ScRNA-seq analysis for CD11c+ cells recruited to the MLNs in colitis induced in mice supplied with PBS or lysine

ABSTRACT: L-lysine (Lysine) is an indispensable amino acid that cannot be synthesized by the human body. Lysine effects on DCs in colitis is unkonwn. We investigated whether whether lysine could alter DC differentiation trajectories in colitis. We sorted CD11c+ cells recruited to the MLNs in colitis induced in mice by DSS and profiled the cells by single-cell RNA sequencing.

Project description:The goal of our study is to determine whether Atg16L1 deficiency leads to differences in the transcriptional profile of CD11c+ Dendritic Cells, ultimately leading to an increased inflammatory phenotype. CD11c+ cell sorted splenic DCs were isolated from 8 week old WT and Atg16L1 hypomorphic mice from spleens of untreated mice and were placed directly into TRIzol LS (Invitrogen). mRNA was isolated, amplified, and hybridized to an Affymetrix GeneChip (MOE430A).

Gut microbiota analysis in WT and Il17d-deficient mice treated with or without colitis

Project description:Microbiota dysbiosis has been reported to contribute to the pathogenesis of colitis, to demonstrate whether IL-17D protects against DSS-induced colitis through regulation of microflora, we performed 16S rRNA sequencing in feces from WT and Il17d-deficient mice. Our data indicate that Il17d deficiency results in microbiota dysibiosis in both steady state and DSS-induced colitis.
