ABSTRACT: We analyzed ORC2 ChIP-Seq from hand dissected salivary glands of wandering third instar larvae from OrR or SuUR Drosophila. Goals were to ascertain the difference in binding profile between salivary glands expressing and not expressing the Supressor of UnderReplication protein.

Project description:ChIP was performed to identify regions of gDNA bound by orc2 in third instar salivary glands of Drosophila WT and SuUR mutants. This demonstrated that ORC does not localize to regions that are under-replicated in SuUR mutant third instar salivary glands.

Goals were to determine the DNA damage profile relative to underreplicated domains. We analyzed SUUR and Cdc45 ChIP-Seq from hand dissected early (stage 10) and late (stage 12/13) egg chambers from adult Drosophila ovaries. The goals was to determine the localization of SUUR relative to replication forks.

Project description:We analyzed Origin Recognition Complex Subunit 2 (ORC2) ChIP-seq from hand-dissected fat body tissue from 68hr (after egg laying, AEL), 92hr AEL, and late-third wandering Drosophila melanogaster larvae. Fat body was dissected from wild-type (OrR) males and testes were removed. We examined ORC2 binding genome-wide with particular focus on the underreplicated regions in the fat body.
