Project description:Transcriptome at the single-cell level of a human embryoid model, which recapitulates aspects of lineage diversification and three-dimensional tissue architecture of the human embryo from the implantation to the onset of gastrulation, was profiled at different time points. Similarly, chimpanzee embryoids were generated and profiled to reveal transcriptome dynamics during the early post-implantation chimpanzee development. Our comparative transcriptome analyses reveal a critical role of NODAL signaling in human mesoderm specification, which is functionally validated by generating embryoids with NODAL-knockout (KO) hPSCs.

Single-cell RNA-sequencing of mouse embryoids at Day 8

Project description:Generating properly organized and differentiated embryonic structures in vitro from aggregates of pluripotent cells remains a major challenge to overcome. In the living embryo, the interplay between morphogens and their antagonists set up gradients of activity that instruct cells about their fate, leading to patterning and morphogenesis. Here we show that experimentally engineering a morphogen signaling center within an aggregate of mouse pluripotent stem cells is sufficient to mimic in vitro the function of an embryo organizer and to trigger embryonic development. The resulting embryoids are extensively patterned along their anterior-posterior and dorsal-ventral axes, with formation of three germ layers through a process of gastrulation and differentiation of germ layer derivatives similar to those of a neurula-stage mouse embryo.

Mechanochemical symmetry breaking during morphogenesis of lateral-line sensory organs.

Project description:Actively regulated symmetry breaking, which is ubiquitous in biological cells, underlies phenomena such as directed cellular movement and morphological polarization. Here we investigate how an organ-level polarity pattern emerges through symmetry breaking at the cellular level during the formation of a mechanosensory organ. Combining theory, genetic perturbations, and in vivo imaging, we study the development and regeneration of the fluid-motion sensors in the zebrafish's lateral line. We find that two interacting symmetry-breaking events - one mediated by biochemical signaling and the other by cellular mechanics - give rise to precise rotations of cell pairs, which produce a mirror-symmetric polarity pattern in the receptor organ.

Left-right symmetry breaking in tissue morphogenesis via cytoskeletal mechanics.

Project description:RationaleLeft-right (LR) asymmetry is ubiquitous in animal development. Cytoskeletal chirality was recently reported to specify LR asymmetry in embryogenesis, suggesting that LR asymmetry in tissue morphogenesis is coordinated by single- or multi-cell organizers. Thus, to organize LR asymmetry at multiscale levels of morphogenesis, cells with chirality must also be present in adequate numbers. However, observation of LR asymmetry is rarely reported in cultured cells.ObjectivesUsing cultured vascular mesenchymal cells, we tested whether LR asymmetry occurs at the single cell level and in self-organized multicellular structures.Methods and resultsUsing micropatterning, immunofluorescence revealed that adult vascular cells polarized rightward and accumulated stress fibers at an unbiased mechanical interface between adhesive and nonadhesive substrates. Green fluorescent protein transfection revealed that the cells each turned rightward at the interface, aligning into a coherent orientation at 20° relative to the interface axis at confluence. During the subsequent aggregation stage, time-lapse videomicroscopy showed that cells migrated along the same 20° angle into neighboring aggregates, resulting in a macroscale structure with LR asymmetry as parallel, diagonal stripes evenly spaced throughout the culture. Removal of substrate interface by shadow mask-plating, or inhibition of Rho kinase or nonmuscle myosin attenuated stress fiber accumulation and abrogated LR asymmetry of both single-cell polarity and multicellular coherence, suggesting that the interface triggers asymmetry via cytoskeletal mechanics. Examination of other cell types suggests that LR asymmetry is cell-type specific.ConclusionsOur results show that adult stem cells retain inherent LR asymmetry that elicits de novo macroscale tissue morphogenesis, indicating that mechanical induction is required for cellular LR specification.

Symmetry-Breaking Triplet Excited State Enhances Red Afterglow Enabling Ubiquitous Afterglow Readout.

Project description:Molecular vibrations are often factors that deactivate luminescence. However, if there are molecular motion elements that enhance luminescence, it may be possible to utilize molecular movement as a design guideline to enhance luminescence. Here, the authors report a large contribution of symmetry-breaking molecular motion that enhances red persistent room-temperature phosphorescence (RTP) in donor-π-donor conjugated chromophores. The deuterated form of the donor-π-donor chromophore exhibits efficient red persistent RTP with a yield of 21% and a lifetime of 1.6 s. A dynamic calculation of the phosphorescence rate constant (kp) indicates that the symmetry-breaking movement has a crucial role in selectively facilitating kp without increasing nonradiative transition from the lowest triplet excited state. Molecules exhibiting efficient red persistent RTP enable long-wavelength excitation, indicating the suitability of observing afterglow readout in a bright indoor environment with a white-light-emitting diode flashlight, greatly expanding the range of anti-counterfeiting applications that use afterglow.

Efficient generation of ETX embryoids that recapitulate the entire window of murine egg cylinder development

Project description:The murine embryonic–trophoblast–extra-embryonic endoderm (ETX) model is an integrated stem cell–based model to study early postimplantation development. It is based on the self-assembly potential of embryonic, trophoblast, and hypoblast/primitive/visceral endoderm-type stem cell lines (ESC, TSC, and XEN, respectively) to arrange into postimplantation egg cylinder–like embryoids. Here, we provide an optimized method for reliable and efficient generation of ETX embryoids that develop into late gastrulation in static culture conditions. It is based on transgenic Gata6-overproducing ESCs and modified assembly and culture conditions. Using this method, up to 43% of assembled ETX embryoids exhibited a correct spatial distribution of the three stem cell derivatives at day 4 of culture. Of those, 40% progressed into ETX embryoids that both transcriptionally and morphologically faithfully mimicked in vivo postimplantation mouse development between E5.5 and E7.5. The ETX model system offers the opportunity to study the murine postimplantation egg cylinder stages and could serve as a source of various cell lineage precursors.
