RNA-seq analysis of SPL integrin b7hi or integrin b7lo PCs, or BM PCs

ABSTRACT: Newly generated plasma cells in secondary lymphoid organs migrate to niches in the bone marrow, wherein they survive as long-lived plasma cells (LLPCs). Although characterization of LLPCs has been well performed, it is still unclear what is key determinant(s) for longevity of plasma cells. One postulated model is that heterogeneity of plasma cells is formed by such as BCR affinity and/or microenvironment at the induction site, and some of these variables may instruct their longevity. Here, we found that, among newly generated IgG plasma cells, integrin b7hi marks plasma cells prone to home to the bone marrow, whereas integrin b7lo cells remain in secondary lymphoid organs.

Project description:We found that, among newly generated IgG plasma cells, integrin b7hi marks plasma cells prone to home to the bone marrow, whereas integrin b7lo cells remain in secondary lymphoid organs. Moreover, we also found that the differences in KLF2 expression between b7hi and b7lo populations in secondary lymphoid organs is likely to explain that only integrin b7hi plasma cells can egress. Next, to look for the functional target(s) downstream of KLF2 involved in egress of newly generated plasma cells, we carried out RNA-seq analysis of KLF2-sufficient and -deficient splenic plasma cells. Wildtype control splenic plasma cells were further divided into integrin b7hi and b7lo populations. We identified S1pr1 and Itgam as functional targets downstream of KLF2.

Gene expression profile at single cell level of plasma cells (PCs) from human bone marrow

Project description:Antibody-secreting plasma cells (PCs) are generated in secondary lymphoid organs, but are reported to reside in an emerging range of anatomical sites. Analysis of the transcriptome of different tissue-resident (Tr)PC populations revealed that they each have their own transcriptional signature indicative of functional adaptation to the host tissue environment. In contrast to expectation, all TrPCs were extremely long-lived, regardless of their organ of residence, with the longevity influenced by intrinsic factors like the immunoglobulin isotype. Analysis at single-cell resolution revealed that the bone marrow is unique in housing a compendium of PCs generated all over the body that retain aspects of the transcriptional program indicative of their tissue of origin.

Absence of integrin α3 modulates the integrin landscape of human keratinocytes

Project description:Integrins are among the most abundant cell surface receptors constituting the principal adhesion receptors for the extracellular matrix (ECM), providing a physical anchor for the cell and triggering multiple intracellular signalling events. Loss-of function mutations of the integrin α3 gene (ITGA3) have been recently disclosed in patients with interstitial lung disease, congenital nephrotic syndrome and junctional epidermolysis bullosa, a multiorgan disorder with fatal outcome. In these patients, the respiratory function is strongly impaired and the kidneys are variably affected, whereas skin fragility is rather mild, has a delayed onset after birth or remains unrecognized, suggesting that integrin α3 differently influences the development and homeostasis of these organs. Here we employed authentic human keratinocytes bearing a naturally occurring integrin α3 loss-of-function mutation as a prototype to characterize the molecular mechanisms launched by the constitutional absence of this integrin subunit. To validate our findings, we generated new cellular models, including an additional ILNEB patient cell line, ITGA3 rescued and knockdown cells. We show that keratinocytes lacking a functional α3 subunit have an activated cellular phenotype with a switch in the pattern of integrin α subunits on the cell surface. These assure spreading and adhesion of epidermal keratinocytes but also drive the migratory phenotype of these cells.

The Protective Role of Integrin α4β7 and Amphiregulin-Expressing Innate Lymphoid Cells in Lupus Nephritis

Project description:Type-2 innate lymphoid cells (ILC2s) have emerged as key immune-response regulators in renal-inflammatory diseases such as lupus nephritis. However, the adhesion and migration of ILC2s, including in the homeostatic and diseased kidney, are poorly understood. By integrating transcriptomic profiling, flow cytometry, live-cell imaging, and in-vivo models, we showed that renal ILCs are retained in the homeostatic kidney by the adherence of their cell-surface integrin α4β7 to VCAM-1, E-cadherin or fibronectin on structural kidney cells. When cultured on these ligands, ILC2s demonstrated remarkable migration. Knocking down integrin-α4β7 expression reduced ILC2 Areg production. In lupus nephritis, TLR7/9 may downregulate ILC2 expression of integrin-α4β7, thereby both reducing Areg expression and promoting ILC2 egress. Areg loss may promote the proinflammatory-cytokine secretion by T cells. IL-33 upregulated ILC2 integrin-α4β7 and Areg expression. Notably, IL-33 treatment enhanced survival in lupus nephritis by mitigating kidney inflammation. Thus, ILC2 adhesion may be a therapeutic target for autoimmune kidney diseases.
