Expression profiling of human umbilical cord mesenchymal stem cell line hMSCs, human esophageal carcinoma cell line EC9706 and their hybrids EMFs.

ABSTRACT: Our findings demonstrate that hMSCs can inhibit the malignant phenotypes of ECs through induced cell fusion without potentially cancerous reprogramming. To further understand the differences between before-after cell fusion and the mechanism of suppression, we used gene expression profiling technology from Capitalbio to find out the differences between EC cells, hMSCs and their derived heterokaryons.

Six dual channel Capitalbio 22K Human oligo arrays was used to hybrid six paired samples (EC9706-cy3+EMF1-cy5, EC9706-cy3+EMF2-cy5, EC9706-cy3+EMF3-cy5, EMF1-cy3+hMSCs-cy5, EMF3-cy3+hMSC-cy5, hMSC-cy3+EMF2-cy5).One replicate per array.

Project description:To identify the key microRNAs (miRNAs) of hMSCs required for fate determination, miRNA profiling was performed with hMSCs from three different sources including adipose-derived stem cells (ADSCs), bone-marrow-derived stem cells (BMSCs), and umbilical cord-derived stem cells (UCSCs) versus fibroblasts, a more differentiated mesenchymal cell types. We compared the expression profiles of two different donors per hMSCs to that of fibroblasts. All hMSCs were used for profiling at passage 3-6.

Changes in gene expression of hMSCs and NOD/scid mouse lung after IV infusion of hMSCs

Project description:Quantitative assays for human DNA and mRNA were used to examine the paradox that intravenously (IV) infused human multipotent stromal cells (hMSCs) can enhance tissue repair without significant engraftment. After 2 X 106 hMSCs were IV infused into mice, most of the cells were trapped as emboli in lung. The cells in lung disappeared with a half-life of about 24 hr but < 1,000 cells appeared in 6 other tissues. The hMSCs in lung up-regulated expression of multiple genes with a large increase in the anti-inflammatory protein TSG-6. After myocardial infarction, IV hMSCs but not hMSCs transduced with TSG-6 siRNA decreased inflammatory responses, reduced infarct size, and improved cardiac function. IV administration of recombinant TSG-6 also reduced inflammatory responses and reduced infarct size. The results suggest improvements in animal models and patients after IV infusions of MSCs are at least in part explained by activation of MSCs to secrete TSG-6.

Stabilizing heterochromatin by ZKSCAN3 safeguards hMSCs from senescence

Project description:Zinc finger protein with KRAB and SCAN domain 3 (ZKSCAN3), a transcriptional repressor, involves in multiple cellular functions. However, the functions of ZKSCAN3 in the homeostatic maintenance of human stem cells remains elusive. Here, we demonstrated that ZKSCAN3 was crucial for preventing human mesenchymal stem cells (hMSCs) from senescence in an autophagy-independent manner. Downregulation of ZKSCAN3 was observed in senescent hMSCs, and depletion of ZKSCAN3 led to premature aging in hMSCs. Further study uncovered that ZKSCAN3 maintained heterochromatin (HC) stability by interacting with heterochromatin associated proteins KAP1 and HP1 as well as nuclear envelope proteins Lamin B1, LBR and Emerin. Deficiency of ZKSCAN3 resulted in detachment of Lamina-associated domains (LADs) from the lamina, loss of heterochromatin and more accessible chromatin status within the heterochromatin and aberrant expression of repetitive sequences. Overexpression of ZKSCAN3, KAP1 or HP1 respectively rescued the senescent phenotypes in ZKSCAN3-/- hMSCs. Notably, reintroduction of ZKSCAN3 protein also retarded the cellular senescence in the replicative senescent hMSCs, as well as the pathological or physiological aging hMSCs. Together, our study reveals a novel, autophagy-independent role of ZKSCAN3 in the maintenance of heterochromatin stability and attenuation of hMSC senescence. Thus, ZKSCAN3 may be a candidate for alleviating human aging-related disorders.

Genome-wide analysis of serum starved prelamin A-accumulating hMSCs gene expression.

Project description:Analysis of serum starved prelamin A-accumulating hMSCs at gene expression level. The hypothesis tested in the present study was that prelamin A accumulation induces the dysregulation of genes that are essensial for cell survival under a stress condition such as serum starvation. The results provide important information about these genes and the functional categories that are dysregulated due to prelamin A accumulation in serum starved hMSCs. Two samples are analyzed in this microarray experiment: human mesenchymal stem cell cultured under serum starvation conditions (during 24 hours) which accumulate prelamin A (pre-hMSCs) and control mesenchymal stem cells (ctrl-hMSCs). 2 biological replicates (hMSCs derived from 2 different bone marrow donors) and 1 technical replicate are included in this analysis.

Genome-wide pairwise analysis of normal and spontaneously transformed adipose tissue derived hMSCs

Biosafety of cultivated in vitro multipotent cells prior transplantation is one of the important issues. Genome-wide pairwise expression analysis of normal and immortalized hMSCs was performed to determine aspects of spontaneous transformation and immortalization mechanisms during cultivation of hMSCs. Data has provided insight into functionally connected genes involved in spontaneous transformation of hMSCs in vitro. Multipotent stromal cells (MSCs) were isolated from adipose tissue samples obtained from 6 healthy donors. Total RNA was isolated from 6 normal and 6 donor-matched spontaneously transformed hMSC cultures in vitro for pair-wise global comparison.

Chip-chip from undifferentiated and neuron-differentiated hMSCs cells with EZH2

Project description:EZH2 plays an important role in stem cell renewal and maintenance by inducing gene silencing via its histone methyltransferase activity. Previously, we showed EZH2 downregulation markedly enhances neuron differentiation of human mesenchymal stem cells (hMSCs). To understand how EZH2 regulates neuron differentiation of hMSCs, we wanted to identify the target genes of EZH2. For this reasons we performed ChIP-on-chip experiments using specific EZH2 antibodies followed by a human promoter array for the whole human genome. The 3A6-hMSCs were differentiated into neuron for 5 days, and then 109 cells were harvested for the ChIP-on-chip assay. The procedure was based on the manufacturer's instructions (NimbleGen).

RNA sequencing analyses of fused heterokaryons between mouse embryonic stem cells and human B lymphocytes collected at early timepoints after fusion

Project description:Tcf3 is an effector of Wnt canonical pathway and deletion of Tcf3 in mouse embryonic stem cells (mESCs) strongly enhances reprogramming efficiency after fusion. We fused murine Tcf3-/- mESCs with human B-lymphocytes using the Polyethylene glycol (PEG) reagent and isolated heterokaryons at different time points to analyze early reprogramming events starting from 4 hours after fusion. We performed a genome-wide expression profile of the heterokaryons by RNA-sequencing at a high resolution.
