Project description:4 Adult male Sprague-Dawley rats (275-350 g) were anesthetized and subjected to hepatectomy sham surgery (abdominal cavity was opened, liver was handled, but no tissue resection was made). 1 hour after the surgery rats were killed and liver samples were harvested. This study was conducted to analyzes the effects of surgical stress on gene expression levels in rat liver. It provides additional data to 1-6 h partial hepatectomy study (Series GSE7415). Project description:We report the expression profiles of ileal samples extracted from obese male C57Bl6 mice following vertical sleeve gastrectomy or sham surgery, focusing of gene signatures indicative of altered bile acid metabolism. We induced obesity in male C57bl6 mice through a high-fat diet. At 8 weeks, they were submitted to either a vertical sleeve gastrectomy (VSG) or a sham surgery; following surgery VSG mice were fed ad libitum while sham controls were pair-fed (SPF) to the experimental animals. During the four perioperative days, mice were fed a liquid Osmolite diet. Approximately 8 weeks after surgery mice were fasted overnight and gavaged with .5mL Osmolite; after one hour mice were sacrificed and the terminal ileum were extracted. RNA was extracted, quantified, and compared between VSG and SPF experimental groups.

The Epigenome of Spermatozoa is Affected in Obesity and Dynamically Changes after Bariatric Surgery [Lean/Obese MNase]

Project description:Obesity is a heritable disorder, with children of obese fathers at higher risk of developing obesity. Â Environmental factors epigenetically influence somatic tissues, but the contribution of these factors to the establishment of epigenetic patterns in human gametes is unknown. Here, we hypothesized that weight loss remodels the epigenetic signature of spermatozoa in human obesity. Comprehensive profiling of the epigenome of sperm from lean and obese men showed similar histone positioning, but small non-coding RNA expression and DNA methylation patterns were markedly different. In a separate cohort of morbidly obese men, surgery-induced weight loss was associated with a dramatic remodeling of sperm DNA methylation, notably at genetic locations implicated in the central control of appetite. Our data provide evidence that the epigenome of human spermatozoa dynamically changes under environmental pressure, and offers Â insight into how obesity may propagate metabolic dysfunction to the next generation. Examination of the DNA methylation status, histone retention and sncRNA expression of the semen of 13 lean and 10 obese individuals; as well as the DNA methylation status of the semen of 6 obese men undergoing Roux-en-Y GBP surgery, at three time points: approximately 1 week before, 1 week after and 1 year after the surgery.

Gene Expression of brain cortex microvessels in two rat models of acute liver failure (ALF)

Project description:Evaluation of early transcriptional changes in blood brain barrier genes involved in the patogenesis of brain edema associated with ALF. Two different well stablished rat models were analyzed: Portacaval anastomosis plus hepatic artery ligation and portacaval anastomosis plus total hepatectomy. ALF was induced by portocaval anastomosis (PCA) and 24 hours after the surgery animals underwent hepatic artery ligation (HAL) or total hepatectomy (HEP). Sham rats were subjected to a fictitious two-step surgery. Between 8 and 12 hours after the last surgery rats were euthanized by decapitation, to avoid anesthetic effects, and cortical microvessels isolated.
