Transcription factors Sox8 and Sox10 contribute differentially to maintenance of the differentiated state in oligodendrocytes

ABSTRACT: To identify target genes of the transcription factors Sox8 and Sox10 in oligodendrocytes in a genome-wide and unbiased approach, we induced oligodendocyte-specific deletion of Sox8, Sox10 or a combination thereof (Sox8 knockout, Sox10 knockout, double knockout) in 2 months-old mice, dissected the corpus callosum of control and mutant mice after 21 days and isolated total RNA for ensuing RNA-Seq studies. Among others we detected substantial changes in the expression of genes associated with differentiation of oligodendrocytes and myelination in Sox10 knockout mice as well as double knockout mice but not in Sox8 knockout mice.

Transcription profiling of mice mutant for Sox8 and Sox9 has early testicular sterility

Project description:To study a possible redundant role of mouse Sox8 and Sox9 after testis induction we generated double mutants by AMH-Cre conditional inactivation of Sox9 in a Sox8-/- background. Double mutants arrest spermatogenesis in early pubescence. We performed expression profiling to explore causes. AMH-Cre/+;Sox9flox/flox;Sox8-/- is a conditional homozygous knockout of SOX9 in a sox8-/- background, using an AMH-CRE line where the CRE recombinase is expressed under the promoter region of the human AMH gene in Sertoli cells starting at E13.5. The specific cross was between males of the genotype +/+;Sox9flox/flox;Sox8-/- and females of the genotype AMH-Cre/+;Sox9flox/flox;Sox8+/-.

Sox10 Binding Analysis in the Central and Peripheral Nervous System

Project description:ChIP-seq of Sox10 in spinal cord and sciatic nerve was performed to determine shared and unique binding sites for Sox10 in oligodendrocytes and Schwann cells, respectively. Sox10 is required for both Schwann cell and oligodendrocyte development. In addition, differentiation of myelinating glia is dependent upon axonal signaling, so these studies were performed in vivo.

RNA-seq analysis of in vivo region-specific PDX (G144) tumour cell populations and in vitro SOX10 overexpression in G144 cells

Project description:For in vivo experiments GFP+ G144 human GBM cells were injected in the caudoputament of CD-1 nude mice. After tumour formation, GFP+ G144 cells were acultely purified from microdissected mouse brain regions (tumour bulk, corpus callosum, striatum) and sequenced to determine gene expression profiles of the different invasive populations. In vitro G144 cells engineered to express inducible SOX10 vector were exposed to Doxycyline for two days and sequenced to determine gene expression profiles following SOX10 overexpression.

Testis-to-ovary reprogramming and testis regression after ablation of Sox9 in Sox8-/- mice

SOX10 Single Transcription Factor Based Fast and Efficient Generation of Oligodendrocytes from Human Pluripotent Stem Cells

Project description:We report the use of a single transcription factor, SOX10, for the generation of oligodendrocytes from human pluripotent stem cells. We further used RNA sequencing to compare the oligodendrocytes derived from pluripotent stem cells to different primary cells. It was revealed that oligodendrocytes derived from pluripotent stem cells are representative of intermediate oligodendrocytes during embryonic development.

Expression Patterns of SOX10 deficient BRAFi/MEKi tolerant and SOX10 KO A375 cell lines

Project description:Given the heterogeneous expression of SOX10 in naïve melanoma, we sought to characterize SOX10 deficient population. To this end, we generated SOX10 CRISPR/Cas9 knockouts using two different guide RNAs (gRNA, #2 and #4) in the A375 (BRAF mutant) metastatic cell line. Using this approach, we identified multiple clones with loss of SOX10 expression. RNA-seq was performed to characterize the SOX10-regulated transcriptome. We used GSEA analysis to evaluate significant pathway changes in SOX10 knockout cells when compared to parental cells. We observed an enrichment in pathways associated with the tumor microenvironment (epithelial-mesenchymal transition; TGF beta signaling; extracellular structure organization; apical junction; hypoxia; angiogenesis), alterations in metabolic pathways (increase in the glycolysis pathway), a reduction in MYC and E2F targets and upregulation in p53 pathway and TNFA signaling via NFkB in the SOX10 knockout cells compared to parental cells. SOX10 negative clones have been identified in the minimal residual disease in BRAF mutant PDX models following BRAF and MEK inhibition and in patient samples while on treatment and SOX10 loss has been described as a resistant mechanism in BRAF mutant melanoma patients following vemurafenib treatment. Thus, we also analyzed the transcriptome profile of SOX10 low/deficient cells that arose following BRAFi+MEKi treatment in vivo. We performed GSEA analysis on RNA-seq data of CRT34 and CRT35 cells compared to parental A375 cells. CRT34 and CRT35 showed a very similar transcriptome profile to SOX10 KO cells when compared to A375 parental cells

Project description:We performed ChIP-Seq analysis of SOX10, histone H3 lysine 27 acetylation (H3K27ac) and H3K27 trimethylation (H3K27me3) in melanocytes to profile the genomic binding sites of SOX10 and the chromatin landscape. In parallel, we generated Sox10 haploinsufficient cell lines using gene knockout approaches and conducted microarray gene expression analysis to identify functional gene targets of SOX10 transcriptional regulation in melanocytes. We demonstrate that SOX10 predominantly engages “open” chromatin, binds to melanocyte enhancer elements and plays a central role in transcriptional activation and repression of functionally distinct classes of genes. Furthermore, we identified cis-regulatory sequence motifs of putative co-regulatory transcription factors that define SOX10-activated and SOX10-repressed target genes. Our results uncover novel mechanisms and roles of SOX10 in global transcriptional regulation of diverse regulatory pathways in the melanocyte lineage. These results indicated that SOX10 plays a role in activation and repression of distinct classes of genes. Chromatin Remodeler CHD7 mutated in CHARGE Syndrome Interacts with Sox10 to Regulate Timing of CNS Myelination and Remyelination [ChIP-seq]

Project description:Mutations in CHD7, encoding ATP-dependent chromodomain-helicase-DNA-binding protein 7, in CHARGE syndrome leads to multiple congenital anomalies including growth retardation, craniofacial malformations and neurological dysfunction. Currently, mechanisms underlying the CNS phenotypes remain poorly understood. Here, we show that Chd7 is a direct transcriptional target of oligodendrogenesis-promoting factors Olig2 and Brg1 and required for proper timing of CNS myelination and remyelination. Genome-occupancy analyses coupled with transcriptome profiling reveal that Chd7 cooperates with Sox10 to target the enhancers of key myelinogenic genes, and identify novel Chd7 target. Sox8 reprogram ectoderm into ear vesicles and auditory neurons [smartseq2]

Project description:The vertebrate inner ear arises from a pool of progenitors with the potential to give rise to all the sense organs and cranial ganglia of the head1-6. Here we explore the molecular mechanisms that control ear specification from these progenitors. Using a multi-omics approach combined with loss-of-function experiments we identify a core transcriptional circuit that imparts ear identity, along with non-coding elements that integrate this information. This analysis places the transcription factor Sox8 at the top of the ear determination network. Introducing Sox8 into cranial ectoderm not only converts non-ear cells into ear progenitors, but also activates the cellular programmes for ear morphogenesis and neurogenesis. Thus, Sox8 emerges as a master regulator of ear identity and may be a key factor for sense organ cell reprogramming.
