Effects of eyestalk ablation on the gene expression levels in female black tiger shrimp broodstock (Penaeus monodon)

ABSTRACT: Comparisons of gene expression profiles between ovaries of before (day 0) and after eyestalk ablation (days 1, 4 and 7) of domesticated 14-month-old black tiger shrimp (Penaeus monodon) were made using a cDNA microarray. Differentially expressed genes were identified through the microarray analysis, and the microarray results were confirmed by real-time PCR. Selected genes were further characterized.

Project description:Comparisons of gene expression profiles between ovaries of before (day 0) and after eyestalk ablation (days 1, 4 and 7) of domesticated 14-month-old black tiger shrimp (Penaeus monodon) were made using a cDNA microarray. Differentially expressed genes were identified through the microarray analysis, and the microarray results were confirmed by real-time PCR. Selected genes were further characterized. A cDNA microarray consisting of 5,568 features was constructed from EST libraries of P. monodon. RNA samples were extracted from the ovaries of before (day 0) and after eyestalk ablation at days 1, 4 and 7 from shrimp from the Shrimp Genetic Improvement Center, Thailand. The RNA was converted into cDNA by indirect aminoallyl-cDNA labelling method (LabelStar Array, Qiagen). aa-cDNA from day 0 samples were coupled with Cy3 dye as a reference, and those from days 1, 4 and 7 were coupled with Cy5 dye.

Thus, the experiment was to compare transcriptomic profiles of two different parts of reproductive organs, namely testes (TT) and vas deferens (VD), of domesticated 17-month-old between two different feeds, namely commercial pellet and polychaetes after feeding for one month. Differentially expressed genes were identified through the microarray analysis, and the microarray results were confirmed by real-time PCR. Selected genes were further characterized.

Differentially expressed genes were identified through the microarray analysis, and the microarray results were confirmed by real-time PCR. Selected genes were further characterized. A cDNA microarray was constructed from the EST libraries of P. monodon, consisting of 5,568 features. RNA samples were extracted from testes of pooled juveniles (4 month-old, pooled from n=114), broodstocks from West and East sides of Thailand, and domesticated broodstocks of 10-, 14-, and 18-month old. The RNA from testis of pooled juveniles was labeled with Cy3 dye as a reference and those of the broodstocks were labeled with Cy5 dye.

Stress induced gene expression profiling in Penaeus monodon

Project description:Cultured shrimp are continuously exposed to variable environmental conditions which are associated with stress and subsequent outbreaks of disease. To investigate the effect of environmental stress on Penaeus monodon gene expression, a 3853 random cDNA microarray chip was generated with clones originating from 6 stress-enriched hemocyte libraries generated by suppression subtractive hybridization and a normal hemocyte cDNA library. Changes in temporal gene expression were analyzed from shrimp exposed to hypoxic, hyperthermic and hypoosmotic conditions. 3.1% of the cDNAs were differentially expressed in response to at least one of the environmental stressors. 70% of the differentially expressed clones had no significant sequence similarity to previously known genes. Among those genes with high identity to known sequences, the most common functional groups were immune related genes and non-LTR retrotransposons. Hierarchical clustering revealed a set of cDNAs with temporal and stressspecific gene expression profiles as well as a set of cDNAs indicating a common stress response between stressors. Hypoxic and hyperthermic stressors induced the most severe short-term response in terms of gene regulation, and the osmotic stress had the least variation in expression profiles relative to the control. These expression data agree with observed differences in shrimp physical appearance and behavior following exposure to stress conditions. Keywords: stress response, shrimp, time series
