Identification of genes characteristic of primary inguinal or epididymal preadipocyte fibroblasts

ABSTRACT: Committed preadipocyte fibroblasts were genetically labelled in transgenic mice by expressing GFP under the control of the locus for Zfp423, a gene controlling preadipocyte determination. These mice are herein referred to as Zfp423-GFP mice. The overall goal was to identify genes differentially expressed between adipogenic GFP+ firboblasts and non-adipogenic GFP- fibroblasts from either inguinal or epididymal fat stromal vascular cultures obtained from Zfp423-GFP mice.

Project description:Committed preadipocyte fibroblasts were genetically labelled in transgenic mice by expressing GFP under the control of the locus for Zfp423, a gene controlling preadipocyte determination. These mice are herein referred to as Zfp423-GFP mice. The overall goal was to identify genes differentially expressed between adipogenic GFP+ firboblasts and non-adipogenic GFP- fibroblasts from either inguinal or epididymal fat stromal vascular cultures obtained from Zfp423-GFP mice. Adipose stromal vascular cultures were obtained from inguinal or epididymal fat of 6-8 week-old male Zfp423-GFP mice. GFP+ and GFP- fibroblasts were sorted from fat stromal vascular cultures and then returned to culture. Subconfluent cultures of GFP+ and GFP- cells were compared in triplicate for Affymetrix analysis.

Project description:In order to identify the molecular mechanisms controlling preadipocyte commitment we derived new sublines of Swiss 3T3 fibroblasts with varying potential for adipogenesis. Swiss 2, Swiss 8, Swiss 9, and Swiss 19 differentiate into fat cells while Swiss 3, Swiss 5, and Swiss 22 have a lower propensity to differentiate into fat cells. Identification of Zfp423-dependent genes in adult inguinal white adipocytes

Project description:We derived a model that allows for doxycycline-inducible deletion of Zfp423 in mature adipocytes of adult mice (Adiponectin-rtTA; TRE-CRE; Zfp423 loxP/loxP). In these animals deletion of Zfp423 results in a spontaneous conversion of white adipocytes into beige-like adipocytes at room temperature. The goal of this expression analysis was to 1) determine the gene programs dependent on adipocyte Zfp423 in inguinal WAT, and 2) determine the similarity between the converted beige-like cells to normal beige adipose tissue that accumulates upon cold exppsure.

RNAi profiling of mouse 3T3-L1 adipocytes knocked down for PPAR gamma returned the expression of most adipocyte genes towards preadipocyte levels

Project description:The nuclear receptor PPAR gamma is required for adipocyte differentiation, but its role in mature adipocytes is less clear. Here we report that knockdown of PPAR gamma expression in 3T3-L1 adipocytes returned the expression of most adipocyte genes towards preadipocyte levels. Consistently, down regulated but not up regulated genes showed strong enrichment of PPAR gamma binding. Surprisingly, not all adipocyte genes were reversed and the adipocyte morphology was maintained for an extended period after PPAR gamma depletion. To explain this, we focused on transcriptional regulators whose adipogenic regulation was not reversed upon PPAR gamma depletion. We identified GATA2, a transcription factor whose down-regulation early in adipogenesis is required for preadipocyte differentiation, remaining low after PPAR gamma knockdown. Forced expression of GATA2 in mature adipocytes complemented PPAR gamma depletion and impaired adipocyte functionality with a more preadipocyte- like gene expression profile. Ectopic expression of GATA2 in adipose tissue in vivo had similar effect on adipogenic gene expression. These results suggest that PPAR gamma-independent down regulation of GATA2 prevents reversion of mature adipocytes after PPAR gamma depletion. Experiment Overall Design: This dataset consists of three sample groups: preadipocytes, control siRNA treated adipocytes, and PPAR gamma siRNA treated adipocytes. Each sample group consists of three replicates samples. Each sample was hybridized to a separate array for a total of nine arrays. RNA sequencing Facilitates Quantitative Analysis of luteolin or quercetin on adipogenesis Transcriptomes

Project description:We report the use of transcriptome sequencing in the intervention of luteolin or quercetin in adipogenesis. We constructed the adipogenic differentiation model of mouse embryonic fibroblasts and obtained the differential genes of luteolin or quercetin regulating adipocyte differentiation by transcriptomic sequencing. We identified 2085 differentially expressed lipogenesis response genes, 181 luteolin response genes and 574 quercetin response genes. This study provides a framework for the network regulation mechanism of luteolin or quercetin inhibiting preadipocyte differentiation.

Perirenal and epididymal preadipocytes from young and old rats.

Project description:Inherent depot- and age-dependent preadipocyte characteristics may contribute to age-related fat redistribution. Both aging and depot origin affect preadipocyte replication and adipogenesis. To define responsible mechanisms, we analyzed genome-wide expression profiles in epididymal (E) and perirenal (P) preadipocytes cultured from young (3 month) and old (30m) rats.
