Project description:Commercially available antibodies raised against condensin subunits have been widely used to characterise their cellular interactome. Here we have assessed the specificity of a polyclonal antibody (Bethyl A302-276A) that is commonly used as a probe for NCAPH2, the kleisin subunit of condensin II, in mammalian cells. We find that, in addition to its intended target, this antibody cross-reacts with one or more components of the SWI-SNF family of chromatin remodelling complexes in an NCAPH2-independent manner.

Commercially available antibodies raised against condensin subunits have been widely used to characterise their cellular interactome. Here we have assessed the specificity of a polyclonal antibody (Bethyl A302-276A) that is commonly used as a probe for NCAPH2, the kleisin subunit of condensin II, in mammalian cells. We find that, in addition to its intended target, this antibody cross-reacts with one or more components of the SWI/SNF family of chromatin remodelling complexes in an NCAPH2-independent manner. This cross-reactivity with an abundant chromatin-associated factor is likely to affect the interpretation of protein and chromatin immunoprecipitation experiments that make use of this antibody probe.

Project description:Condensin complexes are highly conserved for chromosome compaction to ensure their faithful segregation in mitosis. Condensin II is present in the nucleus throughout the cell cycle, including interphase. The aim of these experiments is to investigate the changes of gene expression in knockdown of NCAPH2, a condensin II subunit, in mouse embryonic stem cells compared to their control cells.

Project description:A novel knock-in mouse model was reported (Minella et al., Genes Devel 2008) to have a mixed hyper-proliferative and defective maturation phenotype in the erythroid series. Microarray analyses were performed to identify gene expression alterations resulting from dysregulated cyclin E control. Analysis of gene expression changes in cyclin E (T74A T393A) knock-in erythroblasts (Ter119-positive, CD71-high) isolated by FACS sorting. Comparison is to cells similarly isolated from littermate controls.

Genome-wide transcriptional analysis of the primitive erythroid (EryP) lineage differentiation

Project description:During mammalian postimplantation development, primitive erythroblasts (EryP) or red blood cells are the first cell type to be specified from nascent mesoderm, late in gastrulation. EryP arise in the yolk sac from a transient wave of progenitors, enter the circulation soon thereafter, and continue to mature in a stepwise and synchronous fashion. We have taken advantage of a transgenic mouse line in which a green fluorescent reporter is expressed specifically in the EryP lineage to identify and purify EryP at different stages of maturation from E7.5 through E12.5. A transcriptional profile that encompasses key developmental landmarks of this lineage was determined using a genome-wide microarray analysis (Illumina platform).
