Multi-omics study on the effect of moderate intensity exercise on protein of lactylation in mouse muscle tissue

ABSTRACT: To explore the effects of moderate intensity exercise on protein of lactylation in mouse muscle tissue metabolism. The healthy adult mice running for 6 weeks as exercise model and sedentary mice as control were used to perform transcriptomic, proteomic, lactylation-proteomics, and metabolomics analysis. In addition, correlation analysis between transcriptome and proteome, and proteome and metabolome was conducted as well.

RNA-Seq of effects of exercise-induced Extracellular Vesicles on an F344 rat (Rattus norvegicus) model of forced running wheel exercise

Project description:n total, 16 7-week-old male F344 rats were subjected to model regular exercise and sedentary lifestyle. In the second week after arrival and acclimatization to the reversed light/dark cycle, rats were introduced to the forced running wheel without a specific running mode. It was followed by a 12-day training phase for all 16 experimental animals (started at 9 weeks of age) by gradually increasing the running speed and duration. During the training phase, 8 best performers were selected for the runner group while the remaining 8 rats were assigned to the sedentary group. The regular forced running exercise phase for the runner group lasted for 5 weeks in total, including 4 weeks of regular running. After 4 weeks of regular exercise, rats underwent brief anesthesia and of blood from the tail vein were taken. This step was repeated for the same animals immediately after 1h of running. On the same day, a blood sample was collected from the sedentary rats following the same protocol. Plasma was collected and immediately frozen for further EV isolation. EVs were isolated from rat plasma by size exclusion chromatography. RNA libraries were constructed using CleanTag® Small RNA Library Prep Kit (Trilink Biotechnologies, USA) and sequenced on Illumina NextSeq500 instrument using NextSeq 500/550 Mid Output Kit v2.5 (150 cycles).

Exercise intensity and sex alter neurometabolic, transcriptional, and functional recovery following traumatic brain injury.

Project description:In order to assess the impact of treadmill exercise on traumatic brain injury outcomes, we subjected male and female swiss webster mice to a controlled cortical impact (CCI), followed by 10 days of sedentary, low-, moderate-, or high-intensity treadmill exercise. Outcome measures included neurometabolic function, cognitive recovery, oxidative stress, pathophysiology, and single nuclei RNA sequencing (snRNA seq). The snRNA seq study was conducted on both male and female mice, and included a total of 2 replicates (each was a pool of 2 tissue samples) from each of the following groups: male sham sedentary, male CCI sedentary, male CCI low, male CCI high, female sham sedentary, female CCI sedentary, female CCI low, female CCI high. Our data reveal exercise intensity- and sex-dependent effects of treadmill exercise following injury. Transcriptomic changes were largely limited to the low-intensity exercised CCI males.

Regular physical activity followed by detraining increases the sensitivity of global gene expression to acute exercise stress

Project description:To determine the influence of a previous physcial activity period (e.g., exercise) on global gene expression, we compared differential expression in sedentary (SED) and detrained (DETR) rats relative to trained (TR) rats with regular access to physcial activity. TR and DETR rats had voluntary access to caged running wheels for 8 and 4 weeks, respectively. DETR rats were moved to sedentary conditions (normal cage activity) for another 4 weeks. SED rats experienced 8 weeks of normal caged activity; n=8 per group. At the end of each group's respective 8 weeks, all rats underwent 1 hr of moderate of acute exercise stress (e.g., forced wheel running at a speed of 20 m/min). Soleus muscles were harvested 24h post-running.

Histone lactylation antagonizes Senescent via facilitating gene-expression reprogramming [snRNA-seq]

Project description:The hallmark of aging is a remarkable rearrangement of histone modifications, which reflect alterations in the cellular environment. Here, we show that histone lactylation, a novel histone modification that bridges metabolism and epigenetics, plays a crucial role in antagonizing Senescent. The level of histone lactylation is markedly decreased during Senescent but restored following anti-Senescent treatment such as physiological hypoxic conditions and the application of nicotinamide mononucleotide, a potent precursor for NAD+. We describe the genome-wide distribution profile and gene expression network of histone lactylation during Senescent, revealing that histone lactylation prevents the Senescent program by increasing the expression of anti-aging genes and activating related signaling pathways. Moreover, running exercise enhances the level of histone lactylation and reconstructs the cell composition of mouse skeletal muscle, leading to functional improvement. Our data reveal the function of histone lactylation during Senescent and demonstrate that this modification can be used as a novel marker of Senescent, and perhaps even a potential target for aging intervention.
