Expression data of age-specific ovaries in rhesus monkey

ABSTRACT: The biological function of human ovaries declines with increasing age. Moreover, disorders that cause reproductive failure, such as premature ovarian failure (POF), seriously affect the female health and may be related to abnormal gene expression in the ovaries. The objective of this study was to generate the gene expression proﬁles of ovaries in rhesus monkeys of different ages, so as to identify age-related changes in the ovaries and understand their significance in various human reproductive problems.

Moreover, disorders that cause reproductive failure, such as premature ovarian failure (POF), seriously affect the female health and may be related to abnormal gene expression in the ovaries. The objective of this study was to generate the gene expression proM-oM-,M-^Ales of ovaries in rhesus monkeys of different ages, so as to identify age-related changes in the ovaries and understand their significance in various human reproductive problems. Ovaries were obtained from young (Y, 3 to 4 years), middle-age (M, 7 to 8 years) and old (O, 18 to 19 years) monkeys, and each group had 3 monkey replications. Half of ovary from each monkey was isolated mRNA for whole genome gene expression microarray analysis to detect differentially expressed genes, and followed by the Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses by using a free web-based Molecular Annotation System 3.0 (MAS 3.0).

Nano-encapsulated senolytics cocktail ameliorates germ cell senescence in female mice

Project description:Low-quality oocytes directly affect fertilization and embryo developmental ability, and further contributes to infertility in women. Dasatinib and quercetin, as a senolytics, has been explored extensively in various age-related diseases. Here, we report that nano-encapsulated senolytics cocktail (D+Q) efficaciously ameliorates the the quantity and quality of follicles and oocytes in vitro and in vivo. D+Q cocktail supplementation reduces the level of ROS in aged oocytes, decreases the frequency of fragmentation, maintains the spindle integrity, rescues mislocalized cortical granules, rescues mitochondrial membrane potential and alleviates DNA damage and apoptosis in vitro. Nano-encapsulated D+Q cocktail effectively ameliorates the fertility deficits in the cyclophosphamide-induced primary ovarian failure (POF) mice model. Moreover, RNA sequencing analysis shows that D+Q cocktail improves the fecundity of POF mice by increasing development gene expression and reduces the senescence-associated secretory phenotype (SASP) accumulation. Taken together, our data show that the D+Q cocktail helps to improve assisted reproductive technology and reproductive outcomes in POF.

Exceptional longevity of ovarian and oocyte macromolecules throughout the reproductive lifespan of mammals

Project description:The mechanisms contributing to age-related deterioration of the female reproductive system are complex but aberrant protein homeostasis is a major contributor. We elucidated the exceptionally stable proteins, structures, and macromolecules that persist in ovaries and gametes throughout the reproductive aging timeline in mammals. Ovaries exhibit localized structural and cell-type specific enrichment of stable macromolecules throughout both the follicular and extrafollicular environments. Moreover, both ovaries and oocytes harbor a panel of exceptionally long-lived proteins, including cytoskeletal components, mitochondrial, and oocyte-derived proteins. The exceptional persistence of these long-lived molecules might play a critical role in both lifelong maintenance and age-dependent deterioration of reproductive tissues.

Proteomic quantification of native and ECM-enriched mouse ovaries reveals an age-dependent fibro-inflammatory signature

Project description:The ovarian microenvironment becomes fibrotic and stiff with age due in part to increased stromal collagen and decreased hyaluronan. However, the extracellular matrix (ECM) is a complex network of hundreds of proteins, glycoproteins and glycans which are highly tissue specific and undergo pronounced changes with age. We used a label-free quantitative proteomic methodology to obtain an unbiased and comprehensive protein profile of age-associated alterations of the mammalian ovarian proteome. We analyzed ovaries from reproductively young and old mice using both native and ECM-enriched tissues to specifically interrogate the extracellular matrix. We optimized and validated conditions to enrich for the ECM prior to proteomic analysis and determined that treatment of ovaries with 0.1% SDS for 12.5 hours efficiently removed nuclear material, while maintaining the integrity of the ECM. Following the analysis by data-independent acquisition (DIA), both native and ECM-enriched samples clustered separately based on age, indicating that ovaries from reproductively young and old mice feature clearly distinct proteomic signatures. We identified a total of 4,721 proteins and 383 proteins that are significantly-altered with advanced reproductive age, including key ECM proteins. In addition to ECM proteins, immunoglobulins, proteins that regulate metal homeostasis, and tumor suppressors were highly upregulated with age, whereas downregulated proteins were consistent with the age-dependent decline in fertility. Pathways regulating genomic instability and proteostasis were downregulated with age. Biological processes associated with immune function and ECM remodeling were upregulated with age. These findings provide evidence from a proteomic perspective that the aging ovary provides a fibroinflammatory milieu, and our study suggests target proteins which may drive these age-associated phenotypes for future investigation.
