Project description:To identify potenital target genes of the transcription factor KUODA1 (AT5G47390), we performed expression profiling using an estradiol inducible overexpression system. Estradiol-treatment allows for the nuclear translocation of the constitutively expressed chimeric transcription factor XVE and subsequent expression of KUA1 from the LexA promoter. Stable trangenic T3 seedlings were grown for 14 days and either treated with 15 M-BM-5M estradiol or ethanol (mock) for 4 hours. Two mock and two estradiol treated samples were used for RNA isolation and hybridization.

Transcriptome analysis of estradiol-inducible overexpression of transcription factor WRKY75 in Arabidopsis thaliana

Project description:Sequence-specific transcription factor WRKY75 is highly responsive to reactive oxygen species on transcriptional level in the rosettes of Arabidopsis thaliana. In addition, it acts in developmental responses, acquisition of nutrients, and in stress responses. In the root, WRKY75 is a repressor of root hair formation, it regulates the phosphate starvation response, and the response to certain pathogens. In order to find the target genes of WRKY75, the effects of estradiol-inducible overexpression of WRKY75 on transcriptome was studied using RNA-seq. Two independent inducible WRKY75 overexpressor lines (N2102362 and N2102363) were treated with estradiol for 3 hours and four biological replicates, five plant per replicate, were used for RNA-seq. As a control, wild type Col-0 was treated with estradiol or water for 3 hours and three biological replicates, five plant per replicate, were used for RNA-seq.

Project description:The aim was to identify early target genes of the senescence-associated transcription factor: ORS1. For this purpose we used DEX-inducible system and studied the expression profile 5h after treatment using Affymetrix microarray. Keywords: genetic modification Arabidopsis 46 day-old transgenic plants (Arabidopsis C24 background, inducible overexpression) were sprayed with 30µM DEX for 5h. As control mock (0.05% ethanol) treated ORS1-inducible over-expression plants were used. Whole rosettes were harvested 5h after induction and used for expression profiling using Affymetrix near-full genome ATH1 arrays. Experiment performed in two biological replications.

For this purpose, three independent amiR-tor lines (amiR-tor9, amiR-tor17 and amiR-tor 20) and EV lines 3 and 6 days after EST- or non-induction were used for expression profile analysis using Affymetrix microarray. Two weeks-old transgenic seedlings were transferred to MS plate with 20ÂµM estradiol to induce the overexpression of amiR-tor (consequently, repression of AtTOR levels) under the control of an estradiol-(EST) inducible promoter.
