Project description:To explore the molecular basis of validamycin overproduction at the transcriptional level, the transcriptomes of strain 5008 and TL01 cultivated in Yeast extract-Malt extract-Glucose (YMG) and rice-peanut cake based industrial (IND) fermentation medium were compared by microarray analysis. Global gene expressions in the strain 5008 and TL01 were measured in fermentation medium YMG and IND, respectively. Three independent experiments were performed at each condition.

Regulation by growth temperature of Pseudomonas aeruginosa quorum-sensing depedent virulence factors production involves two RNA-thermometers.

Project description:Several bacterial human pathogens regulate the production of virulence factors by temperature, expressing them only at 37 °C. Accordingly we show that the production of all P. aeruginosa virulence factors that are dependent on the QS transcriptional regulator RhlR, but only a fraction that are activated by LasR, are induced at 37 °C compared to 30 °C or 25 °C. The RhlR-dependent induction at 37 °C is a posttranscriptional effect due to an RNA thermometer of the ROSE family that thermoregulates the expression of rhlAB operon involved in rhamnolipids production, a virulence associated trait. This RNA structure also affects the expression of the downstream rhlR gene. A second thermometer is present upstream lasI and causes a reduced expression of this gene at lower temperatures without causing a significant decrease of the autoinducer 3-oxo-dodecanoyl homoserine lactone. Using transcriptomic analysis and quantitative real time PCR we show that these RNA thermometers are the main mechanism of thermoregulation of P. aeruginosa virulence factor production

Microbial succession and exploration of higher alcohols-producing core bacteria in northern Huangjiu fermentation

Project description:Higher alcohols (HAs) are abundant compounds that provide important flavors in Huangjiu, but they also cause hangover. Previous studies have shown the production of HAs to be related to yeast, but the correlations between HAs and other microorganisms are rarely reported. In this study, we detected changes in levels of HAs and microbial dynamics during the Huangjiu fermentation process. Relationships were characterized using Pearson’s correlation coefficient. The functional core HA-producing bacteria were selected by bidirectional orthogonal partial least squares (O2PLS). The result showed that 2-methyl-1-propanol, phenethyl alcohol and 3-methyl-1-butanol were the principle HAs present at high levels. Lactococcus and Saccharomyces were predominant at the genus level of bacteria and fungi, respectively. A total of 684 correlations between HAs and microorganisms were established. Five genera were screened as functional core HA-producing bacteria. Our findings might provide some new inspiration for controlling the content of HAs, enhancing international prestige and market expansion of Huangjiu. Rice yields decline with higher night temperature from global warming.

Project description:The impact of projected global warming on crop yields has been evaluated by indirect methods using simulation models. Direct studies on the effects of observed climate change on crop growth and yield could provide more accurate information for assessing the impact of climate change on crop production. We analyzed weather data at the International Rice Research Institute Farm from 1979 to 2003 to examine temperature trends and the relationship between rice yield and temperature by using data from irrigated field experiments conducted at the International Rice Research Institute Farm from 1992 to 2003. Here we report that annual mean maximum and minimum temperatures have increased by 0.35 degrees C and 1.13 degrees C, respectively, for the period 1979-2003 and a close linkage between rice grain yield and mean minimum temperature during the dry cropping season (January to April). Grain yield declined by 10% for each 1 degrees C increase in growing-season minimum temperature in the dry season, whereas the effect of maximum temperature on crop yield was insignificant. This report provides a direct evidence of decreased rice yields from increased nighttime temperature associated with global warming.

Samples were harvested at O.D600 of 1.5. Three biological replicates of each growth condition was used for RNA extraction and hybridization on Affimetrix microarrays.

Lactobacillus delbrueckii ssp. bulgaricus Milk- and Low-Temperature-Associated Proteomes

Project description:We identified the proteins synthesized by Lactobacillus delbrueckii ssp. bulgaricus strain LBB.B5 in laboratory culture medium (MRS) at 37 �C and in milk at 37 and 4 �C. Cell-associated proteins were measured by gel-free, shotgun proteomics using high-performance liquid chromatography coupled with tandem mass spectrophotometry. A total of 635 proteins were recovered from all cultures, and among which 72 proteins were unique to or significantly more abundant in L. bulgaricus after growth in milk compared to MRS at 37 �C.
