Gene expression profile at a single cell level of differentiating J1 mouse embryonic stem cells cocultured with OP9 cells

ABSTRACT: The OP9 coculture system is widely used to differentiate pluripotent stem cells into mesodermal lineages. We used single cell RNA sequencing to analyze differentiation trajectories and heterogeniety of differentiating pluripotent stem cells using the OP9 coculture system.

RNA-seq analysis of mouse pluripotent stem cells derived cells

Project description:To detect the role of OP9 stromal cells in our optimized 3D self-assembling peptide induction system followed by the OP9 coculture system. First, we used RNA-seq to analyze mouse pluripotent stem cells derived total cells at day5 between the 3D+OP9 and 3D+0.1% group in our hematopoietic differentiation system. In addition, to further evaluate hematopoietic transcriptome differences between Lin-Sca-1+c-kit+CD201+ cells and Lin-Sca-1+c-kit+CD201- cells, we used RNA-seq to analyze mouse pluripotent stem cells derived Lin-Sca-1+c-kit+CD201+ and Lin-Sca-1+c-kit+CD201- cells at day5 in our 3D+OP9 hematopoietic differentiation system.Meanwhile,we used the mouse embryonic stem cell(mESC) as negative control and bone marrow derived Lin-Sca-1+c-kit+CD201+ and fetal liver derived Lin-Sca-1+c-kit+CD201+ as positive control.

Gene expression profile of AU019823-gene-trapped early mesodermal cells and their revertant cells

Project description:Using our previously established homozygous mutant mouse ESC bank, we identified a previously uncharacterized novel gene AU019823, which we named “attenuated hematopoietic development (Ahed)”, whose null mutant ESCs exhibited attenuated production (70-fold fewer) of blood cells during the hematopoietic differentiation supported by coculture with OP9 stromal cells. Both mutant and revertant (almost identical to wild-type) ESCs normally differentiate into Flk-1-positive early mesodermal cells in vitro. In this study, to explore the function of this gene, we analyzed the gene expression profile of Flk-1-positive cells with these two genotypes.

Project description:Umbilical cord blood (UCB) transplantation shows pro-angiogenic effect and contributes to symptom amelioration in animal models of cerebral infarction. OP9 is a stromal cell line used as feeder cells to promote hematoendothelial differentiation of embryonic stem cells. We co-cultured UCB 24hr with OP9 (i.e. OP9 preconditioning) and investigated its change in angiogenic properties and underlying mechanisms. Single cell RNA sequencing showed prominent phenotypic shift toward M2 in monocytic fraction of OP9 pre-conditioned UCB.

Expression data from Hh signaling-deficient OP9 stromal cells

Project description:In the bone marrow B cell development occurs in initimate and essential association with stromal cells. Known effects of stromal cells on B cell development have been shown to be mediated through direct contact and by soluble factors produced by stromal cells. Our findings suggest that cell-nonautonomous Hh signaling may play an important role in B cell lymphopoiesis. We therefore interrogated OP9 stromal cells for Hh-dependent transcripts that may confer B lymphopoietic identity. We generated OP9 stromal cells deficient in Hh signaling through targeting the non-redundant, pathway-obligate GPCR Smoothened. OP9 cells were transduced with RNAi pLKO.1-Puro lentiviral particles expressing a scrambled control shRNA (NT) or shRNA targeting Smoothened (Smo-kd). RNA from stable NT and Smo-kd OP9 cells were isolated in triplicate and global changes in transcripts were analyzed using the Affymetrix Mouse Exon 1.0 ST gene chip.

A mesoderm-derived mesenchymal stem/stromal cells (MSC) precursor: time course experiment

Project description:Mesenchymal stem/stromal cells (MSCs) are multipotent cells that can differentiate into a variety of cell types forming connective tissue and skeleton, and are essential participants in the development of all organs. However, MSC precursors remain largely unknown. In human embryonic stem cells (hESCs) directed to mesendodermal differentiation through coculture with OP9 stromal cells, we identified a population of mesodermal cells by surface expression of apelin receptor (APLNR1). APLNR+ cells were enriched with precursors generating compact spheroid colonies in semisolid suspension culture. Being formed by single cells, these colonies consisted of a uniform population of mesenchymal cells with a transcriptional profile representative of embryonic mesenchyme originating from lateral plate/extraembryonic mesoderm. Mesenchymal colony formation required serum-free medium and FGF2 as a colony-forming factor, could be significantly enhanced by PDGF-BB, but suppressed by VEGF. When transferred to the adherent cultures in serum-free medium with FGF2, individual colonies gave rise to multipotential mesenchymal cell lines with typical phenotype (CD146+CD105+CD73+CD31-CD43-CD45-), differentiation (chondro-, osteo-, and adipogenesis) and proliferation (>80 doublings) potentials. Consistent with lineage-restricted differentiation pattern, neither endothelial nor hematopoietic cells could be produced from adherent mesenchymal cultures, however endothelial cells could be derived from mesenchymal colonies in the early days of colony-forming culture suggesting that mesenchymal cells arose from cells with primary angiogenic potential (mesangioblasts). Together these studies identified mesangioblasts as the earliest clonogenic mesenchymal precursors at this stage of their specification from mesoderm. This set (8 samples) of expression data is a time-course experiment of hESC (H1) differentiated in OP9 coculture for 1-7 days.

Project description:The differentiation of human pluripotent stem cells (hPSC) towards clinically relevant cells for regenerative medicine has been hampered by poor differentiation efficiencies. This may be a result, in part, of the heterogeneity that exists within stem cell cultures. We have generated a new stem cell medium PRIMO, which biases cells towards mesodermal differentiation without losing pluripotency. This experiment characterizes the gene expression of cells grown in PRIMO and compares this to cells grown in standard culture conditions and a 72 hour time course of mesodermal differentiation
