Vascular smooth muscle cell phenotype switching during cancer progression.

ABSTRACT: Smooth muscle cell plasticy and phenotypic switching play a prominent role in pathogenesis of multiple diseases. We used scRNAseq to look at the diversity of vascular smooth muscle cells in tumorigenesis.

KLF4-PFKFB3-driven glycolysis is essential for phenotypic switching of vascular smooth muscle cells

Project description:Vascular smooth muscle cells (VSMCs) within atherosclerotic lesions undergo a phenotypic switching in a KLF4-dependent manner. Glycolysis plays important roles in transdifferentiation of somatic cells, however, it is unclear whether and how KLF4 mediates the link between glycolytic switch and VSMCs phenotypic transitions. Here, we show that KLF4 upregulation accompanies VSMCs phenotypic switching in atherosclerotic lesions. KLF4 enhances the metabolic switch to glycolysis through increasing PFKFB3 expression. Inhibiting glycolysis suppresses KLF4-induced VSMCs phenotypic switching, demonstrating that glycolytic shift is required for VSMCs phenotypic switching. Mechanistically, KLF4 upregulates expression of circCTDP1 and eEF1A2, both of which cooperatively promote PFKFB3 expression. TMAO induces glycolytic shift and VSMCs phenotypic switching by upregulating KLF4. Our study indicates that KLF4 mediates the link between glycolytic switch and VSMCs phenotypic transitions, suggesting that a previously unrecognized KLF4-eEF1A2/circCTDP1-PFKFB3 axis plays crucial roles in VSMCs phenotypic switching.

Coordinated Regulation of Smooth Muscle Cell Remodeling by Non-coding and Coding products of Phenotype Switching Regulator Gene.

Project description:We identified a novel nuclear peptide Arteridin encoded by an annotated lncRNA Phenotype Swithching Regulator (PSR). To test the DNA binding capacity of Arteridin, we perfomed Arteridin ChIP-seq. We found both Arteridin and lncPSR transcript could induce vascular smooth muscle cell phenothype switching. We also found the interaction between Arteridin, lncPSR and transcription factor YBX1. To further illustrate how Arteridin and lncPSR work coordinately, we performed YBX1 ChIP-seq in VSMCs with Arteridin-FLAG overexpression or lncPSR ATT mutant overexpression.

RNA-seq identifies novel lncRNAs involved in vascular smooth muscle cell proliferation

Project description:Smooth muscle cell (SMC) phenotypic switching from a contractile to a synthetic state is implicated in diverse vascular pathologies, including neointimal formation. This study was designed to identify lncRNAs that may play a role in vascular pathologies. Primary smooth muscle cells cultured from surplus human saphenous vein tissue were treated with inflammatory and proliferative stimuli, IL1α and PDGF, for 72h and RNA extracted for RNA-sequencing. Using edgeR processed data we found expression of many lncRNAs was altered following treatment and could play a role in vascular disease.

Inhibition of smooth muscle cell TGFβ signaling in hyperlipidemic mice induces vessel wall inflammation and vessel wall dilation/dissection and leads aortic aneurysm. We performed scRNAseq method to examine smooth muscle cell gene expression profile using Apoe and SMC specific TGFbR2 KO in Apoe background mice.
