Response of Escherichia coli and glutathion mutants to Cadmium

ABSTRACT: We investigated the impact of cadmium on the global transcriptome of E. coli wild type, ∆gshA and ∆gshB mutant cells to evaluate the molecular basis of cadmium toxicity in the presence or absence of cellular thiols. This global transcriptome analysis were done with cells synthezising GSH (wild type), gamma-glutamyl-cysteine (∆gshB mutant) or neither of the two cellular thiols (∆gshA mutant) under the influence of 100 µM Cd(II).

Project description:We investigated the impact of cadmium on the global transcriptome of E. coli wild type, â??gshA and â??gshB mutant cells to evaluate the molecular basis of cadmium toxicity in the presence or absence of cellular thiols. This global transcriptome analysis were done with cells synthezising GSH (wild type), gamma-glutamyl-cysteine (â??gshB mutant) or neither of the two cellular thiols (â??gshA mutant) under the influence of 100 ÂµM Cd(II). E. coli cells, wild type, â??gshA and â??gshB mutant strain, were grown at 37 Â°C . At a cell turbidity of 100 Klett, cells were treated for 10 min with 100 ÂµM Cd(II) or no metal as a control in TMM medium. Total RNA was extracted, DNAse digested and reverse-transcribed with either Cy3- or Cy5-labeled dCTP. Both labeled cDNA were hybridized to a slide at 42 Â°C. All experiments were performed with three independent biological repeats.

Quantitative transcriptome analysis of a high-GSH producing Saccharomyces cerevisiae and the wild type based on the second generation sequencing.

Project description:The mutant Saccharomyces cerevisiae Y518 generated much more intracellular glutathione (GSH) than its counterpart dose. The RNA-seq based global transcriptome analysis was performed for exploring the potential mechanisms. Statistical analysis indicated that 1125 differentially expressed genes (fold-change>=2.0, FDR<=0.001) were up-regulated and 503 were down-regulated. There were 12 genes involved in glutathione metabolism. Of which GSH1, encodes gamma-glutamine cysteine synthetase, the rate-limiting enzyme in GSH biosynthesis process, was up-regulated. And MET17, encodes cysteine synthase A, an enzyme catalyzes the biosynthesis of one of the precursor amino acids L- cysteine, was also up-regulated. Besides, regulator SKN7 and several genes involved in oxidative stress response (GPX2, CTT1, SOD1, TRX2) were up-regulated. Intracellular ROS level of Y518 was also enhanced compared to that of 2-10515. Our results indicate that up-regulations of GSH1 and MET17 might be associated with the increased intracellular GSH content of the mutant, and up-regulated GSH1 may be caused by increased intracellular ROS level. Threonine dehydratase couples L-cysteine metabolism to cadmium resistance in Pseudomonas stutzeri 273 through cadmium sulfide nanoparticle biomineralization

Project description:Cysteine is an amino acid containing sulfhydryl (-SH) group which is the basis of cysteine involved in sulfur metabolism and heavy metal detoxification in microorganisms. Here, we demonstrate that adding L-cysteine significantly improves the cadmium resistance and removal ability of Pseudomonas stutzeri 273. Threonine dehydratase (TSD) connects L-cysteine metabolism and cadmium resistance through CdS nanoparticle biomineralization with the ability of catalyzing L-cysteine desulfuration and H2S generation in P. stutzeri 273. Gene knockout of TSD in P. stutzeri 273 results in the decrease of L-cysteine and cadmium resistance, decline of H2S generation, and reduction of CdS biosynthesis ability.

Project description:The transcriptome approach was used to characterize the global change in slr1738 null-mutant gene expression in response to cadmium (50 microM, 180 min), compared to wild type strain gene expression in response to cadmium (50 microM, 180 min). Total RNA were isolated from null-mutant and wild type stressed cells, reverse-transcribed, differentially labelled (dye swapped), hybridized together (stressed versus unstressed samples) and analyzed with DNA glass microarrays (two slides per each time point) (Custom-commercial array : IntelliGeneTM CyanoCHIP version 2.0, TAKARA). To identify differentially expressed genes, the median of the normalized ratio of Cy5/Cy3 intensity was calculated for each spot of the replicated dye-swap. Identifying the impact of cadmium stress on the pneumococcal metalloproteome

Project description:The toxicity of the heavy metal cadmium is well established. However, the molecular basis for how this toxicity perturbs cellular viability is still not well understood. This project investigates the cellular and molecular impacts of cadmium stress in the bacterial pathogen, Streptococcus pneumoniae. We find that cadmium depletes cellular manganese and zinc stores, leading to disruption of metalloproteins and the corresponding biochemical pathways. Furthermore, the over-accumulation of cadmium within the cells appears to facilitate mismetallation events, whereby native metal cofactors are displaced by cadmium, leading to reduced or abrogated enzymatic activity. These metalloproteomic analyses have been conducted in conjunction with transcriptomics and metabolomics to elucidate the global impacts of cadmium stress within a prokaryotic organism.

Effect of pH on Cadmium Toxicity in Escherichia coli

Project description:Metals at high concentrations can exert toxic effects on microorganisms. It has been widely reported that lowering environmental pH reduces effects of cadmium toxicity in bacteria. Understanding the effects of pH-mediated cadmium toxicity on bacteria would be useful for minimizing cadmium toxicity in the environment and gaining insight into the interactions between organic and inorganic components of life. Growth curve analysis confirmed that cadmium was less toxic to Escherichia coli at pH 5 than at pH 7 in M9 minimal salts medium. To better understand the cellular mechanisms by which lowering pH decreases cadmium toxicity, we used DNA microarrays to characterize global gene expression patterns in E. coli in response to cadmium exposure at moderately acidic (5) and neutral (7) values of pH. Higher expression of several stress response genes including hdeA, otsA, and yjbJ at pH 5 after only 5 minutes was observed and may suggest that acidic pH more rapidly induces genes that confer cadmium resistance. Genes involved in transport were more highly expressed at pH 7 than at pH 5 in the presence of cadmium. Of the genes that showed an interaction between pH and cadmium effects, 46% encoded hypothetical proteins, which may have novel functions involved in mitigating cadmium toxicity.

Proteome glioma cell lines LC-MS/MS

Project description:Mutations in isocitrate dehydrogenase 1 or 2 (IDH1/2) define glioma subtypes and are 38 considered primary events in gliomagenesis, impacting tumor epigenetics and metabolism. 39 IDH enzymes are crucial for the generation of reducing potential, yet the impact of the mutation 40 on the cellular antioxidant system is not understood. Here, we investigate how glutathione 41 (GSH) levels are maintained in IDH1 mutant gliomas, despite an altered NADPH/NADP 42 balance. We find that IDH1 mutant astrocytomas specifically upregulate cystathionine γ-lyase 43 (CSE), the enzyme responsible for cysteine production upstream of GSH biosynthesis. Genetic 44 and chemical interference with CSE in patient-derived glioma cells carrying the endogenous 45 IDH1 mutation, sensitized tumor cells to cysteine depletion, an effect not observed in IDH1 46 wild-type gliomas. This correlated with reduced GSH synthesis as shown by in vitro and in vivo 47 serine tracing and led to delayed tumor growth in mice. Thus we show that IDH1 mutant 48 astrocytic gliomas critically rely on NADPH-independent de novo GSH synthesis to maintain 49 the antioxidant defense, which uncovers a novel metabolic vulnerability in this dismal disease.

Impact of cadmium stress on the transcriptome of Streptococcus pneumoniae

Project description:This study reports the impact of cadmium toxicity on the transcriptome of S. pneumoniae. The most transcriptionally-responsive pathways were found to be carbon source metabolism, fatty acid biosynthesis, cellular metal homeostasis and competence. This data provides a global overview of how the pneumococcus responds to metal ion intoxication, and subsequently, how it maintains viability during this stress.
