Gene expression profile during Ebselen treatment in budding yeast

ABSTRACT: Ebselen is synthetic selenium containing organic compound. It exhibits glutathione peroxidase (GSH-Px) like activity hence acts as a potent antioxidant therefore has been widely used in the treatment of various diseases. In contrast, certain studies also demonstrate its toxic nature; however its mechanism of action is not well understood. Presently, Ebselen has been used at 100 µM concentration for the elucidation of its molecular and cellular mechanism of action. Microarray analysis identified the differentially expressed genes in 100 µM Ebselen concentration.

Transcriptomic analysis of Alfalfa response to low selenium concentration

Project description:We studied the application of transcriptome technology in alfalfa selenium (Se) treatment. Alfalfa had different states after different concentrations of Se treatment. It shows that lower concentration promoted growth and higher concentration produced toxicity. The positive regulatory effects of moderate Se (100 mg / kg) on alfalfa was determined through preliminary experiments, and the gene expression of Alfalfa under this treatment was further analyzed by transcriptome.

Effect of LPS and glutathione depletion on gene expression profiling in mouse macrophage-like cells

Project description:By microarray gene expression profiling, we investigated whether glutathione (GSH) has a signaling role in the response of RAW 264.7 cells to lipopolysaccharide (LPS). GSH was depleted by prior incubation with 120 µM buthionine sulfoximine (BSO) for 24 h, then cells were treated with 10 ng/ml LPS for 2 or 6 h. DNA microarray analysis was performed in triplicate samples in control and GSH-depleted cells, in the presence or absence of LPS.

Gene expression in broiler livers upon supplementation with selenized yeast

Project description:Selenium (Se) is an essential cofactor of the antioxidant enzyme glutathione peroxidase beside other functions. The evaluation of optimal selenium supplementation in chicken feed and the subsequent effects on animal health and performance requires comprehensive knowledge of the overall metabolic effects of selenium. Therefore the gene expression was measured in the control group with a standard diet and in the group with a Se supplemented diet (0.5mg Se/kg diet) to determine significantly altered gene expression. The selenium was supplemented in the form of selenized yeast (Se-yeast), which mainly consists of organic Se in the form of L-selenomethionine and L-selenocysteine. The control group received a diet, which contained 70μg of Se / kg diet and the Se-yeast group 620μg of Se / kg diet (analyzed).

Global transcriptomic profiling of ischemic/reperfusion injury in an in vivo Gpx1 -/- transgenic mouse model.

Project description:Glutathione peroxidase (Gpx) is a selenium-containing enzyme that catalyses the reduction of a variety of biological peroxides at the expense of reduced glutathione (GSH). Gpx1 is the most abundant isoform and its role has been implicated in neurodegenerative disorders such as Parkinson’s disease (PD), dementia with Lewy bodies tissue (DLB) (Power and Blumbergs, 2009) and traumatic brain injury (Tsuru-Aoyagi et al., 2009). Due to its high abundance, mutation of the Gpx1 allele would lower overall Gpx activity in the brain significantly. Gpx1 knockout (Gpx1-/-) mice do not show overt phenotypic differences, but all indications suggest that these mice are in a chronic “pro-oxidant” state (Cheng et al. 1999; de Haan et al. 2004). Indeed, a recent study from our laboratory illustrated that the absence of Gpx1 exacerbated stroke injury via increased ROS production and vascular permeability (Wong et al. 2008). Furthermore, Gpx1-/- mice demonstrated an increase in caspase-3 activation and greater infarct volume (Crack et al. 2001)

Four selenoproteins, protein biosynthesis, and Wnt signalling are particularly sensitive to limited selenium intake in mouse colon

Project description:Selenium is an essential micronutrient. Its recommended daily allowance is not attained by a significant proportion of the population in many countries and its intake has been suggested to affect colorectal carcinogenesis. Therefore, microarrays were used to determine how both selenoprotein and global gene expression patterns in the mouse colon were affected by marginal selenium deficiency comparable to variations in human dietary intakes. Two groups of 12 mice each were fed a selenium-deficient (0.086mg Se/kg) or a selenium-adequate (0.15mg Se/kg) diet. After 6wk, plasma selenium level, liver, and colon glutathione peroxidase (GPx) activity in the deficient group was 12, 34, and 50%, respectively, of that of the adequate group. Differential gene expression was analysed with mouse 44K whole genome microarrays. Pathway analysis by GenMAPP identified the protein biosynthesis pathway as most significantly affected, followed by inflammation, Delta-Notch and Wnt pathways. Selected gene expression changes were confirmed by quantitative real-time PCR. GPx1 and the selenoproteins W, H, and M, responded significantly to selenium intake making them candidates as biomarkers for selenium status. Thus, feeding a marginal selenium-deficient diet resulted in distinct changes in global gene expression in the mouse colon. Modulation of cancer-related pathways may contribute to the higher susceptibility to colon carcinogenesis in low selenium status.

A study exploring the potential of selenium incorporated in dairy proteins to improve health and reduce the risk of colon cancer.

Project description:Interventions: The aim is to explore the potential of selenium incorporated in dairy proteins to improve bowel health and reduce the risk of colon cancer. Participants will include a special dairy product in their diet in which the dairy proteins have been naturally enriched with selenium. High selenium=100 micrograms per day, Placebo =1.5 micrograms selenium per day, in Milk protein concentrate prepared as a powdered food drink. Each intervention is 8 weeks in length with a 2 week wash-out between each phase. Markers of the health value of selenium will then be examined in samples of blood and samples taken from the lining of the bowel wall. The duration of the intervention will be 2 weeks baseline+ 8 weeks for each treatment phase (16wks) and 2 weeks wash out between treatment phases.
