Project description:Inflorescence architecture of Arabidopsis thaliana is regulated by ER-EPFL4/6 signaling module. To analyze the genes governed by this module, the transcriptional profiles of er-2 (allelic to er-106) mutant and epfl4 epfl6 double mutant were investigated. 9 samples are included in this study. Three biological replicates of 33 d post germination inflorescence tips of er-2 (allelic to er-106, Lease et al., 2001) and epfl4 epfl6 were compared with wild type (Col-0) sample for changes in gene expression.

Regulation of inflorescence architecture by intertissue layer ligand-receptor communication between endodermis and phloem.

Project description:Multicellular organisms achieve final body shape and size by coordinating cell proliferation, expansion, and differentiation. Loss of function in the Arabidopsis ERECTA (ER) receptor-kinase gene confers characteristic compact inflorescence architecture, but its underlying signaling pathways remain unknown. Here we report that the expression of ER in the phloem is sufficient to rescue compact er inflorescences. We further identified two Epidermal Patterning Factor-like (EPFL) secreted peptide genes, EPFL4 and EPFL6/CHALLAH (CHAL), as redundant, upstream components of ER-mediated inflorescence growth. The expression of EPFL4 or EPFL6 in the endodermis, a layer adjacent to phloem, is sufficient to rescue the er-like inflorescence of epfl4 epfl6 plants. EPFL4 and EPFL6 physically associate with ER in planta. Finally, transcriptome analysis of er and epfl4 epfl6 revealed a potential downstream component as well as a role for plant hormones in EPFL4/6- and ER-mediated inflorescence growth. Our results suggest that intercell layer communication between the endodermis and phloem mediated by peptide ligands and a receptor kinase coordinates proper inflorescence architecture in Arabidopsis.

Comparison of infloresence transcriptome of bp er vs. bp er fil

Project description:BP and ER encode proteins that act synergistically to regulate Arabidopsis inflorescence architecture. To search for genes/proteins that influence the BP/ER signaling pathways, we conducted mutagenesis of the bp er double mutant and found that a mutation in FILAMENTOUS FLOWER (FIL) suppresses many of the morphological/developmental defects in bp er. Given that FIL encodes a Zn-finger containing transcription factor, microarray analysis was conducted on bp er vs. the bp er fil line to identify genes that are misregulated and which might implicate specific genes/proteins/pathways that are involved in regulating inflorescence development.

HB21 induction in Arabidopsis inflorescence apexes

Project description:HB21 is a homeodomain leucine zipper transcription factor involved in the establishment of bud axillary dormancy. In this work we have characterized the role of HB21 in the control of the inflorescence arrest at the end of flowering. HB21, together with HB40 and HB53, are upregulated in the inflorescence apex at the end of the flowering phase promoting the floral bud arrest associated with the process. We also show that ABA accumulation occurs in the inflorescence apex at the end of flowering in physiological conditions, and that this accumulation depends on HB genes. Our work has allowed us to propose a physiological role of ABA in the control of floral bud arrest at the end of flowering

miR396-OsGRF6 module controls rice inflorescence architecture

Project description:Developing strategies to increase rice productivity to meet global demand is one of the main challenges for breeders around the world. Here, we report a novel microRNA mediated process that increases rice grain yield in field trials. Expression of target mimicry of microRNA396 (MIM396) significantly increases rice yield by modulating the development of the auxiliary branches and spikelets through promoting the expression of Growth Regulation Factor 6 (OsGRF6). OsGRF6 coordinately induces the expression of several key factors involved in branch and spikelet development, auxin (IAA) biosynthesis, and auxin signaling pathway. Our results demonstrate that the miR396b-GRF6 module acts as a key player in shaping the inflorescence architecture of rice, which could be engineered to generate high-yield rice. This dataset records the profile of the binding peaks of OsGRF6 with GFP antibody in 35S:GRF6-GFP overexpression lines and the differential expressed genes between MIM396 and WT plants.
