine and salicylic acid metabolism of the wild type and salicylic acid-deficient Arabidopsis mutants, which in turn influenced other hormones. To our knowledge, such a side-by-side comparison of the influence of eds5-1 and sid2-2 mutations on polyamines has not been reported yet. To achieve our goals, wild and mutant genotypes were tested after putrescine, spermidine or spermine treatments. Polyamine and plant hormone metabolism was investigated at metabolite and gene expression levels. Individual polyamines induced different changes in the Arabidopsis plants, and the responses were also genotype-dependent. Polyamines upregulated the polyamine synthesis and catabolism, and remarkable changes in hormone synthesis were found especially after spermidine or spermine treatments. The sid2-2 mutant showed pronounced differences compared to Col-0. Interactions between plant hormones may also be responsible for the observed differences.

Comparison of osmotic stress related gene expression in Col-0, sid2-1 single, wrky54wrky70 double and wrky54wrky70sid2-1 triple mutants.

Project description:To explore the possible cause for the enhanced tolerance to osmotic stress exhibited by wrky54wrky70 double mutant, we have used Agilent Arabidopsis (V4) Gene Expression Microarray to characterize the effect of these mutations under the osmotic stress treatment. In addition to wild type and wrky54wrky70 double mutant, sid2-1 single and wrky54wrky70sid2-1 triple mutant were also included in the microarray experiment. When comparing to untreated control samples, 58 genes were identified as representatives to show the different expression level among different mutants under the 15% PEG6000 treatment for one day. Expression of six genes (RAB18, LTI78, KIN1,NCED3,P5CS1 and PRODH) from this gene list were quantified by qRT-PCR, confirming the suppressed induction in wrky54wrky70 double mutant under the osmotic stress. Osmotic stress induced gene expression in Col-WT, sid2-1 single, wrky54wrky70 double and wrky54wrky70sid2-1 triple mutants was measured after exposure to 15% PEG6000 treatment for one day. For each array, three labeled aRNA sample were hybridized and three biological replicates for each sample with dye swaps were made.
