Transcriptome study in rhesus monkey

ABSTRACT: RNA-Seq technology was widely used to study gene expression, splicing structure and RNA editing. To facilitate transcriptome study in rhesus macaque, and provide an evolutionary context to understand human biology, we performed 90bp × 2, paired-end, strand-specific, polyA-postive RNA-Seq in 10 tissues of the same macaque animals. The data were deposited under accession number GSE42857 and GSE34426.

Trancriptome study in rhesus monkey

Project description:While the next generation sequencing technology is accelerating the discovery of sites in RNA editing, the strategies to accurately identify the editome, the mechanism by which its profile is maintained and its functional significance remain controversial. Here, 90bp × 2, paired-end, strand-specific, polyA-postive RNA-Seq were performed in 3 rhesus monkey tissues, and 90bp × 2, paired-end, whole exome sequencing was performed in blood cells. Combining genome-wide identification and other quality control in multiple tissues from the same individual, we identified a list of editing sites in coding regions from the rhesus macaque, one of our closest evolutionary relatives. Low-scale verification validated all of these sites and the corresponding levels of editing. The editome in macaque coding region suggests RNA editing as a type of controlled, conserved regulation shaped by purifying selection. This submission represents RNA-Seq: cerebellum, lung, kidney and heart component of study.

Project description:The primary goal of this study was to compare the performances of Rhesus Macaque Genome Array and Human Genome U133 Plus 2.0 Array with respect to the detection of differential expressions when rhesus macaque RNA extracts were labeled and hybridized. The secondary goal of this study was to investigate the effect of mismatch position on signal strength in Affymetrix GeneChips by examining naturally occurring mismatches between rhesus macaque transcripts and human probes from Human Genome U133 Plus 2.0 Array. Transcriptional profiling of rhesus monkey nuclear transfer embryonic stem cells

Project description:Derivation of embryonic stem cells (ESC) genetically identical to a patient by somatic cell nuclear transfer (SCNT) holds the potential to cure or alleviate the symptoms of many degenerative diseases while circumventing any immunorejection issues. However, no primate nuclear transfer embryonic stem (ntES) cell lines have been derived to date. Here, we used a modified SCNT technique to produce rhesus macaque SCNT blastocysts at a relatively high efficiency from adult donor cells and we successfully derived two primate ntES cell lines from 304 oocytes (an overall efficiency of 0.7%). Nuclear and mitochondrial DNA analysis confirmed the ntES cell lines were derived from rhesus monkey SCNT blastocysts and both rhesus monkey ntES cell lines exhibited a normal ESC morphology, expressed key stemness markers, were transcriptionally indistinguishable from control ESC lines and differentiated into multiple cell types. This is, to our knowledge, the first confirmed derivation of primate ntES cell lines. Keywords: rhesus monkey somatic cell nuclear transfer embryonic stem cells

Comparison of proteome alternations during aging in temporal lobe from human and rhesus macaque

Project description:Aging of population is a great challenge of healthcare. In china, the number of the elderly is rapidly growing, and it was estimated that there will be approximately 400 million citizens above 65 years old in 2050.Study on the changes of brain during aging may help elucidate the mechanism of the pathological process, and hence prevent or treat these neurological diseases.Rhesus macaque (Macaca mulatta) and human have a genetic homology of 95%, and their anatomy structures or physiological process are highly similar, which make rhesus macaque one of the most important nonhuman primate models.Thus, the comparison between the change of protein profile during aging in human and rhesus macaque is still necessary, and the characteristics of proteins that are conservative or divergent are of interest.The aim of the(our) study is to identify the conservative changes of pathways during aging, and to reveal the potential difference between human and rhesus macaque so that relevant studies based on primate models can be interpreted more accurately.
