Project description:Alterations in the presence of sperm RNAs have been identified using microarrays in teratozoospermic (abnormal morphology) or other types of infertile patients. However, so far no studies had been reported on the sperm RNA content using microarrays in asthenozoospermic patients (low motility). We started the present project to with the goal to characterize the RNA expression in asthenozoospermic infertile patients as compared to normozoospermic fertile controls. We selected four normal fertile donors and four severe asthenozoospermic infertile patients. Equal amounts of RNA were extracted from the sperm samples, subjected to different quality controls and hybridized to the Affymetrix U133 Plus version 2 arrays.

Some of genes were up-regulated or down-regulated in asthenozoospermia, and their abnormal expression were the causes of the impaired sperm motility. Results provide insight into the mechanisms by which asthenozoospermia is controlled. We used microarrays to detail the global programme of gene expression of ejaculated spermatozoa between normozoospermic and asthenozoospermic men and identified distinct classes of genes expressed differentially in two groups. The liquefied semen samples from normozoospermic and asthenozoospermic men were purified by Percoll on a discontinuous density gradients for RNA extraction and hybridization on Affymetrix microarrays. 30 sperm samples of each group were pooled to abtain enough total RNA.

Project description:Male factor infertility is increasing and recognized as playing a key role in reproductive health and disease. The current primary diagnostic approach is to assess sperm quality associated with reduced sperm number and motility, which has been historically of limited success in separating fertile from infertile males. The current study was designed to develop a molecular analysis to identify male infertility using genome wide alterations in sperm DNA methylation. A signature of differential DNA methylation regions (DMRs) was identified to be associated with male infertility patients. A promising therapeutic treatment of male infertility is the use of follicle stimulating hormone (FSH) analogs which improved sperm numbers and motility in a sub-population of infertility patients. The current study also identified genome-wide DMRs that were associated with the patients that were responsive to FSH therapy versus those that were non-responsive.

Project description:Normal human spermatogenesis concludes with the formation of large numbers of morphologically well developed spermatozoa. While transcriptionally quiescent these cells carry an RNA payload that reflects the final spermiogenic phase of transcription. We report here the spermatozoal transcript profiles characteristic of normally fertile individuals and infertile males suffering from a consistent and severe teratozoospermia in which under 4% of spermatozoa are morphologically normal. RNA was extracted from the purified sperm cells of ejaculate and hybridized to Affymetrix U133 (v2) Microarrays. Spermatozoal RNAs were prepared from the semen samples of 21 individuals. An asymmetric dual block design was adopted with biological replicates in both blocks. 13 semen samples were assessed from normally fertile males who had fathered at least one child. 8 semen samples were assessed from infertile individuals with a severe and consistent heterogeneous teratozoospermia who showed no other abnormal semen parameters.

Project description:Growing evidences have shown that many transcripts have been discovered in sperm and small noncoding RNAs, such as miRNA and tsRNA, of sperm paly an important role in the transgenerational transmission of paternal lifetime experiences. However, the expression profile of long noncoding RNAs (lncRNAs) in mammalian sperm has not been systematically investigated. Furthermore, Abnormal expression of lncRNA whether or not related with the asthenozoospermic is completely unknown. In the present study, highly purified RNA was served to investigate lncRNA expression profiles in human mature sperm by stranded-specific RNA-seq. We identified 38,307 known and 27,472 novel lncRNA genes, and the existence of intact LncRNA transcripts was confirmed by RT-PCR and fluorescence in situ hybridization (FISH) on two representative LncRNAs. Many lncRNAs exclusive were expressed in testis and sperm. 880 lncRNA sequences were conserved between human and mouse with > 80% overlap. Compared with normal population, the lncRNA profiles in asthenozoospermic patients’ sperm showed significant difference with 2437 up-regulation and 306 down-regulation of lncRNA. This study will provide a preliminary database valuable for identifying lncRNAs critical on sperm function regulation or important for screening the lncRNA markers of asthenozoospermia.

Project description:The aim of this study was to achieve a higher resolution of the human sperm proteome for better understanding of the molecular causes of male infertility. For this purpose, we examined the proteomic profiles of sperm from 76 men for proteins with deregulated abundances. Infertile men had abnormal semen parameters and were involuntarily childless. Men in the cohort with normal fertility had normozoospermia and had fathered children without medical assistance. Mass spectrometry analysis led to an in-depth reconstruction of the human sperm proteome. The corresponding genes were mainly involved in cellular motility, response to stimuli, adhesion, and reproduction. We also observed increasing numbers of at least three-fold differentially abundant sperm proteins from oligozoospermia and oligoasthenozoospermia to oligoasthenoteratozoospermia, compared with normozoospermia. Primarily, flagellar assembly and sperm motility, sperm-egg interaction, and male gametogenesis were affected. This suggests that sperm of infertile men carry signatures of impaired sperm production and are functionally impaired. Not least, we present new male infertility markers in addition to established ones.

Project description:Although piwi-interacting RNAs (piRNAs) play pivotal roles in spermatogenesis, little is known about piRNAs in the seminal plasma of infertile males. In this study, we systematically investigated the profiles of seminal plasma piRNAs in infertile males to identify piRNAs that are altered during infertility and evaluate their diagnostic value. Seminal plasma samples were obtained from 211 infertile patients (asthenozoospermia and azoospermia) and 91 fertile controls. High-throughput sequencing technology was employed to screen piRNA profiles in seminal plasma samples pooled from healthy controls and infertile patients. The results identified 61 markedly altered piRNAs in the infertile patient groups compared with the control group. Next, a quantitative RT-PCR assay was conducted in the training and validation sets to measure and confirm the concentrations of altered piRNAs. The results identified a panel of 5 piRNAs that were significantly decreased in the seminal plasma of infertile patients compared with healthy controls. The areas under the ROC curves for these piRNAs ranged from 0.796 to 0.996, suggesting that the diagnostic potential of these 5 piRNAs to distinguish asthenozoospermic and azoospermic individuals from healthy controls was high. In summary, this study identifies a panel of piRNAs that can accurately distinguish fertile from infertile males. This finding may provide pathophysiological clues that are involved in the development of infertility. Project description:Aberrant DNA methylation profiles have been associated with male infertility and some semen parameters. However, few studies systematically surveyed DNA methylation profiles associated with sperm motility in normozoospermia and asthenozoospermia. In this study, based on promoter targeted bisulfite sequencing technology, we provided a quantitative description on global DNA methylation profiles. The average global methylation values were 24.7% and the inter-individual variance was about 14.4%, while the intra-individual variance was about 3.9%. The difference between different motile sperm population or different participant groups was subtle and not significant. Furthermore, we identified 134 differentially methylated CpGs and 134 differentially variable CpGs in low motile sperm from asthenozoospermic patient (P < 0.05). Based on the literature, we further found 16 differentially methylated or variable genes which were required for spermatogenesis or sperm motility or dominantly expressed in testis. This study will provide potential markers for clinical diagnosis and a promising basis for understanding the effect of DNA methylation on asthenozoospermia.
