Project description:To examine fosB regulation of neurogenesis, depression and epilepsy, we compared the gene expression profiles of wild type, fosBd/d and fosB-null mice by microarray analysis. Microarray analysis revealed that genes related to neurogenesis, depression and epilepsy are altered in the hippocampus of fosB-null mice. Total RNA from the hippocampus of wild-type, fosBd/d and fosB-null mice was prepared using ISOGEN and RNeasy Mini kits. For DNA microarray experiments, total RNA (100ng) was added to poly-A control and treated with WT Expression Kit. cDNA was labeled using WT terminal labeling kit and hybridized with Affymetrix GeneChip Mouse Gene 1.0 ST platform (GPL6246).

Project description:The Fosb gene encodes subunits of the activator protein-1 transcription factor complex. Two mature mRNAs, Fosb and ΔFosb, encoding full-length FOSB and ΔFOSB proteins respectively, are formed by alternative splicing of Fosb mRNA. Fosb products are expressed in several brain regions. Moreover, Fosb-null mice exhibit depressive-like behaviors and adult-onset spontaneous epilepsy, demonstrating important roles in neurological and psychiatric disorders. Study of Fosb products has focused almost exclusively on neurons; their function in glial cells remains to be explored. In this study, we found that microglia express equivalent levels of Fosb and ΔFosb mRNAs to hippocampal neurons and, using microarray analysis, we identified six microglial genes whose expression is dependent on Fosb products. Of these genes, we focused on C5ar1 and C5ar2, which encode receptors for complement C5a. In isolated Fosb-null microglia, chemotactic responsiveness toward the truncated form of C5a was significantly lower than that in wild-type cells. Fosb-null mice were significantly resistant to kainate-induced seizures compared with wild-type mice. C5ar1 mRNA levels and C5aR1 immunoreactivity were increased in wild-type hippocampus 24 hours after kainate administration; however, such induction was significantly reduced in Fosb-null hippocampus. Furthermore, microglial activation after kainate administration was significantly diminished in Fosb-null hippocampus, as shown by significant reductions in CD68 immunoreactivity, morphological change and reduced levels of Il6 and Tnf mRNAs, although no change in the number of Iba-1-positive cells was observed. These findings demonstrate that, under excitotoxicity, Fosb products contribute to a neuroinflammatory response in the hippocampus through regulation of microglial C5ar1 and C5ar2 expression.

Project description:To identify genes whose expression is regulated by Fosb gene products in microglia, we performed microarray analysis using RNA prepared from wild-type and fosB-null microglial cells. Microarray analysis revealed that expression levels of 6 genes are altered in primary cultured microglial cells prepared from fosB-null mice compared with wild type. fosB+/G mice with a null-mutant allele, fosBG, were established and maintained by backcrossing to C57BL/6J. To yield homozygous mutant mice, fosB+/G (N20 generation) mice were intercrossed. C57BL/6J mice were used as wild-type control.

Project description:We examined here the regulation of gene expression in ventral hippocampus neurons that project to nucleus accumbens (vHPC-NAc) by the transcription factor, ΔFosB. The activity of this circuit is critical to stress-induced social withdrawal. ΔFosB regulates the physiology and behavior of this circuit to produce resilience. Circuit-specific translating ribosomal affinity purification (TRAP) was conducted followed by RNASeq. A strain of floxed Fosb mice (the gene that encodes ΔFosB) was crossed with a ribosomal L10-GFP fusion protein line (Rosa26). To target vHPC-NAc neurons, a retrograde recombinant HSV-Cre was injected into NAc to retrogradely drive expression of GFP and knockout the Fosb gene in vHPC-NAc. Bilateral biopsy punches of vHPC containing GFP-labeled projections were collected, pooled, and processed by TRAP followed by 3rd-generation sequencing of actively translating mRNA. We have now identified Fosb-regulated gene expression in a specific hippocampal-to-accumbens circuit that is important for resilience to stress-induced social withdrawal.

We performed hippocampal ChIP-sequencing to identify ∆FosB target genes in APP mice, a model of AD, and in pilocarpine-treated wildtype mice (Pilo mice), a model of epilepsy. Functional domains modulated by ∆FosB target genes are expanded and diversified in APP mice and in Pilo mice (vs respective controls), and primarily involve neuronal excitability and neurotransmission, neurogenesis, chromatin remodeling, and cellular stress and immunity. To identify genes bound by ∆FosB regardless of seizure etiology, we focused on the 442 ∆FosB target genes in both APP and Pilo mice (not respective controls), which related to pathways including membrane potential regulation, glutamatergic signaling, complement activation, neuron-glia population maintenance, and chromatin dynamics. RNA-sequencing and RT-qPCR in independent mice detected altered expression of several ∆FosB targets shared in APP and Pilo mice. Our findings indicate that seizure-induced ∆FosB can bind genes in seizure etiology-dependent patterns, but can also bind genes in patterns regardless of seizure etiology. Understanding factors that influence ∆FosB binding patterns could reveal novel insights into control of gene expression in disorders with recurrent seizures.

Project description:Adult Neurogenesis and Gene Expression Changes in 5-HT7 Receptor Knockout Mice: In the adult, the formation of new nerve cells in the CNS is restricted to the subependymal layer and to the subgranular zone of the hippocampus (Duman et al., 2001). Clinically adult neurogenesis has received most attention for its possible role in major depression. Depressed patients have reduced hippocampal neurogenesis (Kasper & McEwen, 2008) and hippocampal volume (Colla et al., 2007). It has also been shown that chronic, but not acute, treatment with currently widely used antidepressants, most notably fluoxetine, results in increased hippocampal neurogenesis (Malberg et al., 2000; Miller et al., 2007). Pharmacologically antidepressants act by elevating the amount of synaptic serotonin (5-HT) and they do that within minutes of administration, but the clinical effect is often delayed, sometimes for weeks (Miller et al., 2007). This delay is believed to involve changes in plasticity and neurogenesis. With currently available treatment options for depression 20% or more of patients do not respond to the therapy. Thus, there is a need for an increased understanding of the mechanism behind plasticity and neurogenesis, and for the development of improved therapies for depression. We have in an ongoing project with the aim to functionally characterize the 5-HT7 receptor, one of the most recently discovered receptor subtypes for 5-HT (Lovenberg et al., 1993). Interestingly, we have found that this receptor might be a new target for the treatment of depression. Inactivation or pharmacological blockade of this receptor induces an antidepressant-like profile in several preclinical models of depression (Hedlund et al., 2005; Guscott et al., 2005; Wesolowska et al., 2006a, 2006b) and in sleep patterns (Thomas et al., 2003; Hedlund et al., 2005). Of special interest is also the finding that inhibition of the 5-HT7 receptor potentiates the effect of antidepressants in behavioral models of depression (Wesolowska et al., 2007; Bonaventure et al., 2007). We have recently determined that mice lacking lacking the 5-HT7 receptor show a 25% decrease in the number of proliferating cells in the subgranular zone of the hippocampus compared to wildtype mice. The mice were treated with BrdU, a thymidine analog that is incorporated into the DNA of cells undergoing mitosis. BrdU was then detected immunohistochemically. Both wildtype and knockout mice responded to fluoxetine treatment with an increase in the number of dividing cells. The reduction in the number of dividing cells in the 5-HT7 receptor knockout mice seemingly contradicts the antidepressant-like profile previously observed in these mice, but might on the other hand help to explain previously observed impairments in hippocampus-dependent learning seen in the knockout mice (Roberts et al., 2004) and after pharmacological blockade (Meneses, 2004). The observation that 5-HT7 receptors influence neurogenesis constitutes an entirely new regulatory mechanism for this physiologically and pathophysiologically important process. In order to further understand hippocampal neurogenesis and how it is influenced by 5-HT in general and 5-HT7 receptors in particular we propose to perform a microarray study using the glycogene array designed by the Consortium for Functional Glycomics using total RNA from the hippocampus. A comparison will be made between young adult male mice lacking the 5-HT7 receptor and wildtype sibling controls. Such a study should yield new valuable insights into the pathways regulating neurogenesis. The glycogene array is of particular interest as it includes several genes that have been shown to be either directly or indirectly involved in adult neurogenesis and to be regulated by 5-HT and antidepressants. Of particular interest are cell adhesion molecules which are glycoproteins. One such molecule is NCAM, a marker for immature neurons undergoing remodeling and plasticity (Nakagawa et al., 2002), which is upregulated by fluoxetine treatment (Varea et al., 2006). In contrast, NCAM is downregulated following 5-HT depletion (Brezun & Daszuta, 1999). Chronic fluoxetine treatment has also been shown to upregulate neuropeptide Y, Galectin I, Activin A receptor and Inhibin beta-A, all included in the glycogene array (Miller et al., 2007). Another relevant gene on the glycogene chip is ST8SiaIV (Nakagawa et al., 2002). Experimental Design: The hippocampus will be dissected and total RNA will be extracted according to recommended protocols for array studies. RNA from a sufficient number (2-3 animals) of male 5-HT7 receptor knockout mice will be pooled for each of the three replicates. Control samples will be similarly generated from wildtype siblings. The 5-HT7 receptor mice are on a genetically homogeneous background (backcrossed for 16 generation on a C57BL6/J background) so inter-animal variability should be negligible. The RNA will be analyzed on the glycogene array. 3 KO replicates + 3 WT replicates per glycogene array = 6 arrays

Project description:Among fos family genes modulating the cell fates such as cell proliferation, differentiation and cell death, only the fosB gene produces two forms of mature mRNA for FosB and deltaFosB proteins by alternative splicing of an exonic intron in the exon 4. FosB dramatically enhances the transcription regulation of AP-1 dependent promoters by Jun, while deltaFosB, a truncated form of FosB lacking its C-terminal transactivation domain, suppresses the function of Jun. We and other have shown that FosB and deltaFosB have a distinct function to control the cell fate as well as neuronal functions based on their exogenous expression. To elucidate the authentic function of each protein, it is essential to control each expression separately. We established a mutant mouse embryonic stem (ES) cell line carrying homozygous fosBd/d alleles which encode exclusively deltaFosB, and compared its gene expression profile and phenotypes with fosB-null and wild-type ES cell lines. Both mutant ES cells are devoid of FosB, therefore the common phenotypes between the two mutant ES cells in comparison to wild type depict the effects of FosB deficiency. Opposite phenotypes between the two are considered to be determined by deltaFosB itself. We analyzed the gene expression and cellular function among these two mutant and wild-type ES cells. Experiment Overall Design: Total RNA was purified from ES cells grown in the absence of feeder cells using the Isogen kit (Nippon Gene), according to the manufacturerâs instructions. For DNA microarray experiments, Total RNA (10 microg) were labeled using the Agilent Linear Amplification/Labeling Kit (Agilent Technologies) according to the manufacturer's instructions. One microgram of each Cy3-labeled wild-type and Cy5-labeled mutant cDNA or each Cy3-labeled mutant and Cy5-labeled wild-type cDNA, respectively, were mixed, then hybridized to Agilent Mouse cDNA Microarrays (G4104A, design file number: 000522R000679, Agilent Technologies) with 8500 unique clones from Incyteâs mouse UniGene 1 clone set, according to the manufacturer's hybridization protocol. After the washing step, the microarray slides were analyzed with an Agilent G2565AA microarray scanner system. These experiments were carried out in duplicate using exchanged dye-labeled cDNA probes (i.e., Cy3 and Cy5 dye-swapping experiments).
